    Abstract Cyclodextrin (CD)-based polyrotaxanes (PRXs) are supramolecular polymers comprising multiple CDs mechanically interlocked onto a linear polymer chain by capping the polymer ends with bulky stoppers. Among various PRX derivatives, propionylated PRXs (Pr-PRXs) composed of propionylated α-CD and high molecular-weight poly(ethylene glycol) (PEG) form self-assembled nanoparticles in aqueous solution through hydrophobic interactions. Although Pr-PRX nanoparticles can encapsulate hydrophobic drugs in their hydrophobic domains, their release rate is limited. To improve the efficiency of drug release from Pr-PRX nanoparticles, ultraviolet (UV) light-dissociable Pr-PRXs were designed using 4,5-dimethoxy 2-nitrobenzyl groups as UV-cleavable bulky stopper molecules to facilitate UV-induced drug release.     Abstract Recently, the application of β-cyclodextrins (β-CDs) as therapeutic agents has received considerable attention. β-CDs have been reported to have therapeutic effects on various diseases, such as Niemann-Pick type C (NPC) disease, a family of lysosomal storage disorders characterized by the lysosomal accumulation of cholesterol. To further improve the therapeutic efficacy of β-CDs, the use of β-CD-threaded polyrotaxanes (PRXs) has been proposed as a carrier of β-CDs for NPC disease. PRXs are supramolecular polymers composed of many CDs threaded onto a linear polymer chain and capped with bulky stopper molecules. In this review, the design of PRXs and their therapeutic applications are described. To achieve the intracellular release of threaded β-CDs from PRXs, stimuli-cleavable linkers are introduced in an axle polymer of PRXs. The stimuli-labile PRXs can dissociate into their constituent molecules by a cleavage reaction under specific stimuli, such as pH reduction in lysosomes. The release of the threaded β-CDs from acid-labile PRXs in acidic lysosomes leads to the formation of an inclusion complex with the cholesterol that has accumulated in NPC disease patient-derived fibroblasts, thus promoting the extracellular excretion of the excess cholesterol. Moreover, the administration of PRXs to a mouse model of NPC disease caused significant suppression of the tissue accumulation of cholesterol, resulting in a prolonged life span in the model mice. Additionally, the induction of autophagy by the methylated β-CD-threaded PRXs (Me-PRXs) is described. Accordingly, the stimuli-labile PRXs are expected to be effective carriers of CDs for therapeutic applications.

    Abstract A 67-year-old male was admitted to our hospital for sudden onset chest pain and hoarseness. He underwent 2-debranching thoracic endovascular aortic repair for a ruptured aortic arch aneurysm four years prior. However, computed tomography (CT) revealed an aneurysmal rerupture due to a typeⅠa endoleak. We performed partial arch replacement with uncovered stent removal under intermittent hypothermic circulatory arrest. We needed to be more careful than usual open heart surgery because a non-anatomical bypass procedure was performed. The surgery was successful without any major complications, and the patient was discharged on the 23th postoperative day. Reinterventions post-endovascular repair are sometimes difficult;thus, open surgery could be useful for arch replacement.

    Abstract Windswept Sasa grasslands are stable communities but may have the potential to change into other vegetation such as forests through germination of the soil seed bank. I investigated the compositions of the vegetation and soil seed banks in two windswept Sasa grasslands in the Tanzawa Mountains, eastern Japan: Ryugababa (RGB) and Fudonomine (FDM). Tall tree and sub-tall tree seedlings were present in both grasslands but were below a dense cover of Sasa hayatae. The number of buried viable seeds was estimated as 2831 seeds m⁻² comprising 39 species at RGB and 5786 seeds m⁻² comprising 58 species at FDM, 70% of which were graminoids and 5−23% of which were sub-tall trees at both sites. Most sub-tall tree seeds were Weigela decora, which forms windswept scrub forests around Sasa grasslands. No seedlings of S. hayatae germinated from any of the soil seed bank samples. A habitat type analysis of the soil seed banks showed that the “other” type (species that are infrequent or do not belong to a particular plant association) had the highest relative dominance (approximately 70%), followed by windswept scrub forest (13−25%) at both sites. Sørensen’s similarity index between the vegetation and soil seed bank was low (39.6% in RGB and 47.1% in FDM), possibly because the dense Sasa cover prevents seedlings from germinating and growing. These results suggest that the soil seed banks in windswept Sasa grasslands have different species compositions from the vegetation and consequently have the potential to cause a shift to W. decora scrub forest.

