    Abstract Abstract The ecological environment of the hilly and gully area of the Loess Plateau in northern Shaanxi is fragile and the soil fertility is low. As a result, the yield and quality of Pear-jujube which constitute one of the dominant economic forests in this region, have been severely restricted. At present, the scientific application of fertilizer is important for comprehensively improving the quality of fruit trees, and for devising the optimal management of fruit trees. In particular, the application of organic fertilizers plays an important role in improving soil and improving fruit quality. In this experiment, a field study was conducted to understand the effects of different organic fertilizer applications on physiological growth, photosynthetic characteristics, reproductive growth and nutritional quality of Pear-jujube in the Loess Plateau. The results showed that organic fertilizer significantly promoted the physiological growth of Pear-jujube. The Pear-jujube bearing branch and leaf area under the soybean cake fertilizer (SC) treatment were 20.17 cm and 1246 mm2/leaf, respectively, which are increased by 34% and 44.46% compared with the no fertilizer treatment, which was a control check (CK). The total chlorophyll content of fertilization treatment was significantly higher than that of CK (P < 0.05). The maximum of chlorophyll content was 10.90 mg/dm2 under the biogas fertilizer (BM). The content of LAI was in the order BM > conventional fertilizer (CF) > sheep manure (SM) > SC > CK. The changing trend of gap fraction was opposite to that of LAI, and the density of light was consistent with that of LAI. The density of light BM was the largest, reached 38.06 mol/(m2 d), which was 15.13% higher than that of CK. Organic fertilizer significantly improved the net photosynthetic rate (Pn) and water use efficiency (WUEp) of Pear-jujube. The WUEp of SC was up to 3.30%.     Abstract Environmental changes and ground subsidence along railway lines are serious concerns during high-speed railway operations. It is worth noting that AutoRegressive Integrated Moving Average (ARMA), Long Short-Term Memory (LSTM), and other prediction methods may present limitations when applied to predict InSAR time series results. To address this issue, this study proposes a prediction method that decomposes the nonlinear settlement time series of feature points obtained through InSAR technology using Ensemble Empirical Mode Decomposition (EEMD). Subsequently, multiple Intrinsic Mode Functions (IMFs) are generated, and each IMF is individually predicted using the Prophet forecasting model. Finally, we employ an equal-weight superimposition method to combine the results, resulting in the prediction of the InSAR settlement time series. The predicted values of each component are subsequently weighted equally and combined to derive the final prediction outcome. This paper selects InSAR monitoring data along a high-speed railway in inland China and uses the proposed method and ARMA and Prophet models to carry out comparative experiments. The experimental results show that compared with the ARMA and Prophet models, the method in this paper improves the root mean square error by 58.01% and 32.3%, and increases the mean absolute error by 62.69% and 33.78%, respectively. The predicted settlement values generated by our method exhibit better agreement with the actual InSAR monitoring values.

    Abstract The aim of this study is to explore suitable drip irrigation system on the water saving and high yield of pear-jujube from 2009 to 2012 years in the mountain of northern Shaanxi. The treatments consisted of combinations of 5 drip irrigation systems (DP). The irrigation quota of DP-1, DP-2 and DP-3 treatment was 100 m3 hm-2, 135 m3 hm-2 and 180 m3 hm-2, respectively, irrigated 4 times. The irrigation quota of DP-4 and DP-5 treatment was 135 m3 hm-2 and irrigated 3 and 2times, respectively; and with no irrigation as the control (C). Results indicated that bearing branch length of jujube, fruit set and yield of different drip irrigation system are significantly better than C (P<0.05). Bearing branch length and yield of DP-3 treatment are reached maximum in 2012, which are 22.0 cm and 16772.8 m3 hm-2. And they are increased by 47.7% and 13.2% compared with C, respectively. In addition, the water consumption of different irrigation treatment increases along with the increasing of irrigation amount. And the DP-3 treatment is the highest in different years. The water use efficiency of pear-jujube of low irrigation quota is better than the high irrigation quota. Water use efficiency of 135 m3 hm-2 and irrigated 2 times treatment is the best, which is 1.92 m3 hm-2. Considering the lack of high annual precipitation, we conclude that DP-5 treatment was the best drip irrigation system in the mountain of northern Shaanxi.

