    Abstract Background: Schizophrenia is a severe psychiatric disease affecting approximately 0.5-1% of the general population. The relative contribution of genetic factors has been estimated to be 64-81%.
    Objective: This review summarizes recent efforts to identify genetic variants associated with schizophrenia.
    Methods: Relevant linkage and candidate genes as well as genome wide association studies, studies on copy number variants and next generation sequencing are presented and discussed.
    Results: The latest and worldwide largest study on the genetics of schizophrenia found 128 genome wide significant single nucleotide polymorphisms (SNP) and 108 genome wide loci. The most obvious association is with genetic variations in the major histocompatibility complex (MHC). Besides polymorphisms, structural variants in the form of copy number variants (CNV), such as microdeletions and microduplications have a very high impact in a subgroup of patients. These CNVs are mainly microdeletions on 1q21.1, 2p16.3, 3q29, 15q13.3 and 16p11.2 as well as a large deletion on 22q11.21 and a microduplication on 16p11.2.
    Conclusion: A large new body of evidence on the genetics of schizophrenia is expected through next generation sequencing approaches. Future studies will particularly address the functional characterization of genetic variants.

    Abstract Schizophrenia is a severe psychiatric disease with a relative contribution of genetic factors of up to 64–81 %. Beside linkage- and candidate gene studies, especially genome wide association studies (GWAS), studies on copy number variants (CNVs) as well as studies on next generation sequencing (NGS) were published. The so far latest and worldwide largest study on genetics of schizophrenia showed 128 genome wide significant polymorphisms (SNPs, single nucleotide polymorphisms). Especially the MHC (major histocompatibility complex) association shows highest impact. Beside polymorphisms also structural variants (CNVs, copy number variants) do have a highest impact in a subsample of patients. These CNVs are mainly microdeletions (1q21.1; 2p16.3; 3q29; 15q11.2; 15q13.3; 16p11.2; 17p12; 17q12, 22q11.21) as well as microduplications (1q21.1; 7q11.23; 7q36.3; 15q11-13; 16p11.2; 16p13.11).brA large, new body of evidence on genetics of schizophrenia is expected through next generation sequencing techniques. In the future, these findings could help to improve diagnostics and early detection and to adapt drug treatment on the genetic profile of patients. p bKurzfassung: /bDie Schizophrenie ist eine schwerwiegende psychiatrische Erkrankung mit einem genetischen Anteil von ca. 64–81 %. In den letzten Jahren wurden neben Kopplungs- und Kandidatengenstudien v.a. relevante genomweite Assoziationsstudien (GWAS), Studien zu strukturellen Variationen (CNVs, Copy Number Variants) und Sequenzierungen (NGS, Next Generation Sequencing) publiziert. Die bislang letzte und weltweit größte genetische Studie zur Schizophrenie zeigte 128 genomweit signifikante Polymorphismen (SNPs, single nucleotide polymorphisms) auf. Die deutlichste Assoziation liegt im MHC-Bereich (major histocompatibility complex). Zusätzlich zu Polymorphismen haben strukturelle Variationen (CNVs, copy number variants) bei einer Teilgruppe der Patienten einen sehr hohen, vermutlich ursächlichen Einfluss. Hierzu zählen v.a. Mikrodeletionen (1q21.1; 2p16.3; 3q29; 15q11.2; 15q13.3; 16p11.2; 17p12; 17q12) als auch Mikroduplikationen (1q21.1; 7q11.23; 7q36.3; 15q11-13; 16p11.2; 16p13.11).brEs ist davon auszugehen, dass die neuen Sequenziertechniken zu einem enormen weiteren Wissenszuwachs hinsichtlich der Pathophysiologie der Schizophrenie führen werden. In Zukunft kann dies zum einen dazu beitragen, die Diagnostik und Früherkennung zu verbessern und zum anderen die medikamentöse Behandlung individuell auf das genetische Profil der Patienten anzupassen.
