    Abstract We formulate a discrete version of Green's theorem such that a summation of a two-dimensional function over a discrete region can be evaluated by the use of a summation over its discrete boundary. In many cases, the discrete Green theorem can result in computational gain. Applications of the discrete Green theorem to several typical image processing problems are demonstrated. We also apply it to analyze shapes of particle aggregates of Fe2O3. Experimental results of the shape study are presented.

    Abstract Objective: Dysfunction of vascular endothelial cells was associated with diverse human diseases, including cardiovascular disease. Long noncoding RNAs (LncRNAs) were involved in the regulation of cell injury. We aimed to investigate the role of lncRNA testis-specific transcript, Y-linked 15 (TTTY15) in hypoxia-induced cell injury in human umbilical vein endothelial cells (HUVECs).
    Materials and methods: Cell counting kit-8 (CCK8) assay was used to check cell viability. Lactate dehydrogenase (LDH) assay kit and flow cytometry were utilized to evaluate the leakage rate of LDH and cell apoptosis, respectively. The protein levels of Cyclin D1 and B-cell lymphoma-2-Associated X (Bax) in hypoxia-induced HUVECs were measured by Western blot. Quantitative Real-time polymerase chain reaction (qRT-PCR) was conducted to detect the expression levels of TTTY15 and miR-186-5p in hypoxia-induced HUVECs. The starBase was utilized to predict the binding sites between TTTY15 and miR-186-5p and the dual-luciferase reporter assay was performed to verify the interaction.
    Results: Hypoxia inhibited cell viability and promoted the release of LDH and cell apoptosis in HUVECs. Besides, hypoxia significantly decreased the protein level of Cyclin D1 and increased the protein level of Bax in HUVECs. In addition, the expression level of TTTY15 was obviously upregulated in hypoxia-induced HUVECs, opposite to the level of miR-186-5p. Meanwhile, knockdown of TTTY15 or upregulation of miR-186-5p mitigated hypoxia-induced cell injury in HUVECs. Further studies suggested that TTTY15 targeted miR-186-5p and regulated hypoxia-induced cell injury via interacting with miR-186-5p in HUVECs.
    Conclusions: Downregulation of TTTY15 ameliorated hypoxia-induced cell injury by targeting miR-186-5p in HUVECs.

    Abstract A syntactic-semantic approach to information extraction from images is described. The methodology involves the injection of semantic considerations into context-free grammars. The semantic considerations include feature vectors, selection restrictions, feature transfer functions, semantic well-formedness, etc. With such injection, we can make a description scheme which carries the numerical, the structural, and the a prior real world knowledge about the pattern we want to extract. From the description we can construct an analytical mechanism, the creation machine, which wil find the desired pattern amid a chaos of noisy primitives.

    Abstract NAC is one of the most abundant plant-specific groups of transcription factors, which play important roles in plant growth and development regulation, as well as in biotic and abiotic stress responses. In the present study, a stress-responsive NAC gene, AhNAC4, was isolated from a cDNA library of peanut (Arachis hypogaea L.) immature seeds, and characterized for its role in drought tolerance. AhNAC4 shared high amino acid similarity with NAC proteins belonging to the ATAF subfamily. The expression analysis indicated that AhNAC4 was highly induced by drought, salinity and ABA treatments. Transient expression analysis showed the AhNAC4-GFP fusion protein was exclusively localized in the nucleus of onion epidermal cells. Transactivation assays in yeast cells demonstrated that AhNAC4 functioned as a transcription activator and its C-terminus contained the activation domain. Overexpression of AhNAC4 confers enhanced drought tolerance in transgenic tobacco plants. The improved drought tolerance was associated with more stomatal closure and higher water use efficiency. Collectively, our results indicated that AhNAC4 functions as an important regulator in response to drought stress.

    Abstract The media and indicators were absorbed in a paper-disk (P-D) which is made of filters of mixed cellulose. When P-D is inoculated and cultured at 37 degrees C, the presence of Candida albicans can be shown by the color change from white to brown within 24 hours. Results showed that P-D was specific to Candida species, and the susceptibility was 380 CFU/ml. Fifty patients suspected of oral candidosis were detected by P-D. Its positive percentage of findings was insignificant compared with that of the conventional cultures (P > 0.05). However, P-D is easy to manipulate and inexpensive, and particularly useful because result can be obtained within 24 hours.

    Abstract Acyl carrier protein (ACP), as an essential protein cofactor, plays an important role in de novo synthesis of fatty acids in plastids. In this study, the expression profile of peanut (Arachis hypogaea) AhACP1-1 and AhACP1-2 was analyzed in different tissues. The expression level of AhACP1-1 was highest in the seed, whereas expression was barely detected in the shoot, and AhACP1-2 was expressed in every tissue analyzed with the highest expression level detected in the leaf and seed. Overexpression (OE) and antisense-inhibition (AT) of AhACP1 in transgenic tobacco modified the transcript level of endogenous NtACPs, and the content of total lipids and composition of fatty acid in leaves were altered compared with the wild-type control. Transgenic OE-AhACP1 or AT-AhACP1 tobacco exhibited a significant increase or decrease in polyunsaturated C18:2 and C18:3 fatty acid content, and were more tolerant or sensitive to cold stress, respectively. It is suggested that AhACP1 bound with C18:1 might be the specific substrate of oleoyl-ACP thioesterase or glycerol-3-phosphate acyltransferase, and participates in membrane lipid synthesis.

    Abstract Despite improvements in immunosuppressive therapy, acute rejection remains an important cause of morbidity and late graft loss in patients undergoing liver transplantation. Increasing evidence supports an important role for chemokines and their receptors in transplant immunology. An acute liver graft rejection model in rats was used to study the role of the chemokine receptor CXCR3 in acute transplant rejection after liver transplantation by lentivirus-mediated gene transfer of small interfering RNA (siRNA) against CXCR3. Using reverse transcription-polymerase chain reaction it was first shown that three lentivirus-CXCR3 siRNA vectors inhibited the in vitro expression of CXCR3 in activated T-cells bearing CXCR3. Then, it was shown that treatment of the animals with lentivirus-CXCR3 siRNA before liver transplantation reduced CXCR3 mRNA and protein, and protein levels of interleukin (IL)-2, IL-4, IL-6 and IL-10 and interferon-gamma measured as indices of acute graft rejection. Based on the results from this animal model, targeting chemokines by the use of siRNA may become a feasible option for therapy after transplantation.
