    Abstract With the development of industry and agriculture, polycyclic aromatic hydrocarbons (PAHs) in the agricultural sector have gradually increased to different degrees, leading to an escalation in environmental pollution. In turn, this escalation has presented a significant possibility of endangering agricultural practices on farmland and has had a serious impact on regional sustainable development. Therefore, a total of 117 samples of soil were gathered to research the pollution level, distribution, sources, and health risk of PAHs in Helan farmland soils. A reference was used for the identification and quantification of PAH content using high-performance liquid chromatography (HPLC) with an ultraviolet detector, and their spatial distribution was analyzed utilizing the Arc Geographic Information System (ArcGIS). The source of PAHs was analyzed by absolute principal component scores/multiple linear regression (APCS-MLR). The lifetime cancer risk increment model and Monte Carlo sensitivity analysis were used to assess the potential health hazards to humans associated with PAHs in soil. Within the current study area, PAHs were higher in the northwest. The results showed that the total content of PAHs in Helan farmland soil ranged from 17.82 to 1544.73 ng·g −1 with a mean of 408.18 ng·g −1 , which indicated the middle degree of pollution in farmland soil. The verification results of the APCS-MLR model showed that the correlation coefficient between the measured values and the predicted values ranged from 0.661 to 0.984, which suggested that the APCS-MLR model demonstrated favorable suitability for conducting source analysis of PAHs in the soil within the study region. Based on the contribution of PAHs from each source, the main sources of PAHs in Helan farmland soil were the combustion source (biomass, diesel, and natural gas combustion) and the transportation source (gasoline for vehicles and traffic exhaust emissions).     Abstract Proton exchange membrane fuel cells are devices that directly convert chemical energy to electricity. A hydrogen oxidation reaction takes place on the anode side, generating protons and electrons. In the cathode, oxygen reduction reaction involving oxygen, proton and electron occurs, producing water and heat. The water content in PEMFCs should be maintained at a reasonable amount to avoid water flooding or membrane dehydration. The thermal management and water management of PEMFCs are important for an efficient and stable operation of PEMFCs. Inside the multiscale spaces of PEMFCs, multiphase flow with a phase change, heat and mass transfer, proton and electron conduction, and electrochemical reaction simultaneously take place, which play important roles in the performance, lifetime and cost of PEMFCs. These processes should be well understood for better designing PEMFCs and improving the thermal management and water management.

    Abstract Coral sand foundation formed by hydraulic fill often faces the problem of poor bearing capacity. This paper proposed for the first time to apply CFG pile composite foundation to coral sand sites to verify the feasibility of this scheme and understand its mechanical characteristics. Firstly, taking on-site coral sand as the research object, a pile sand interface shear test was conducted to clarify the mechanism of pile side friction. At the same time, the ultimate bearing capacity of CFG pile and its composite foundation was measured through in-situ static load tests. Then, based on the strength parameters of the pile sand interface revealed by indoor tests, numerical simulations were conducted to analyze the bearing characteristics of CFG piles and their composite foundations. Finally, a method for calculating the vertical bearing capacity of rigid piles in composite foundation considering interface parameters was proposed. The results showed that the bearing capacity characteristic values of single pile and composite foundation meet the design requirements; The interface friction angle and cohesion together increased the ultimate side friction by 64.41%; The load is mainly borne by the pile tip resistance, and the increase of the interface friction angle will make the proportion of the side friction load first increase and then decrease more obviously; The pile soil stress ratio first increased and then tended to stabilize as the interface strength increased. Compared with the field static load test results, the rationality of the calculation method for composite foundation rigid piles was verified. This study may have reference significance for the design and construction of coral sand foundation treatment in offshore island and reef projects.