    Abstract High throughput technology has prompted the progressive omics studies, including genomics and transcriptomics. We have reviewed the improvement of comparative omic studies, which are attributed to the high throughput measurement of next generation sequencing technology. Comparative genomics have been successfully applied to evolution analysis while comparative transcriptomics are adopted in comparison of expression profile from two subjects by differential expression or differential coexpression, which enables their application in evolutionary developmental biology (EVO-DEVO) studies. EVO-DEVO studies focus on the evolutionary pressure affecting the morphogenesis of development and previous works have been conducted to illustrate the most conserved stages during embryonic development. Old measurements of these studies are based on the morphological similarity from macro view and new technology enables the micro detection of similarity in molecular mechanism. Evolutionary model of embryo development, which includes the “funnel-like” model and the “hourglass” model, has been evaluated by combination of these new comparative transcriptomic methods with prior comparative genomic information. Although the technology has promoted the EVO-DEVO studies into a new era, technological and material limitation still exist and further investigations require more subtle study design and procedure.

    Abstract Evolutionary developmental biology (EVO-DEVO) tries to decode evolutionary constraints on the stages of embryonic development. Two models—the “funnel-like” model and the “hourglass” model—have been proposed by investigators to illustrate the fluctuation of selective pressure on these stages. However, selective indices of stages corresponding to mammalian preimplantation embryonic development (PED) were undetected in previous studies. Based on single cell RNA sequencing of stages during human PED, we used coexpression method to identify gene modules activated in each of these stages. Through measuring the evolutionary indices of gene modules belonging to each stage, we observed change pattern of selective constraints on PED for the first time. The selective pressure decreases from the zygote stage to the 4-cell stage and increases at the 8-cell stage and then decreases again from 8-cell stage to the late blastocyst stages. Previous EVO-DEVO studies concerning the whole embryo development neglected the fluctuation of selective pressure in these earlier stages, and the fluctuation was potentially correlated with events of earlier stages, such as zygote genome activation (ZGA). Such oscillation in an earlier stage would further affect models of the evolutionary constraints on whole embryo development. Therefore, these earlier stages should be measured intensively in future EVO-DEVO studies.

    Abstract The Goto-Kakizaki (GK) rat, which has been developed by repeated inbreeding of glucose-intolerant Wistar rats, is the most widely studied rat model for Type 2 diabetes (T2D). However, the detailed genetic background of T2D phenotype in GK rats is still largely unknown. We report a survey of T2D susceptible variations based on high-quality whole genome sequencing of GK and Wistar rats, which have generated a list of GK-specific variations (228 structural variations, 2660 CNV amplification and 2834 CNV deletion, 1796 protein affecting SNVs or indels) by comparative genome analysis and identified 192 potential T2D-associated genes. The genes with variants are further refined with prior knowledge and public resource including variant polymorphism of rat strains, protein-protein interactions and differential gene expression. Finally we have identified 15 genetic mutant genes which include seven known T2D related genes (Tnfrsf1b, Scg5, Fgb, Sell, Dpp4, Icam1, and Pkd2l1) and eight high-confidence new candidate genes (Ldlr, Ccl2, Erbb3, Akr1b1, Pik3c2a, Cd5, Eef2k, and Cpd). Our result reveals that the T2D phenotype may be caused by the accumulation of multiple variations in GK rat, and that the mutated genes may affect biological functions including adipocytokine signaling, glycerolipid metabolism, PPAR signaling, T cell receptor signaling and insulin signaling pathways. We present the genomic difference between two closely related rat strains (GK and Wistar) and narrow down the scope of susceptible loci. It also requires further experimental study to understand and validate the relationship between our candidate variants and T2D phenotype. Our findings highlight the importance of sequenced-based comparative genomics for investigating disease susceptibility loci in inbreeding animal models.
