Abstract The olive is a fruit tree species economically very important in countries of the Mediterranean basin. Somatic embryogenesis is a powerful in vitro technique with multiple applications in different fields, including breeding programs performed by both classical and innovative procedures. This editorial paper presents a special issue focused on "Somatic embryogenesis in olive". In this manuscript, the conceptual framework of the special issue is established and the contributions are summarized and put into context. Finally, the main bottlenecks limiting the practical applicability of somatic embryogenesis are identified and the future research prospects are discussed.

Abstract Telemedicine has rapidly evolved, offering expanded applications including virtual consultations, remote patient education, and therapeutic options. It provides advantages such as increased accessibility, reduced travel time, and convenience. Challenges include privacy concerns, the digital divide, and the need for regulatory frameworks. Virtual preoperative assessment shows promise in safely identifying patients who do not require in-person consultations. Legal considerations and liability issues need to be addressed. While the COVID-19 pandemic has accelerated the adoption of telemedicine, it has also highlighted the need for comprehensive policies and equitable access to maximize its potential in health care delivery.

Abstract As the global population is aging and surgical needs rise, the occurrence of perioperative neurocognitive disorders (PND) is becoming a significant concern. PND refers to cognitive changes that occur before or after surgery, including neurocognitive disorders, postoperative delirium, and delayed neurocognitive recovery. To address this issue, a brain health assessment initiative within a multidisciplinary team is an emerging concept. Assessing cognitive function, comorbidities, severity of neurocognitive disorders, medications, nutritional status, sleep quality, and other factors can help mitigate the risk of PND and improve patient outcomes.

Abstract This study reports improvement of a protocol for cryopreservation of olive somatic embryos by using the droplet-vitrification method on aluminum foil strips. Two different approaches were considered in order to optimize cryopreservation: investigating the effect of somatic embryos growth conditions, specifically, the influence of the inoculum density of suspension cultures from which cryopreservation explants were collected, and studying the impact of preculture with different sucrose concentrations in both solid and liquid medium. The results obtained showed a significant effect of inoculum density on cryopreservation evaluated as regrowth rate, although a significant influence on recovery percentage could not be inferred. Sucrose preculture significantly improved recovery rates after cryopreservation. The best results were achieved with somatic embryos previously incubated in liquid ECO medium supplemented with 0.2 M sucrose for 28 days. After this treatment, 90% of explants resumed embryogenesis 12 weeks after thawing. Preculture in liquid medium containing 0.2 M sucrose also had a significant influence on the initial response after cryopreservation, with first signs of regrowth resumption 3 days after thawing, compared to 24-25 days in control, non-precultured embryos. Surviving explants continued actively growing, exhibiting the growth pattern normally observed in olive. As revealed by the histological analysis, cell proliferation greatly increased in somatic embryos precultured on sucrose-enriched media. Cellular features of meristematic and proembryogenic cells, mostly constituting the proliferative regions, make them more prone to survive cryopreservation, thus explaining higher recovery rates found after these treatments.

Abstract Cryopreservation is considered the best technique for the safe, long-term conservation of embryogenic cultures. However, before integrating it into a somatic embryogenesis system, the influence of cryopreservation on the final production of plants should be investigated. The objective of this investigation was to evaluate the effect of cryopreservation on the regeneration performance of olive embryogenic cultures as well as on the quality of the plants obtained and their response to ex vitro establishment. In order to analyze the influence of the genotype, all the investigations were carried out in two genetically distinct embryogenic lines. The results obtained revealed no variation in the regeneration potential or the quality of the regenerated plants due to cryopreservation. The subsequent multiplication, rooting, and acclimatization steps were not influenced by cryopreservation either, although a significant genotype × cryopreservation interaction was found for shoot length during the multiplication step. The genotype played an important role, determining the quality of the regenerated plants and some aspects of the multiplication and rooting phases. This investigation revealed that the droplet-vitrification procedure optimized for the cryopreservation of olive somatic embryos can be efficiently used for the long-term conservation of olive embryogenic lines.

Abstract Purpose of review: Postcraniotomy headache (PCH) is a common adverse event and can lead to various complications and decreased quality of life.
Recent findings: To reduce postcraniotomy pain and associated complications, a multimodal pain therapy including analgesics, analgesic adjuncts, and regional anesthesia is essential. The use of opioids should be minimized to facilitate prompt postoperative neurosurgical assessment. Here, we provide an update on the latest evidence regarding the role of scalp nerve blocks in the pain management of patients undergoing craniotomy procedure. Nerve blocks are effective in alleviating postoperative pain after craniotomy. Scalp blocks contribute to lower pain levels and less opioid consumption in the first 48 h following surgery. Moreover, there is a significant decrease in patients suffering from PONV among patients who receive scalp block.

Abstract Purpose of the review: Acute pain management remains a challenge and postoperative pain is often undermanaged despite many available treatment options, also including cannabinoids.
Recent findings: In the light of the opioid epidemic, there has been growing interest in alternative care bundles for pain management, including cannabinoids as potential treatment to decrease opioid prescribing. Despite the lack of solid evidence on the efficacy of cannabinoids, their use among patients with pain, including those using opioids, is currently increasing. This use is supported by data suggesting that cannabinoids could potentially contribute to a better pain management and to a reduction in opioid doses while maintaining effective analgesia with minimum side effects. The scientific basis for supporting the use of cannabis is extensive, although it does not necessarily translate into relevant clinical outcomes. The use of cannabinoids in acute pain did not always consistently show statistically significant results in improving acute pain. Large randomized, controlled trials evaluating diverse cannabis extracts are needed in different clinical pain populations to determine safety and efficacy.
