Abstract The present study evaluated the avian macrophage responses against Clostridium perfringens that varied in their ability to cause necrotic enteritis in chickens. Strains CP5 (avirulent- netB +), CP1 (virulent- netB +), and CP26 (highly virulent- netB + tpeL +) were used to evaluate their effect on macrophages (MQ-NCSU cells) and primary splenic and cecal tonsil mononuclear cells. The bacilli (whole cells) or their secretory products from all three strains induced a significant increase in the macrophage transcription of Toll-like receptor (TLR)21, TLR2, interleukin (IL)-1β, inducible nitric oxide synthase (iNOS), and CD80 genes as well as their nitric oxide (NO) production and major histocompatibility complex (MHC)-II surface expression compared to an unstimulated control. The CP1 and CP26-induced expression of interferon (IFN)γ, IL-6, CD40 genes, MHC-II upregulation, and NO production was significantly higher than that of CP5 and control groups. Furthermore, splenocytes and cecal tonsillocytes stimulated with bacilli or secretory products from all the strains showed a significant increase in the frequency of macrophages, their surface expression of MHC-II and NO production, while CP26-induced responses were significantly higher for the rest of the groups. In summary, macrophage interaction with C. perfringens can lead to cellular activation and, the ability of this pathogen to induce macrophage responses may depend on its level of virulence.

Abstract Multiple myeloma is a malignant plasma cell condition that mostly affects the skeletal system and bone marrow. Pleural effusions are uncommon and typically result from other conditions coexisting with multiple myeloma. Malignant myelomatous pleural effusions are rare complications of multiple myeloma, occurring in less than 1% of patients and are associated with poor prognosis having mean survival of less than 4 months. The present case report is a 41-year-old multiple myeloma patient who developed bilateral pleural effusion at a disease relapse. Chemotherapeutic regimen of cyclophosphamide, bortezomib, and dexamethasone given. Despite a positive response to treatment, the patient's condition worsened over the course of following month and he eventually passed away. Myelomatous pleural effusion indicates poor prognosis and early consideration helps in quick diagnosis and initiation of treatment which may help in improving prognosis.

Abstract Necrotic enteritis (NE) is an economically important disease of chickens. We have recently shown that inflammatory responses in chickens inoculated orally with virulent Clostridium perfringens were spatially regulated. Here, we used previously virulence-characterized netB + C. perfringens strains, avirulent CP5 and virulent CP18 and CP26, to assess the severity of NE and immune responses in broiler chickens when inoculated intracloacally. The results showed that CP18- and CP26-infected birds had a reduced weight gain and developed milder/less severe NE lesions, as determined by the gross lesions scores, suggesting a subclinical-grade infection. Gene expression analysis in infected birds revealed three statistically significant observations compared to uninfected-control: (1) Increased expression of anti-inflammatory/immunoregulatory interleukin (IL)-10/transforming growth factor (TGF)β in cecal tonsil (CT) and bursa of Fabricius in the CP18/CP26-infected groups. (2) Increased CT transcription of pro-inflammatory IL-1β, IL-6 and interferon (IFN)γ and decreased Harderian gland (HG) expression of IFNγ in the CP18/CP26-infected birds. (3) Increased HG or bursal expression of IL-4 and IL-13 in CP5-infected birds. Collectively, intracloacal C. perfringens inoculation seems to induce a highly regulated inflammatory response in the CT and other mucosal lymphoid organs and an intracloacal infection model may be useful in evaluating immune responses in chickens with subclinical NE.

Abstract Aquatic biodiversity has enormous economic and aesthetic value and is largely responsible for maintaining and supporting overall aquatic environmental health. Aquatic organisms also rely upon the great diversity of aquatic habitats and resources for food and materials and breeding ground. The aquatic biodiversity gets affected by several factors such as industrial pollution or anthropogenic activities. Present investigation is an attempt to document abundance and diversity of aquatic insects of Ramala reservoir in Chandrapur city of Maharashtra. It has been estimated that total 16 species of insects belonging to 14 families and 4 orders inhabit in and around this reservoir. The study was carried out from June 2017 to April 2018, including the three seasons. Season wise fluctuations were clearly observed in the population of insects which were noticed.

Abstract Necrotic enteritis (NE) in poultry is an economically important disease caused by Clostridium perfringens type A bacteria. A global trend on restricting the use of antibiotics as feed supplements in food animal production has caused a spike in the NE incidences in chickens, particularly in broiler populations. Amongst several non-antibiotic strategies for NE control tried so far, probiotics seem to offer promising avenues. The current review focuses on studies that have evaluated probiotic effects on C. perfringens growth and NE development. Several probiotic species, including Lactobacillus , Enterococcus , Bacillus , and Bacteroides bacteria as well as some yeast species have been tested in chickens against C. perfringens and NE development. These findings have shown to improve bird performance, reduce C. perfringens colonization and NE-associated pathology. The underlying probiotic mechanisms of NE control suggest that probiotics can help maintain a healthy gut microbial balance by modifying its composition, improve mucosal integrity by upregulating expression of tight-junction proteins, and modulate immune responses by downregulating expression of inflammatory cytokines. Collectively, these studies indicate that probiotics can offer a promising platform for NE control and that more investigations are needed to study whether these experimental probiotics can effectively prevent NE in commercial poultry operational settings.

