    Abstract Abstract Background To examine trends in weight change patterns from young adulthood through midlife to late adulthood and their sex and racial/ethnic disparities among US adults from 1988 to 2018. Methods A total of 48,969 participants from the National Health and Nutrition Examination Survey 1988–1994 and 2001–2018 were included. Results The age-adjusted prevalence of stable non-obesity between young adulthood and midlife declined significantly from 84.1% (95 CI, 82.9-85.3%) in 1988–1994 to 68.7% (67.1-70.2%) in 2013–2018, and between midlife and late adulthood from 71.2% (69.2-73.1%) to 52.4% (50.5-54.2%). The magnitude of increase in the prevalence of weight gain from young adulthood to midlife (from 10.8% [9.9-11.6%] in 1988–1994 to 21.2% [20-22.3%] in 2013–2018; P < 0.001 for trend) was greater than that from midlife to late adulthood (from 14.1% [12.9-15.3%] to 17.2% [16.2-18.1%]; P = 0.002 for trend). The magnitude of increase in the prevalence of stable obesity from young adulthood to midlife (from 3.9% [3.1-4.8%] in 1988–1994 to 9.2% [8.2-10.3%] in 2013–2018; P < 0.001 for trend) was smaller than that from midlife to late adulthood (from 11.2% [10.1-12.2%] to 24.8% [23.3-26.3%]; P < 0.001 for trend). The declining trends in the prevalence of stable non-obesity and increasing trends in the prevalence of weight gain and stable obesity from young adulthood through midlife to late adulthood were also observed for all sex and race/ethnicity subgroups. The magnitude of decrease in the prevalence of stable non-obesity, and the magnitude of increase in the prevalence of weight gain from young adulthood through midlife to late adulthood were greater in men than in women (all P for interaction < 0.01). Weight gain patterns for those aged ≥ 65 years were substantially different from the younger age groups. Conclusions More young people born in later years are encountering obesity and accumulate greater obesity exposure across their lives than young people born in earlier years.

    Abstract Abstract A considerable chronic disease burden existed in people with normal body mass index (BMI), it is imperative to study the prevailing trends in abdominal obesity among Chinese people with normal BMI. Hence, we aimed to analyze updated prevalence data on abdominal obesity trends among Chinese adults with a normal BMI. We used data from the China Health and Nutrition Survey (CHNS) conducted between 1993 and 2015. Abdominal obesity is defined as waist circumference (WC) ≥ 90 cm for men and ≥ 80 cm for women following the International Diabetes Federation recommendations for Asians. Over the 23-year period, the age-standardized mean WC values showed a significant increasing trend among Chinese adults with BMI < 25 kg/m2, with the mean value increased from 74.0 cm to 78.5 cm (P for trend < 0.0001). During the period of 1993–2015, the age-standardized prevalence of abdominal obesity increased from 12.1 to 26.0% (P for trend < 0.0001). Significant increases were observed in both sexes, all age groups, rural and urban residents, and all educational attainment groups (all P for trends < 0.0001), with a greater relative increase noted among men, younger participants, and rural residents. Similar significant trends were noted when a more stringent BMI < 23 kg/m2 cut point (Asian cut point) was applied. A low magnitude of overlap existed between abdominal obesity and general obesity, irrespective of the criteria used. The mean WC and the prevalence of abdominal obesity among Chinese adults with normal BMI increased continuously from 1993 to 2015. The upward trends were noted in both sexes, all age groups, rural and urban regions, and all educational attainment groups. Our estimates emphasize the importance of adding WC in addition to BMI as measures to monitor obesity prevalence.

