    Abstract Quercetin and daidzin are flavonoid and flavonoid glycoside type compounds that have been found in many plants and nutraceuticals. This study aims to examine the in vitro cytotoxic and selectivity properties of quercetin and daidzin on breast and cervical cancers and to study their molecular interaction and stability on epidermal growth factor receptor tyrosine kinase (EGFR-TK) by applying molecular docking and molecular dynamics (MD) simulations. In vitro anticancer activity was performed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method on breast cancer cell (T47D), cervical cancer cells (HeLa), and Vero normal cells, while molecular docking and MD simulation were done by using AutoDock Vina and Amber18 package software, respectively. Quercetin and daidzin showed potent cytotoxic and high selectivity on both cell lines. Daidzin was found to has a higher IC50 and selectivity index than quercetin. Docking and MD results showed that both compounds prefer to interact with epidermal growth factor receptor tyrosine kinase (EGFR-TK). Daidzin showed better interaction than quercetin with a docking score of -9.6 kcal/mol. Also, daidzin was found more stable than quercetin with low RMSD and RMSF values.

    Abstract With the identification of novel antibiotics from nature being pivotal in the fight against human pathogenic bacteria, there is an urgent need for effective methodologies for expedited screening of crude extracts. Here we report the development and validation of a simple and dye-free antimicrobial assay in 96-well microplate format, for both determination of IC 50 values and high-resolution inhibition profiling to allow pin-pointing of bioactive constituents directly from crude extracts. While commonly used antimicrobial assays visualize cell viability using dyes, the developed and validated assay conveniently uses OD 600 measurements directly on the fermentation broth. The assay was validated with an investigation of the inhibitory activity of DMSO against Staphylococcus aureus , temperature robustness, interference by coloured crude extracts as well as inter-day reproducibility. The potential for high-resolution S. aureus growth inhibition profiling was evaluated on a crude extract of an inactive Alternaria sp., spiked with ciprofloxacin.

    Abstract Benalu batu (Begonia sp.) had been used traditionally as an anticancer medicinal plant by Wana tribe in Morowali, Central Sulawesi, This study aims to evaluate the cytotoxic activity of 2-O-β-glucopyranosil cucurbitacin D, isolated from the ethyl acetate soluble fraction of Benalu batu (Begonia sp.) and to determine its action on apoptosis induction. Benalu batu (Begonia sp.) herb was extracted by maceration using ethanol 96% as a solvent. Vacuum liquid column chromatography and preparative thin layer chromatography have been applied on fractionation and isolation of the compound. The structure elucidation was performed by extensive analysis of 1D/2D nuclear magnetic resonance (NMR) and Mass Spectrophotometer (MS). Cytotoxic activity against human breast adenocarcinoma (MCF-7) and human colon colorectal carcinoma (HCT-116) cell lines were performed by 5-diphenyltetrazolium bromide (MTT) method. Annexin V-FITC assay was employed to determine the apoptosis induction. 2-O-β-glucopyranosil cucurbitacin D showed potent cytotoxic activity against MCF-7 and HCT-116 with the IC50 of 19.913 and 0.002 μg/mL, respectively. Annexin V-FITC assay clearly exhibited the cytotoxic mechanism on MCF-7 and HCT-116 via apoptosis induction with a significant percentage of early and late apoptosis of 75.8 and 78.4%, respectively. This study reveals the potential cytotoxic activity of 2-O-β-glucopyranosil cucurbitacin D isolated from Benalu batu and its mechanism via apoptosis induction.