Abstract Plankton are important biotic component of aquatic habitat. They do determine the trophic status and quality of water. The phytoplankton is the base of most of the lake food web. It forms a bulk of food for zooplankton, fishes and other aquatic organisms. The biodiversity of phytoplankton and zooplankton in any water body shows a correlation with reference to their occurrences and physic-chemical factors. The present paper is an attempt to enumerate the phytoplanktonic diversity of the Ramala Lake, Chandrapur, and Maharashtra. Data is collected during the different seasons from November 2006 to October 2007. The plankton community showed the seasonal fluctuations. The phytoplankton population of Ramala Lake was found to be composed of four major groups namely Chlorophyceae (48.40%), Bacillariophyceae (24.81%), Myxophyceae (22.44%) and Euglenophyceae (4.32%). The class Chlorophyceae represented by 11 species stood first quantitatively and qualitatively, Bailloriophyceae by 7 species, Myxophyceae by 5 and Euglenophyceae by only 2 species.

Abstract The diversity of many invertebrate taxa from Western Ghats of Maharashtra remains unknown due to lack of systematic studies. We studied freshwater calanoid copepods from this region, and found eleven species of diaptomid copepods in 180 samples collected from 80 sites in the study area. Most of these are new records for this region. The observed fauna includes Oriental, Gondwanan, Palaearctic and Indian endemic taxa. Locality data for all the observed species, along with notes on some ecological aspects of the sampled sites are provided. Non-parametric estimates of species richness for the region suggested an overall adequacy of sampling effort, and probable missing out of some species from habitats with longer hydroperiod. Hydroperiod, depth, electrical conductivity, altitude, mean annual temperature and latitude largely influenced the distribution of species in the study area. Co-occurrences of species were overall rare but more common in temporary habitats. Assemblages of diaptomid copepods restricted to certain sites were identified. The importance of temporary habitats is discussed in the light of their species richness, faunal composition and the increasing anthropogenic pressures they face.

Abstract The relevance of multiple poly- and perfluoroalkyl substance (PFAS) fate and transport process across multiple sites was established through high-resolution characterization of the spatial distribution of PFAS within aqueous film forming foam (AFFF)-related source areas and the downgradient plumes. The maximum total PFAS concentrations in source area groundwater at the three study sites ranged from 6 to 51 mg/L but consistently decreased by several orders of magnitude with distance from the source area at all sites, indicating that non-destructive attenuation of PFAS occurred along each flow path. The relative distribution of different PFAS classes, including zwitterionic/cationic species, provided site-specific lines of evidence for retardation due to hydrophobic, air-water interfacial, and electrostatic partitioning processes, as well as impacts from biotransformation and matrix diffusion at multiple sites. The only site where one of these processes (air-water interfacial partitioning) was not supported by the data (Site 1) was attributable to disturbance of vadose zone soils as part of historic remedial efforts. In other cases, the magnitude that these processes influenced PFAS transport reflected site-specific conditions. This included apparent salting out of PFAS at Site 2 due to its elevated groundwater salinity, which has implications for plume migration in coastal areas. In addition, PFAS was present in lower-permeability soils at each site, suggesting that longer-term retention of PFAS is occurring in these zones. The finding that multiple processes were active at site-wide scales is consistent with expectations that these are naturally occurring reactions that should be relevant at most AFFF-impacted source zones.

Abstract Background: Cardiovascular Disease (CVD) is the most significant prognostic factor in individuals with Type 2 Diabetes (T2D). The myocardial damage in diabetic subjects affects diastolic function before the systolic function. Aim and Objectives: To compare the diastolic dysfunction and its parameters (E/A ratio, E/e' ratio) by transthoracic echocardiography among diabetic and non-diabetic patient and to find relation between diastolic dysfunction and HbA1c. Material and Methods: This was a prospective, observational and case control study. Diabetes Mellitus (DM) was labeled if Fasting Blood Sugar (FBS) ≥ 126 mg/dL or HbA1c ≥ 6.5%. Standardized Transthoracic Echocardiographic (TTE) examination was performed (American Society of Echocardiography). Left Ventricular Diastolic Dysfunction (LVDD) was labeled if 3 or more of these variables are abnormal: "Septal e <7 cm/sec, Lateral e <10 cm/sec), E/e ratio >14, LA volume index 2 >34 mL/m , Peak TR velocity >2.8 m/sec." Results: A total of 50 subjects with Type 2 Diabetes Mellitus (T2DM) cases and 50 healthy age and gender matched controls were included in this study. The peak early trans-mitral filling wave velocity (E) in diabetic population was low in diabetes subjects [41 ± 12 cm/s vs 48 ± 8.5 cm/s]. Mean of E/A ratio in the Study Group was significantly lower as compared to the Control Group ('p' = 0.02). The E/e' ratio was significantly higher in diabetic group (16.5 ± 2.7 vs 14.13 ± 1.92) compared to Control Group. T2DM group had lower E/Aratio, deceleration time, e' compared to the controls and higher E/e' ('p' < 0.02). Total 21 (42%) subjects among diabetes mellitus had diastolic dysfunction [Grade I: 19 (38%) and Grade-II: 2 (4%)]. Total 2 (4%) subjects among control had diastolic dysfunction [Grade I: 2(4%)] ('p' < 0.00001). Age, Body Mass Index (BMI), Systolic Blood Pressure (SBP), Diastolic Blood Pressure (DBP), Left Ventricular Ejection Fraction (LVEF), duration of DM and HbA1c had negative correlation with parameters of diastolic ...