    Abstract Abstract The contribution of nonalcoholic fatty liver disease (NAFLD) to all-cause and cardiovascular mortality remains controversial. Sarcopenia, a measure of muscle mass, strength and function, may identify which persons are most at risk for adverse effects of NAFLD. We aimed to test the hypothesis that sarcopenia modifies the associations between NAFLD and all-cause and cardiovascular mortality. A total of 2446 older adults (≥ 60 years) from the third National Health and Nutrition Examination Survey were enrolled. Their mortality data were linked to death certificates in the National Death Index. Sarcopenia was defined as having low skeletal muscle mass together with slow gait speed, which captures both muscle mass and muscle function. Ultrasound tests were used for the assessment of hepatic steatosis. During follow-up (median 16.8 years), 1530 older subjects died from any cause, of which 379 were cardiovascular-related. All-cause and cardiovascular mortality rates were 4.31 and 1.07 per 100 person-years, respectively. In a multivariate model, using participants without NAFLD and sarcopenia as the reference group, individuals with both NAFLD and sarcopenia had 1.69 times [95% confidence interval (CI) 1.23–2.31] and 2.17 times (95% CI 1.33–3.54) higher risks of all-cause and cardiovascular mortality, respectively. However, NAFLD persons without sarcopenia had hazard ratios for all-cause and cardiovascular mortality similar to those of the reference group. Sarcopenia modified the associations of NAFLD with all-cause and cardiovascular mortality. Sarcopenia may identify older adults who are at the highest risk for adverse outcomes associated with NAFLD.

    Abstract Acupuncture can effectively improve the sleep state, and most PD patients have sleep disorders. In this study, we used acupuncture to intervene in the sleep state of PDSD, so as to observe the changes and dose effect of Acutreatment on PDSD. 57 patients with PDSD, during medical treatment, aged 40–70 years were recruited to enroll in this trial. Each participant completed one condition, namely, Acutreatment (n=30) and sham Acutreatment (placebo, stick flat needle on skin, n=27). The Acutreatment was applied for 30 min once a day for a 30-day observation. UPDRSIII scores for motor symptom assessment and sleeping quality were assessed by PDSS-2, ESS as well as ActiGraph. Scale evaluation was made on the first day of admission and the thirtieth day. There were significant differences on all outcome indicators, except UPDRSIII, on day 30 compared with day 1 (P<0.01). Compared with sham Acutreatment therapy, Acutreatment therapy has better performance in sleep latency, total sleep time, and sleep efficiency (P<0.01). ActiGraph indicated that sleep efficiency of sham or Acutreatment in day 6 was significantly lower than that in day 5 (P<0.05 and P<0.01) and Acutreatment in day 7 was significantly lower than that in day 6 (P<0.01). The sleep efficiency of Acutreatment in days 5, 6, and 7 was significantly higher than that in sham Acutreatment (P<0.01). Moreover, Acutreatment in days 26, 27, and 28 was significantly higher than that in sham Acutreatment (P<0.01). There was a close correlation between the difference of UPDRSIII and PDSS-2 (r=0.5090, P<0.05), sleep latency (r=0.7201, P<0.01), TST (r=−0.6136, P<0.01), and sleep efficiency (r=−0.6707, P<0.01). The sleep condition of PDSD patients can be improved by acupuncture, which can effectively relieve sleep quality, can also be shown by ActiGraph, and shows a dose-response relationship. Future research should explore Acutreatment with a larger sample size and compare the Acutreatment protocol goal formation of the system scheme.

    Abstract The extracellular heat shock protein 90α (eHSP90α) has been reported to promote cancer cell motility. However, whether pancreatic cancer (PC) cells expressed membrane-bound or secreted HSP90α, as well as its underlying mechanism for PC progression, were still unclear. Our study demonstrated that the amounts of secreted HSP90α proteins were discrepant in multiple PC cells. In addition, highly invasive Capan-2 cells have a higher level of secreted HSP90α compared with those of less invasive PL45 cells. The conditioned medium of Capan-2 cells or recombinant HSP90α treatment stimulated the migration and invasion of PC cells, which could be prevented with a neutralizing anti-HSP90α antibody. Furthermore, secreted HSP90α promoted elements of epithelial–mesenchymal transition in PL45 cells, including increases in vimentin and Snail expressions, decreases in E-cadherin expression, and changes in cell shape towards a mesenchymal phenotype, but these phenomena were reversed by the anti-HSP90α antibody in Capan-2 cells. In addition, high levels of low-density lipoprotein receptor-related protein 1 (LRP1) were associated with worsened patient survival in pancreatic adenocarcinoma. We demonstrated LRP1 as a receptor of eHSP90α for its stimulatory role in metastasis, by activating the AKT pathway. In addition, silencing LRP1 enhanced the chemosensitivity to gemcitabine and doxorubicin in Capan-2 cells. Therefore, our study indicated that blocking secreted HSP90α underlies an aspect of metastasis and chemoresistance in PC.

