    Abstract The search for novel, natural, high-sweetness, low-calorie sweeteners remains open and challenging. In the present study, the structure-based machine learning modeling and sweetness recognition mechanism were investigated to assist this process. It was found that whether or not a compound was sweet was closely related to molecular connectivity and composition (the number of hydrogen bond acceptors and donors), tpsaEfficiency, structural complexity, and shape (nAtomP and Fsp3). While the relative sweetness of sweet compounds was more determined by the molecular properties (tpsaEfficiency and Log P), structural complexity and composition (nAtomP and ATSm 1). The built machine learning models exhibited very good performance for classifying the sweet/non-sweet compounds and predicting the relative sweetness of the compounds. Moreover, a specific binding pocket was found for sweet compounds, and the sweet compounds mainly interacted with the VFT domain of the T1R2-T1R3 through hydrogen bonds. In addition, the results indicated that among the sweet compounds, those that were sweeter bound to the VFT domain stronger than those that had low sweetness. This study provides very useful information for developing new sweeteners.

    Abstract Neuroinflammation and the resulting neurodegeneration is a big challenge for the healthcare system, especially with the aging population. Neuroinflammation can result from a variety of insults to the central nervous system leading to an interplay between immune and brain cells that sustains chronic inflammation and injures neural cells. One facilitator of this toxic interplay are exosomes. Exosomes are nano-sized, bilayer lipid vesicles secreted by cells containing proteins, nucleic acids and lipids. Because exosomes can be internalized by other cells, their contents can elicit inflammatory responses and trigger toxicities in recipient cells. On the flip side, exosomes can act as therapeutic vehicles carrying protective cargo to maintain homeostasis. This review discusses exosome biogenesis, composition, and its role in neuroinflammation and neurodegeneration in the context of multiple sclerosis and Alzheimer's disease. The emerging roles of exosomes as biomarkers of neurologic diseases and as therapeutic delivery vehicles are also discussed. With all of these varying roles, interest and excitement in exosomes continue to grow exponentially and their promise as brain therapeutics is only beginning to be explored and harnessed.

    Abstract International environmental law is the basic charter for dealing with international environmental issues, and it plays an important role in international environmental governance. In recent years, China's economy has achieved great development, but environmental problems are also very prominent. International environmental law provides a basic reference for China's environmental governance, but it also brings challenges to the development of China's environmental productivity to a certain extent. At the same time, due to the unique dominant position of international environmental law, it has played a certain restrictive role in the development of China's environmental productivity. Based on this, in order to promote the sustainable development of China's economy, this paper took China's industrial and agricultural development as the research object. This paper has conducted research on China's industrial and agricultural environmental productivity, aiming to explore the role of international environmental law in China's environmental productivity. The article first analyzed the limitation of international environmental law on China's environmental productivity, and then on this basis, the article conducted a numerical simulation of China's industrial and agricultural environmental productivity. Finally, starting from the actual development situation, the paper aimed to find out the numerical deviation of China's environmental productivity changes through quantitative analysis. In the course of the research on environmental productivity development, the article found a strong correlation between environmental productivity and international environmental law in China. The correlation between environmental productivity and international environmental law reaches 59.93% in South China and 61.02% in North China. Also, after a series of comparative analyses, the article finds that changes in climate characteristics can exacerbate the regulation of China by international environmental law and seriously affect China's economic development. In this case, China's economy declined by 2.6% year-on-year, which further pushed China to practice sustainable development strategies. This shows that it is essential to study the relationship between international environmental law and China's environmental productivity.

    Abstract Context: Due to the different effects of various drugs and the lack of authoritative and unified guidelines in clinical practice, the choice of therapeutic drugs for pulmonary tuberculosis (TB) remains controversial. Rifampicin, levofloxacin, and isoniazid are all anti-TB drugs. However, no comparative studies of the treatment effects of rifampicin + levofloxacin and rifampicin + isoniazid have occurred.
    Objective: The study intended to analyze the therapeutic effects of rifampicin + levofloxacin compared to rifampicin + isoniazid in the treatment of pulmonary TB to provide a clinical reference, which could provide new references for future clinical treatment of the disease.
    Design: The research team conducted a prospective controlled study.
    Setting: The study took place at Zibo Zhoucun District People's Hospital in Zibo, China.
    Participants: Participants were 100 patients with pulmonary TB admitted to the hospital between March 2021 and December 2022.
    Interventions: The research team assigned participants to one of two groups: (1) the intervention group, with 56 participants who received rifampicin + levofloxacin therapy, and (2) the control group, with 44 participants who received rifampicin + isoniazid.
    Outcome measures: The research team measured: (1) clinical efficacy, (2) incidence of adverse reactions, (3) changes in inflammatory factors, (4) changes in immunoglobulins (Igs), and (5) changes in T lymphocyte subsets.
    Results: The intervention group's total effective rate for treatment was significantly higher than that of the control group (P = .049). However, no significant difference existed between the groups in the incidence of adverse reactions (P > .05). Postintervention, the intervention group's inflammatory factors-IL-6, TNF-α, hs-CRP, and β-EP-were significantly lower and its immunoglobulins (Igs) G/A/M (IgG/A/M) were significantly higher than those of the control group (P < .05). In addition, the intervention group's T lymphocyte subpopulations of CD3+ and CD4+ and CD4+/CD8+ were significantly higher and CD8+ was significantly lower than those in the control group (P < .05). In other words, the intervention group had a better immune function.
    Conclusions: Compared with rifampicin + isoniazid, rifampicin + levofloxacin had a better clinical effect in the treatment of pulmonary TB and could effectively regulate patients' immune functions and inhibit inflammatory reactions. The current research team recommends that rifampicin + levofloxacin become the preferred treatment for pulmonary TB in the absence of a drug allergy.