    Abstract [Purpose] This study investigated the effects of transcutaneous electrical nerve stimulation on trunk extension muscle strength, walking ability, and the Japanese Orthopedic Association Back Pain Evaluation Questionnaire items of gait disturbance in one case of a subacute osteoporotic vertebral fracture. [Participant and Methods] An 88-year-old female with the first and third lumbar vertebral fractures underwent standard physical therapy (A1 and A2 phases) and transcutaneous electrical nerve stimulation to the sclerotome region of the fractured vertebra (B1 and B2 phases). Assessments were performed before the A1 phase and the day after each phase. Assessment items included the Visual Analog Scale scores for pain during rest, getting up, standing up, and walking; isometric trunk extension muscle strength; walking ability (10-meter walking, continuous walking distance); and the Japanese Orthopedic Association Back Pain Evaluation Questionnaire items. [Results] Even though the pain intensity did not change, isometric trunk extension muscle strength, continuous walking distance, and the Japanese Orthopedic Association Back Pain Evaluation Questionnaire items of gait disturbance were improved in phase B compared to phase A. [Conclusion] Standard physical therapy and transcutaneous electrical nerve stimulation to the sclerotome area may improve trunk extension muscle strength, walking ability, and the Japanese Orthopedic Association Back Pain Evaluation Questionnaire items of gait disturbance in patients with subacute osteoporotic vertebral fractures.

    Abstract This study aimed to evaluate the potential for vegetation recovery from buried seed banks following exclusion of deer after different periods of chronic herbivory in a beech forest. Using the seedling emergence method, I surveyed the floor vegetation, buried seed banks and compared survey results with that of past flora. The study areas were an area within two deer exclosure fences, one established in 1997 and the other in 2003, and the area outside the fences. With regard to floor vegetation, species diversity and dominance of vulnerable (VUL) species were higher in F1997 than in F2003, and species frequency was different among the three plots. In the soil seed banks, species richness, seed density and life form composition were different among the three plots. However, when I excluded Rubus illecebrosus, which is a VUL shrub species, seed density of VUL species was not greatly different among the plots. In addition, the species composition of soil seed banks compared with that of past flora was similar among the plots. This may be because almost no VUL herb species formed the buried seed banks. In contrast, invulnerable (INV) species formed numerous seed banks. These results demonstrate that chronic deer herbivory causes a negative effect on the composition of floor vegetation through VUL herb species, and a positive effect on the seed density of soil seed banks through INV species. Conservation measures are needed while there are still plant organs remaining aboveground.

    Abstract Here we report a case of total aortic arch replacement for typeⅠendoleak after thoracic endovascular aortic repair( TEVAR) using the concomitant chimney graft technique. An 81-year-old man was admitted with sudden back pain. Six years prior, he had undergone TEVAR for treatment of a distal aortic arch aneurysm. Preoperative computed tomography revealed an 80-mm-diameter arch aneurysm and typeⅠendoleak. The back pain was caused by impending aneurysmal rupture;therefore, urgent total arch replacement was performed. One stent was cut from the main endograft and anastomosed to its distal side. The bare metal stent in the left common carotid artery was removed and reconstructed at a healthy distal artery. Postoperative computed tomography revealed no endoleak of the aneurysm, and the patient's postoperative course was uneventful.