    Abstract Abstract Background Studies evidenced that reduction in cardiovascular disease (CVD) mortality in diabetic patients can be attributed to improvements in major CVD risk factors and evidence-based treatments. Furthermore, studies showed that the relative risk of CVD mortality associated with diabetes compared with non-diabetes is stronger in women than in men. Hence, we aimed to examine trends in CVD risk factors and intervention measures by sex and diabetic status. Methods Analysis of 5 distinct cross-sectional National Health and Nutrition Examination Surveys, 1988–1994, 1999–2002, 2003–2006, 2007–2010, and 2010–2014. Since detailed information on nontraditional risk factors such as sleep apnea was not available in each NHANES survey, traditional CVD risk factors including obesity, hypertension, and dyslipidemia were assessed in the study. To assess whether changes throughout the 27-year period differed by diabetes status, a logistic regression analysis was utilized to examine potential interaction effects between survey and diabetes. The similar process was repeated for sex. Results Means of all risk factors except body mass index and waist circumference decreased and the prevalence of antihypertensive and lipid-lowering medication use increased over time among diabetic and non-diabetic men and women. For both men and women, survey × diabetes status interaction terms for changes in HDL-cholesterol and triglyceride levels were not statistically significant, while the prevalence of antihypertensive and lipid-lowering medication use increased more in diabetic than in non-diabetic persons (all P < 0.001). For women, survey × diabetes status interaction terms indicated that compared with the first survey, total cholesterol, LDL-cholesterol, and non-HDL-cholesterol fallen more in diabetic than in non-diabetic persons (all P < 0.001).     Abstract Abstract Background Over the past decades, the structural and functional genomics of rice have been deeply studied, and high density of molecular genetic markers have been developed. However, the genetic variation in leaf photosynthesis, the most important trait for rice yield improvement, was rarely studied. The lack of photosynthesis phenotyping tools is one of the bottlenecks, as traditional direct photosynthesis measurements are very low-throughput, and recently developed high-throughput methods are indirect measurements. Hence, there is an urgent need for a fast, accurate and direct measurement approach. Result We reported a fast photosynthesis measurement (FPM) method for phenotyping photosynthetic capacity of rice, which measures photosynthesis of excised tillers in environment-controlled lab conditions. The light response curves measured using FPM approach coped well with that the curves measured using traditional gas exchange approach. Importantly, the FPM technique achieved an average throughput of 5.4 light response curves per hour, which was 3 times faster than the 1.8 light response curves per hour using the traditional method. Tillers sampled at early morning had the highest photosynthesis, stomatal conductance and the lowest variability. In addition, even 12 h after sampling, there was no significant difference of photosynthesis rate between excised tillers and in situ. We finally investigated the genetic variations of photosynthetic traits across 568 F2 lines using the FPM technique and discussed the logistics of screening several hundred samples per day per instrumental unit using FPM to generate a wealth of photosynthetic phenotypic data, which might help to improve the selection power in large populations of rice with the ultimate aim of improving yield through improved photosynthesis.     Abstract Pale, soft, and exudative (PSE) meat can cause consumer dissatisfaction and economic losses. This study determined meat quality, glycolytic enzyme activity, and differential gene expression in the longissimus lumborum (LL) and semimembranosus (SM) of normal and PSE pork carcasses. The SM did not result in PSE meat. Hexokinase, lactate dehydrogenase, and pyruvate kinase activities were lower in the SM of PSE carcasses than in the normal carcasses. Functional enrichment analysis revealed that immune, inflammatory, and muscle fibre genes were significantly enriched in PSE pork. More specifically, PPP1R3G and MSS51 may be key genes regulating pork quality in the SM. Meanwhile, the differential expression of PLVAB, ADIPOQ, LEP, MYH4, MYH7, MYL3, MYL6B, FOS, ATF3, and HSPA6 may induce PSE formation in the LL. These results may provide insights into PSE pork formation mechanisms and reveal candidate genes for improving meat quality after validation.
