    Abstract Background: MiR-486-5p expression is restrained in lung adenocarcinoma (LUAD). However, much less has been understood on its role in LUAD. We aimed to explore the biofunctions of miR-486-5p in LUAD.
    Methods: A differential expression analysis based on The Cancer Genome Atlas-LUAD dataset was done to screen the differently expressed miRNAs and mRNAs. MiR-486-5p and SAPCD2 mRNA expression was analyzed by qRT-PCR, and protein level of SAPCD2 was assayed by western blot. Upregulation and downregulation of miR-486-5p or SAPCD2 were achieved by cell transfection. For cell function assays, the proliferation of cancer cells was examined by MTT assay. Cell apoptosis was assessed by flow cytometry and microscopy. Transwell assay was applied to evaluate cell migration and invasion. A dual-luciferase detection was employed to determine the miRNA-mRNA targeting relationship.
    Results: MiR-486-5p expression was notably reduced in LUAD tissue and cell lines. Upregulating miR-486-5p restrained the anti-apoptotic and proliferative abilities, as well as cell migratory and invasive phenotypes in LUAD cells. SAPCD2 was determined as one target of miR-486-5p. Also, SAPCD2 forced expression was able to attenuate the inhibitory impacts of miR-486-5p on the malignant phenotypes of LUAD cells.
    Conclusion: MiR-486-5p suppressed cell malignant progression in LUAD by targeting SAPCD2, suggesting that the two may be targets for LUAD treatment.

    Abstract Organocatalytic desymmetrization reaction is a powerful tool for constructing axial chirality, but the theoretical study on the origin of stereoselectivity still lags behind even now. In this work, the N-heterocyclic carbene (NHC)-catalyzed desymmetrization reaction of biaryl frameworks for the synthesis of axially chiral aldehydes has been selected and theoretically investigated by using density functional theory (DFT). The fundamental pathway involves several steps, i.e., desymmetrization, formation of Breslow oxidation, esterification, and NHC regeneration. The desymmetrization and formation of Breslow processes have been identified as stereoselectivity-determining and rate-determining steps. Further weak interaction analyses proved that the C-H···O hydrogen bond and C-H···π interactions are responsible for the stability of the key stereoselective desymmetrization transition states. This research contributes to understanding the nature of NHC-catalyzed desymmetrization reactions for the synthesis of axially chiral compounds.

    Abstract This paper studies the role of high-throughput measurement technology in cancer molecular typing. Based on the Dendrix algorithm, the model proposed in this paper selects the gene replication time as an inherent attribute that affects the frequency of gene mutations and adds it to the model. After setting the size of the gene set, compared with the Dendrix algorithm, the model does not need to delete the gene set that has been found in the process of searching the pathway, and it can find more driving pathway gene sets. Based on the high coverage and high exclusivity of the driving gene set in the pathway and the influence of gene covariates, this paper constructs an adaptive multiobjective optimization model. In order to overcome the problem of gene mutation heterogeneity, this model introduces gene covariates as the weight of gene mutation frequency so that the model is adaptive to each gene. The analysis of the research results shows the reliability of high-throughput sequencing technology.

    Abstract Barrett's esophagus (BE) belongs to a pathological phenomenon occurring in the esophagus, this paper intended to unveil the underlying function of miR-378a-5p and its target TSPAN8 in BE progression. GEO analysis was conducted to determine differentially expressed genes in BE samples. Non-dysplastic metaplasia BE samples, high-grade dysplastic BE samples and controls were collected from subjects. CP-A and CP-B cells were exposed to bile acids (BA) to mimic gastroesophageal reflux in BE cells. RT-qPCR as well as western blot were applied for verifying expressions of miR-378a-5p, TSPAN8, CDX2 and SOX9. CCK-8, wound scratch together with Transwell assays were exploited for ascertaining cell proliferation, migration as well as invasion. The targeted relationship of miR-378a-5p and TSPAN8 could be verified by correlation analysis, dual-luciferase reporter experiment, and rescue experiments. Through analyzing GSE26886 dataset, we screened the most abundantly expressed gene TSPAN8 in BE samples. miR-378a-5p was reduced whereas TSPAN8 was elevated in CP-A as well as CP-B cells after triggering with BA. Knocking down TSPAN8 could counteract BA-triggered enhancement in BE cell proliferation, migration along with invasion. miR-378a-5p could suppress BE cell proliferation, and migration along with invasion via targeting TSPAN8. In BE, miR-378a-5p targeted TSPAN8 to inhibit BE cell proliferation, and migration along invasion. miR-378a-5p deletion or elevation of TSPAN8 may be key point in regulating CDX2 and SOX9 levels, thereby promoting BE formation.