    Abstract In order to study the characteristics of pile–rock action of steel pipe driven pile in coral reef limestone stratum, coral reef limestone at the China–Maldives Friendship Bridge site was selected to carry out indoor physical and model tests with red sandstone as the control group. The test outcomes indicate the following: (1) when substantial deformation is permitted, the coral reef limestone has a considerable strength dispersion, a low post-peak stress decrease rate, and a high residual strength, roughly 30% of the peak strength; (2) when the steel pipe pile penetrates the coral reef limestone, the pile top load shows an obvious sawtooth shape, and with the increase in penetration depth, the pile end load of the high-porosity rock sample gradually decreases, and the pile end load of the low-porosity rock sample gradually increases; (3) when the steel pipe pile is penetrated, the strain value of the red sandstone is about twice that of the coral reef limestone at the same position from the steel pipe pile. These findings indicate that the high porosity and heterogeneity cementation characteristics of the coral reef limestone make the extrusion effect during piling significantly less than that of the red sandstone. In addition, the steel pipe pile penetration process is numerically simulated using a four-dimensional discrete spring model method based on the multi-body damage criterion. The numerical simulation results further demonstrate that the pile-side rock fragmentation during steel pipe pile penetration is the primary reason for the lower bearing capacity of steel pipe piles in coral reef limestone stratums. This method provides a novel approach for studying the mechanical properties of reef limestone. The findings can serve as a guide for the design and construction of steel pipe piles in the reef limestone stratum.

    Abstract Emerging epidemiological evidence has linked per- and polyfluoroalkyl substances (PFAS) exposure could be linked to the disturbance of gestational glucolipid metabolism, but the toxicological mechanism is unclear, especially when the exposure is at a low level. This study examined the glucolipid metabolic changes in pregnant rats treated with relatively low dose perfluorooctanesulfonic acid (PFOS) through oral gavage during pregnancy [gestational day (GD): 1–18]. We explored the molecular mechanisms underlying the metabolic perturbation. Oral glucose tolerance test (OGTT) and biochemical tests were performed to assess the glucose homeostasis and serum lipid profiles in pregnant Sprague-Dawley (SD) rats randomly assigned to starch, 0.03 and 0.3 mg/kg·bw·d groups. Transcriptome sequencing combined with non-targeted metabolomic assays were further performed to identify differentially altered genes and metabolites in the liver of maternal rats, and to determine their correlation with the maternal metabolic phenotypes. Results of transcriptome showed that differentially expressed genes at 0.03 and 0.3 mg/kg·bw·d PFOS exposure were related to several metabolic pathways, such as peroxisome proliferator-activated receptors (PPARs) signaling, ovarian steroid synthesis, arachidonic acid metabolism, insulin resistance, cholesterol metabolism, unsaturated fatty acid synthesis, bile acid secretion. The untargeted metabolomics identified 164 and 158 differential metabolites in 0.03 and 0.3 mg/kg·bw·d exposure groups, respectively under negative ion mode of Electrospray Ionization (ESI-), which could be enriched in metabolic pathways such as α-linolenic acid metabolism, glycolysis/gluconeogenesis, glycerolipid metabolism, glucagon signaling pathway, glycine, serine and threonine metabolism.     Abstract Abstract Background Previous studies have reported cross-sectional associations between measures of epigenetic age acceleration (EAA) and kidney function phenotypes. However, the temporal and potentially causal relationships between these variables remain unclear. We conducted a bidirectional two-sample Mendelian randomization study of EAA and kidney function. Genetic instruments for EAA and estimate glomerular filtration rate (eGFR) were identified from previous genome-wide association study (GWAS) meta-analyses of European-ancestry participants. Causal effects of EAA on kidney function and kidney function on EAA were assessed through summary-based Mendelian randomization utilizing data from the CKDGen GWAS meta-analysis of log-transformed estimated glomerular filtration rate (log-eGFR; n = 5,67,460) and GWAS meta-analyses of EAA (n = 34,710). An allele score-based Mendelian randomization leveraging individual-level data from UK Biobank participants (n = 4,33,462) further examined the effects of EAA on kidney function. Results Using summary-based Mendelian randomization, we found that each 5 year increase in intrinsic EAA (IEAA) and GrimAge acceleration (GrimAA) was associated with − 0.01 and − 0.02 unit decreases in log-eGFR, respectively (P = 0.02 and P = 0.09, respectively), findings which were strongly supported by allele-based Mendelian randomization study (both P < 0.001). Summary-based Mendelian randomization identified 24% increased odds of CKD with each 5-unit increase in IEAA (P = 0.05), with consistent findings observed in allele score-based analysis (P = 0.07). Reverse-direction Mendelian randomization identified potentially causal effects of decreased kidney function on HannumAge acceleration (HannumAA), GrimAA, and PhenoAge acceleration (PhenoAA), conferring 3.14, 1.99, and 2.88 year decreases in HanumAA, GrimAA, and PhenoAA, respectively (P = 0.003, 0.05, and 0.002, respectively) with each 1-unit increase in log-eGFR.     Abstract Systemic sclerosis (SSc) is a fibrotic autoimmune disease characterized by pathogenic activation of fibroblasts enhanced by local oxidative stress. The tyrosine phosphatase PTP4A1 was identified as a critical promoter of TGF-β signaling in SSc. Oxidative stress is known to functionally inactivate tyrosine phosphatases. Here, we assessed whether oxidation of PTP4A1 modulates its profibrotic action and found that PTP4A1 forms a complex with the kinase SRC in scleroderma fibroblasts, but surprisingly, oxidative stress enhanced rather than reduced PTP4A1’s association with SRC and its profibrotic action. Through structural assessment of the oxo-PTP4A1-SRC complex, we unraveled an unexpected mechanism whereby oxidation of a tyrosine phosphatase promotes its function through modification of its protein complex. Considering the importance of oxidative stress in the pathogenesis of SSc and fibrosis, our findings suggest routes for leveraging PTP4A1 oxidation as a potential strategy for developing antifibrotic agents.