    Abstract Rebung bambu ampel (Bambusa vulgaris Schrad. ex Wendl.) memiliki kandungan nutrisi, namun di samping itu juga rebung mengandung unsur anti nutrisi yang membahayakan kesehatan yaitu kandungan hidrogen sianida (HCN) sehingga dibutuhkan cara pengolahan yang benar agar kandungan sianida dapat dikurangi bahkan dapat dihilangkan sebelum dikonsumsi. Pada penelitian ini rebung diolah dengan cara perendaman dan air mengalir yang dilanjutkan dengan variasi waktu perebusan. Hasil penelitian ini menunjukan bahwa penurunan kadar sianida terendah yaitu pada perendaman dan perebusan selama 5 menit sebesar 68 ppm sedangkan penurunan kadar sianida yang paling besar terjadi pada kombinasi perlakuan rebung yang dicuci dengan air mengalir dan perebusan selama 120 menit sebesar 656,33 ppm. Berdasarkan hasil penelitian ini dapat disimpulkan bahwa waktu perebusan mempengaruhi kadar sianida rebung, semakin lama waktu perebusan kadar sianida semakin menurun.     Abstract Tamoenju (Hibiscus surattensis L.) leaves are included in the malvaceae family which grows in the plateau and are known to have the antidiabetic activity that the standardization of the extract is needed to be conducted. The research aimed to discover the total level of secondary metabolites from ethanol extract. The extraction of Tamoenju leaves through maceration method with 96% ethanol was done to obtain the filtrate. The filtrate obtained was concentrated by using rotary evaporator at 40°C until the concentrated extract was obtained. The extract gained was then tested in a qualitative way with Alkaloid, Flavonoid, Saponin, and Tannin tests by using the suitable reagent for the test parameters. While in the quantitative test the method used was Gravimetric analysis on alkaloid and Saponin compounds, Permanganometry on Tannin compound, and UV-Visible Spectrophotometry on Flavonoid. The qualitative test results reveal that Tamoenju is positive for containing Alkaloid which was characterized by the presence of orange sediment, Flavonoid was characterized by the orange formation, Tannin was characterized by the dark blue and Saponin was characterized by the stable foam. The quantitative test results are Alkaloid by 305.181 g/g, Saponin by 371.112 g/g, Tannin by 55.417 g/g and Flavonoid by 14.999 mg/100 g.     Abstract Objective: To evaluate the cytotoxic activity of wood extracts of Lunasia amara Blanco (L. amara) and to perform further phytochemical standardization. Methods: The wood extracts of L. amara were assessed for cytotoxic activity by in vitro tetrazolium bromide (MTT) method against two human cancer cell lines, cervical cancer cells (HeLa) and breast cancer cells (T47D). Thin layer chromatography, Dragendorf, acetic anhydride-sulfuric acid and ferric chloride were used to detect alkaloids, steroids and polyphenols, respectively. Furthermore, quantitative determination of total alkaloid by ultra fast liquid chromatography-photodiode array detection using lunacrine as a marker compound was performed as well. Results: The ethyl acetate extract exhibited higher cell-growth inhibition than methanol and n-hexane extracts on HeLa and T47D cancer line cells with the IC50 of 71.15 and 79.04 μg/mL, respectively. Total alkaloid in ethyl acetate extract was counted as (10.46 ± 0.28)% (w/w), while lunacrine determined by ultra fast liquid chromatography-photodiode array detection method was found to be (3.55 ± 0.26)% (w/w). Conclusions: The high total alkaloid and lunacrine concentration on the extract confirm the potential cytotoxic property of ethyl acetate wood extract of L. amara.

    Abstract This study aims to determine the levels of nipagin as preservative in body lotions which have no marketing authorization (MA). Each sample was taken from 4 traditional markets located in Palu City. The assay of nipagin in the body lotions was done by using ultraviolet (UV) at wavelength of 257 nm. From 8 different samples, 5 of them contain nipagin of which levels are as follows: A1 = 0.232%; A2 = 0.229%; B1 = 0.124%; B2 = 0.120%; C1 = 0.120%; C2 = 0.117%; and D1 = 0.267%; D2 = 0.273%; and F1 = 0.213%; and F2 = 0.215%. Based on the results obtained, those body lotions that have no marketing authorization meet the requirement of nipagin’s preservative levels according to Analitical Method of National Center for Drug and Food Testing, in which the allowed levels of nipagin for body lotion is 0.4%.
