    Abstract A procedure was proposed to estimate dermal exposures based on a physiologically based pharmacokinetic (PBPK) model developed in rats. The study examined vapor concentrations ranging from 500 to 10,000 ppm for dibromomethane and 2,500 to 40,000 ppm for bromochloromethane. These concentrations were reconstructed based on chemical blood levels measured in 4 hr, with errors varying from 0.0% to 52.0%. The PBPK approach adequately predicted the blood concentrations and helped simulate contaminant transport through the stratum corneum and distribution in the body compartments. The proposed technique made it possible to estimate the skin absorption time (SAT) obtained from acute inhalation toxicity data. An inverse relationship exists between the SAT and exposure concentration. The method can be helpful in toxicology and risk assessment of hazardous volatile organic compounds.

    Abstract With the current clinical method for the treatment of hypothyroidism the target for the optimum individual values for free thyroxine concentrations [FT4] and thyrotropine concentrations [TSH] of the specific patient are unknown. This situation leads to unnecessary long experimental medication administration that can take a period of sometimes one year. In this article a method will be described where hypothyroid patients are characterized with weekly measured FT4 and TSH concentrations during the first three weeks of synthetic thyroxine or levothyroxine (L-T4) treatment to predict their optimum [FT4] and belonging [TSH] endpoint for a euthyroid homeostatic state. The treatment with levothyroxine will start for all patients with a reference dose of 100 µg, which can be adjusted by the treating physician to a more safe and appropriate dose for the individual which is monitored with weekly thyroid function tests to observe the progress. After three weeks all characteristics of the patient can be inferred from the measured data. The final titration target together with the individual thyroxine half life can be calculated. With the known characteristics and the L-T4 titration target the clinician or treating physician has an instrument to reduce the experimental treatment burden for the patient from one year to a maximum of four weeks.

    Abstract A mathematical method was developed to study the skin penetration of volatile organic compounds (VOCs) after exposure to a high dose of the substance. While closed-form solutions exist to describe the diffusion and evaporation from small amounts, numerical approaches are often implemented to predict dermal transport involving large doses. This work offers a Laplace transform-based method to estimate the time constant and dynamic and steady-state behaviors. First, the process was divided into two stages, separated by the time it took for excess chemicals to be depleted from the skin surface. Series solutions were written for the percutaneous VOC concentration, absorption and evaporation in the first stage. Application of Laplace transform methods yielded transient profiles after the compound dissipated from the surface of the stratum corneum. In addition, the procedure facilitated the calculation of the time constant and steady-state values. The method was validated using benchtop and fume hood experiments conducted with N,N-diethyl-3-methylbenzamide (DEET) and air velocities of 0.165 m/s and 0.72 m/s, respectively. The increase in the flow rate decreased the total amount of VOC absorbed and reduced the period required for the surface fluid to disappear.

    Abstract Restoration science is growing fast. The restoration of habitats is increasingly part of the discussion over how to tackle the challenges of climate change, biodiversity loss and rural development. With this increasing role and attendant visibility, restoration science has seen increasing controversy. Here I describe six aspects of robust restoration science that should be kept in mind to help realize its potential: do data-driven studies; focus on robust results; improve reproducibility; contextualize the results; give attention to economics; consider the wider goals of restoration. Realizing the potential of restoration science, via robust scientific studies, will provide society with the knowledge and tools to make better choices about which habitats to restore and where. This article is part of the theme issue 'Understanding forest landscape restoration: reinforcing scientific foundations for the UN Decade on Ecosystem Restoration'.

    Abstract (1) Background: The surge in vape (e-cigarette) use among young adults is concerning, as there is limited knowledge about risk factors and health consequences. This study explores the personality traits linked to vape use, and associations between vaping and chronotype, sleep quality, and mental health, among young adults. (2) Methods: 316 participants, aged 18-25, completed measurements of mindfulness, rumination, self-compassion, anxiety/depression, chronotype, and sleep quality. (3) Results: the vape user group scored significantly lower on mindfulness, higher on rumination, and lower on self-compassion. Vape users were more likely to be evening types and had significantly lower sleep quality and higher anxiety symptoms, as well as higher alcohol use and loneliness (at trend) (4) Conclusions: These novel findings enhance our understanding of what might predispose young adults to vaping and the potential impact on their mental health and sleep quality. Findings point to specific cognitive/personality traits as vaping risk factors, which could inform intervention strategies.
