    Abstract Diversity of fishes was evaluated at the Ranjit Sager Wetland and its three adjoining streams. Factors like altitude, bed gradient, dominated substratum and habitat types of the streams have also been studied. These streams fall under Type-B category on the basis of habitat variability, gradient and sinuosity. 43 fish species represented by 6 orders have been reported from the study area. Out of all 43 fish species, 13 come under threatened categories of Red List of IUCN, out of which 2, 3 and 8 come under Endangered (EN), Vulnerable (VU) and Low Risk near threatened category (LRnt) respectively. It has been analyzed from the above pattern that maximum fish species reported from this wetland have fallen under different threatened categories.

    Abstract Species diversity, dominance and abundance of the phytoplankton community of the Ranjit Sager Wetland were studied during June, 2012 to May, 2014 on monthly basis. The main aim was to provide information about overall phytoplankton diversity of this wetland. Phytoplankton samples were collected using phytoplankton net and preserved in 5% formaldehyde solution on the spot, then brought to the laboratory for further analysis. Algae belonging to Cyanophyceae, Chlorophyceae and Bacillariophyceae were recorded. The Bacillariophyceae was the most dominant group followed by the Chlorophyceae and Cyanophyceae. A total of 26 genera were recorded, 12 were belonged to Bacillariophyceae, 11 to Chlorophyceae and 3 to Cyanophyceae. The different types of indices such as Margalef’s index, Simpson index, Shannon-Weaver index and species evenness index were employed to analysis the data. Phytoplankton converts light energy to chemical energy by the process of photosynthesis so they act as primary producers and food for variety of aquatic organisms. Phytoplankton are also act as efficient bio-indicators to measure quality of water.

    Abstract Abstract Here we demonstrate localization of the isoform3 of DNA Methyltransferase1 (DNMT1) enzyme to mitochondria, instead of isoform1 as reported earlier. The fused DNMT1-isoform1, reported earlier to localize in mitochondria, surprisingly showed its exclusive presence inside the nucleus after its ectopic expression; and failed to localize in mitochondria. On the other hand, ectopically expressed DNMT1-isoform3 targeted itself to mitochondria and subsequently methylated CpG regions in the mitochondrial genome. In addition, overexpression of DNMT1-isoform3 affected mitochondrial biology and regulated its function. Under different conditions of oxidative and nutritional stress, this isoform was down-regulated, resulting in hypomethylation of mitochondrial genome. Our study reveals how DNMT1-isoform3, instead of isoform1, is responsible for mtDNA methylation, influencing its biology.

    Abstract Major histocompatibility complex (MHC) class I molecules (proteins) bind peptides of eight to ten amino acids to present them at the cell surface to cytotoxic T cells. The class I binding groove binds the peptide via hydrogen bonds with the peptide termini and via diverse interactions with the anchor residue side chains of the peptide. To elucidate which of these interactions is most important for the thermodynamic and kinetic stability of the peptide-bound state, we have combined molecular dynamics simulations and experimental approaches in an investigation of the conformational dynamics and binding parameters of a murine class I molecule (H-2Kb) with optimal and truncated natural peptide epitopes. We show that the F pocket region dominates the conformational and thermodynamic properties of the binding groove, and that therefore the binding of the C terminus of the peptide to the F pocket region plays a crucial role in bringing about the peptide-bound state of MHC class I.