    Abstract The correlation between single nucleotide polymorphisms (SNPs) of milk fat globule-epidermal growth factor 8 (MFGE8) and metabolic syndrome (MetS) and metabolism-related indicators are limited. The present study explored the relation in Chinese adults.We conducted a nested case–control study based on the Rural Chinese Cohort Study including 408 people with MetS and 408 controls matched by baseline sex, age (±2 years), marital status, and residence village. Four polymorphisms were selected and genotyped by using the SNPscan Genotyping system. Conditional logistic regression was used to estimate the association of MFGE8 polymorphisms with MetS incidence, and linear regression was used to assess the association with metabolism-related indicators in controls.We found no significant association of MFGE8 SNPs with MetS incidence or systolic blood pressure, or triglycerides level, or fasting plasma glucose (P > 0.05). However, MFGE8 rs4932450 was negatively associated with high-density lipoprotein cholesterol (HDL-C) level under the dominant model (β = −0.0218, P = 0.007) and the additive model (β = −0.0175, P = 0.012) and positively associated with diastolic blood pressure (DBP) under the recessive model (β = 4.8848, P = 0.011). The rs3784751 SNP was associated with increased waist circumference (WC) in controls (β = 0.0307, P = 0.028).MFGE8 polymorphisms were not associated with MetS but were related to DBP, HDL-C level, and WC in Chinese adults.

    Abstract Evidences show that cluster determinant 36 (CD36) protein plays a role in lipid metabolism and insulin resistance, and the expression of CD36 is inducible in obesity. The present study evaluated the association of CD36 variants and the interaction with obesity on type 2 diabetes mellitus (T2DM) risk.We performed a case-control study nested in the Rural Chinese Cohort Study. We included 546 incident T2DM cases matched with non-T2DM controls in a 1:1 ratio by sex, age (within 2 years), marital status, and residence village. Four loci in CD36 (rs1194197, rs2151916, rs3211956, and rs7755) were genotyped by SNPscanᵀᴹ Genotyping system.After adjusting for potential confounding, we observed no statistically significant association between the CD36 polymorphisms and T2DM risk. Compared to wild-type homozygous carriers with normal weight, overweight/obesity participants carrying the mutational allele rs7755 showed increased risk of T2DM, by 114% (OR = 2.14, 95% CI: 1.33–3.46; Pinteraction = 0.007); abdominal obesity participants carrying the mutational allele rs7755 showed increased risk of T2DM, by 133% (OR = 2.33, 95% CI: 1.48–3.66; Pinteraction = 0.002). Furthermore, rs2151916 polymorphism was associated with triglycerides level (P = 0.019), and the rs1194197 variant was related to systolic blood pressure (P = 0.023) within the group of controls.CD36 genotypes were not associated with the progression to T2DM independently. However, our results suggested a positive interaction between the CD36 variants and obesity on T2DM susceptibility, which might be through a cardiometabolic disorder.
