    Abstract Aplastic anemia (AA) is a rare serious hematologic disorder caused by hematopoietic stem cell failure in maintaining hematopoiesis. AA is virtually fatal if not treated, and diagnosis and therapy require extensive hematologic infrastructure. Academic medical centers in Brazil have continuously and significantly contributed to diagnostic tools and therapy development, from novel transplant strategies to drug combinations and implementation science in the national public health system. In the present review, we discuss how the collaborative effort among academic centers in hematology has contributed to improving health care for patients with aplastic anemia. We also discuss what needs are still unmet and how to overcome these challenges.

    Abstract Exquisite discoveries and new interpretations regarding an enigmatic group of cursorial avemetatarsalians led to a new phylogenetic hypothesis regarding pterosaur affinities. Previously thought to be dinosaur precursors, lagerpetids are now considered to be the closest relatives to pterosaurs. This new hypothesis sheds light on a new explorable field, especially regarding the character acquisition and evolution within the pterosaur lineage. In the present study, the morphospace occupation of distinct skeletal regions of lagerpetids within the morphological spectrum of avemetatarsalians is investigated. This approach indicates which portions of the skeleton are more similar to the anatomy of pterosaurs and which portions present different homoplastic signals. The analyses demonstrate that the craniomandibular traits of lagerpetids are pterosaur-like, the pectoral girdle and forelimb are dinosauromorph-like and the axial skeleton and the pelvic girdle and hindlimb are unique and highly specialized among the analyzed sample. So, despite the close phylogenetic relationships, the postcranial skeleton of lagerpetids and pterosaurs are very different. The occurrence of two distinct and highly specialized groups of pterosauromorphs coexisting with a wide ecological range of dinosauromorphs during the Triassic suggests pressure for new niches occupation.

    Abstract COVID-19 is a complex disease manifesting in a broad severity spectrum and involving distinct organs and systems. Hyperinflammation, including complement over-activation, has a pivotal role in severe COVID-19 pathobiology, stimulating the inflammatory response, causing microangiopathy, platelet-neutrophil activation, and hypercoagulability. SARS-CoV-2 can directly activate the complement system by the classic, alternative, and lectin pathways, and infected cells can produce intracellular complement (the complesome). COVID-19 severity appears to be associated with the degree of complement activation, and it has been hypothesized that patients with COVID-19 may benefit from therapeutic complement inhibition. Different complement cascade molecules may be targeted with potential advantages and disadvantages. Which target(s) is the most effective and when is the best timing for intervention remain open questions. Early phase I and phase II clinical trials have shown promising but conflicting results, warranting phase III controlled randomized trials. Upstream complement inhibition appears to better and more effectively block hyperinflammation with potential clinical significance. Understanding how SARS-CoV-2 exploits the complement system can add precious information about the pathogenesis of other infections, inflammatory, and autoimmune diseases beyond COVID-19.

    Abstract Comprising the oldest unequivocal dinosauromorphs in the fossil record, silesaurs play an important role in the Triassic radiation of dinosaurs. These reptiles provide the main source of information regarding the ancestral body plan of dinosaurs, as well as the basis for biogeographic models. Nevertheless, the co-occurrence of silesaurs and the oldest unequivocal dinosaurs is rare, which hampers reliable ecological inferences. Here we present the first species of silesaur from the oldest unequivocal dinosaur-bearing beds from Brazil. Amanasaurus nesbitti gen. et sp. nov. possesses a unique set of femoral traits among silesaurs, including the oldest occurrence of an anterior trochanter separated by the femoral shaft by a marked cleft. Its femoral length indicates that the new species rivals in size with most coeval dinosaurs. This find challenges the assumption that in faunas where silesaurs and unambiguous dinosaurs co-occurred, silesaurs were relatively smaller. Moreover, the presence of dinosaur-sized silesaurs within ecosystems with lagerpetids, sauropodomorphs and herrerasaurids reinforces the complex scenario regarding the early radiation of Pan-Aves. Silesaurs-independent of their phylogenetic position-persisted during most of the Triassic Period, with its plesiomorphic body size advancing through the dawn of dinosaurs, instead of silesaur lineages decrease in body size through time.

    Abstract Saturation is an intriguing phenomenon that has captured the attention of scientists since the time of Froude when he reported it for ship motion in the mid of the nineteenth century. This work presents the demonstration and a comprehensive study of the nonlinear saturation phenomenon on a compound micromachined structure of U-shape (micro portal frame). The frame is designed and fabricated as a multi-input and multi-output device for actuating the 1st (sway) and 2nd (symmetric) in-plane vibration modes. Geometric nonlinearities along with the softening effect of the electrostatic force present the necessary conditions for the activation of a 2:1 internal (auto-parametric) resonance between the 1st and 2nd modes. Experimental data complemented with analytical simulations are obtained showing the internal resonance and the saturation phenomenon. These results are promising for further exploration of such compound structures and for further in-depth studies of the saturation phenomenon on a variety of other systems and applications.

    Abstract Though Paleozoic ray-finned fishes are considered to be morphologically conservative, we report a novel mode of fang accommodation (i.e., the fitting of fangs inside the jaw) in the Permian actinopterygian †Brazilichthys macrognathus, whereby the teeth of the lower jaw insert into fenestrae of the upper jaw. To better understand how fishes have accommodated lower jaw fangs through geologic time, we synthesize the multitude of ways living and extinct osteichthyans have housed large mandibular dentition. While the precise structure of fang accommodation seen in †Brazilichthys has not been reported in any other osteichthyans, alternate strategies of upper jaw fenestration to fit mandibular fangs are present in some extant ray-finned fishes-the needlejaws Acestrorhynchus and the gars of the genus Lepisosteus. Notably, out of our survey, only the two aforementioned neopterygians bear upper jaw fenestration for the accommodation of mandibular fangs. We implicate the kinetic jaws of neopterygians in this trend, whereby large mandibular fangs are more easily fit between the multitude of upper jaw and palatal bones. The restricted space available in early osteichthyan jaws may have led to a proliferation of novel ways to accommodate large dentition. We recommend a greater survey of Paleozoic actinopterygian jaw morphology, in light of these results and other recent reevaluations of jaw structure in early fossil ray-fins.
