    Abstract Background: This study addresses the critical need for differentiating between upper and lower gastrointestinal bleeding by focusing on blood routine parameters to enhance diagnostic precision.
    Objective: This study aims to identify and compare specific blood routine parameters to determine their efficacy in distinguishing between upper and lower gastrointestinal bleeding for improved clinical decision-making.
    Methods: This retrospective study analyzed 119 patients with gastrointestinal bleeding (GIB) admitted to our hospital between January 2017 and June 2020. Among them, 86 were diagnosed with upper GIB (UGIB) and 33 with lower GIB (LGIB). After admission, peripheral blood samples were collected for a comprehensive blood routine examination, including white blood cell count (WBC), red blood cell count (RBC), hemoglobin (Hb), platelet count (PLT), blood urea nitrogen (BUN), creatinine (Cr), and BUN to Cr ratio (BUN/Cr ratio). Differences in blood routine parameters were compared between the UGIB and LGIB groups. Receiver Operating Characteristic (ROC) curve analysis was conducted to assess the efficacy of blood routine examinations in differentiating between UGIB and LGIB.
    Results: The study revealed no significant differences in WBC and Cr levels between LGIB and UGIB patients (P > .05). However, UGIB patients exhibited statistically lower levels of RBC, Hb, and PLT, along with higher BUN and BUN/Cr ratio levels compared to LGIB patients (P < .05). Pearson correlation coefficient analysis indicated an inverse correlation of BUN/Cr with RBC, Hb, and PLT in GIB patients and a positive association between BUN/Cr and BUN (P < .05). ROC analysis demonstrated that RBC, Hb, PLT, BUN, and BUN/Cr ratios were effective in distinguishing UGIB from LGIB (P < .05).
    Conclusions: Blood routine parameters, including RBC, Hb, PLT, BUN, and BUN/Cr ratio, are valuable in differentiating between UGIB and LGIB. These parameters can serve as early evaluation indexes for GIB, facilitating timely intervention and treatment to enhance therapeutic outcomes.

    Abstract Purpose: Acute kidney injury (AKI) is a serious complication of sepsis and is characterized by inflammatory response. MicroRNA-210 host gene (MIR210HG) is upregulated in human proximal tubular epithelial cells under treatment of inflammatory cytokines. This study aimed to explore the role of MIR210HG in sepsis-induced AKI.
    Materials and methods: Cell viability was detected by a cell counting kit 8 assay. The levels of proinflammatory cytokines were detected by enzyme-linked immunosorbent assay kits. The protein levels of p65, IκBα, and p-IκBα were examined by western blot analysis. The nuclear translocation of nuclear factor kappa B (NF-κB) was detected by immunofluorescence assay. The histological changes of kidneys were analyzed by hematoxylin and eosin staining assay.
    Results: Lipopolysaccharide (LPS) treatment significantly inhibited cell viability and increased productions of proinflammatory cytokines in proximal tubular epithelial cells (HKC-8). Additionally, MIR210HG levels in HKC-8 cells were increased by LPS treatment. MIR210HG silencing inhibited the LPS-induced cell inflammatory response. MIR210HG activated the NF-κB signaling pathway by promoting the phosphorylation of IκBα and nuclear translocation of p65. Rescue assays revealed that the MIR210HG-induced increase of cytokines levels and decline of cell viability were rescued by QNZ treatment. Knockdown of MIR210HG decreased blood urea nitrogen, serum creatinine, and proinflammatory cytokine levels in AKI rats. Moreover, the knockdown of MIR210HG protected against AKI-induced histological changes of kidneys in rats.
    Conclusion: MIR210HG promotes sepsis-induced inflammatory response of HKC-8 cells by activating the NF-κB signaling pathway. This novel discovery may be helpful for the improvement of sepsis-induced AKI.

    Abstract Objectives: The aim of the present study is to carry out a simple synthesis of aminoantipyrine analogues and exploration of their antibacterial, cytotoxic, and anticonvulsant potential.
    Methods: The compounds were characterized employing multi-spectroscopic methods. The in vitro pharmacological response of a series of bacteria was screened employing serial dilution method. The derivatives were screened against maximal electro-shock for their anticonvulsant activity. Molecular docking was carried out to optimize the interaction of the compounds with HPV16-E7 receptors. Further, the in vitro cytotoxicity was tested against human cervical cancer (SiHa) cell lines.
    Results: The compounds show protection against maximal electroshock, esp. 3-nirto- and 4- methyl-3-nitrobenzamido derivatives. In addition, they reveal appreciable DNA cleavage activities and interactions with HPV16-E7 protein receptors, esp. 3,5-dinitro- and 4-methyl-3-nitrobenzamido derivatives. Furthermore, they show potent activity against cervical cancer cells (LD50 value up to 1200 in the case of 4-methyl-3-nitrobenzamido derivative and an inhibition of a maximum of ~97% of cells).
    Conclusion: The simply synthesized aminoantipyrine derivatives show a variety of biological activities like antibacterial and anticancer effects. In addition, this is the first study demonstrating that 4-aminoantipyrine derivatives show an anticonvulsant activity.

    Abstract Background: There are no effective serum biomarkers for early diagnosis and prognosis prediction of acute cerebral infarction (ACI). This study is to explore the roles of miR-497 in the prognosis of patients with ACI and the correlation between miR-497 and some cytokines.
    Methods: MiR-497 expression in 90 serum samples from ACI patients compared to 60 normal control samples subjects was determined by real-time PCR. The serum levels of IL-6, IL-22, IFN-γ, and TNF-α were detected using ELISA kits. Moreover, the GOS was used to evaluate the prognosis of ACI patients on the 30th day after onset. In addition, the diagnostic and prognostic value of miR-497 for ACI was systemically assessed.
    Results: Serum miR-497 expression was significantly increased in ACI patients (p < 0.05) compared with the healthy control subjects. Additionally, the serum levels of IL-6, IL-22, IFN-γ, and TNF-α were remarkably up-regulated in ACI patients (p < 0.05, p < 0.01 or p < 0.001). Evaluated via the prognostic indicator GOS, the levels of serum miR-497, IL-6, IL-22, IFN-γ, and TNF-α were negatively correlated with GOS score, while miR-497 expression was positively correlated with IL-6, IL-22, IFN-γ, and TNF-α.
    Conclusions: The level of miR-497 may be used as a biological marker for prognosis of ACI patients. In addition, miR-497 in the serum has the potential to be a prognostic and diagnostic marker of ACI and a therapeutic target.