    Abstract Since the appearance of consumer law, its norm production landscape has been highly diversified and fragmented among the countries. The features of “lawlessness in some sectors” and “the lack of efficiency and effectiveness of national legal systems” give rise to the proliferation of norm producing originated by other bodies such as international organizations, industries, traders and even hybrid entities composing the public, the private and the civil society. There are many variations and differences in consumer law and policy between the Global North and the Global South. In recent years, harmonization of standards, codes of conduct and good practices have already taken place through international organizations and global businesses. In relation to harmonization of consumer laws among different countries, there have been skeptical views regarding such feasibility taking into account varying levels of institutional development. Thomas Bernauer explains “variation of consumer protection standards across countries may be a source of trade disputes.” Starting from the past decade, harmonization of consumer law has been achieved or at least advocated at regional levels. Harmonization easily starts within a region since regional integration creates opportunities for approximation of legal infrastructures. The EU is a very successful case in harmonizing consumer laws further guaranteed by a strong judicial system. In Latin America and in Asia, harmonization projects are ongoing and similar initiatives have also been supported. Whilst acknowledging the existing fragmentation and variations of consumer law worldwide, the author highlights the converging environment of consumer protection due to challenges brought by the globalization and advocates the feasibility of the harmonization approach from the perspective of a dynamically emerging and transitional economy such as China. At the outset, it explores the underpinnings of the consumer law which is a fairly modern legal intervention. China, like other countries, has been modernizing its consumer law by adopting a human-centred, rights-centred and social values-centred approach. Then, it examines the more important role played by emerging countries, particularly in areas such as global B2C e-commerce, telecommunication, financial services and mass tourism, and examines international dialogues and cooperation with active participation by the developing countries including China. There is a need for harmonization of consumer laws in the Mainland China, Macau and Hong Kong regions. China has made some progress on its own, but achieving harmonization in the wider region presents particular challenges. The author hopes that the harmonization would be achieved not only by creating consistency of rules but also by coordinating enforcement across borders. In conclusion, the author seems optimistic for a possible universal model law, which contains the minimum legal requirements and standards of consumer protection and mechanisms to strengthen cross-border cooperation to maximize the harmonization of consumer protection worldwide.

    Abstract The random moderation model (RMM) was developed based on a two-level regression model to cope with heteroscedasticity in moderation analysis, and normal-distributed-based maximum likelihood (NML) estimation was developed to estimate the RMM. To present an alternative to the NML, this article discusses the effectiveness of Bayesian estimation for the RMM, aiming to explore a more practical method using the popular software Mplus. Through a simulation study, the RMM based on Bayesian estimation was investigated and compared to maximum likelihood (ML) estimations, including the NML and the default ML estimation in Mplus. The results indicated that the Bayesian approach outperformed the two ML estimations. It showed (a) higher accuracy for estimation of the effect size of the moderation effect; (b) higher 95% credibility interval coverage of the true value of the moderation effect; and (c) well-controlled and more stable type I error rates, while powers comparable to the ML estimations were provided.

    Abstract Additive manufacturing (AM) exhibits a prime increment in manufacturing technology development. The last few decades have witnessed massive improvement in this field of research, including the growth in the process, equipment, and materials. Irrespective of compelling technological advancements, technical challenges provoke the application and development of these technologies. Metal additive manufacturing is considered a prime sector of the industrial revolution. Various metal AM techniques, including Selective Laser Sintering (SLS), Laser Powder Bed Fusion (PBF-LB/M), and Electron Beam Powder Bed Fusion (PBF-EB/M), have been developed according to materials and process classifications. PBF-LB/M is considered one of the most suitable choices for metallic materials. PBF-LB/M of tantalum has become a hot topic of research in the current century owing to the high biocompatibility of tantalum and its high-end safety applications. PBF-LB/M of porous Ta can direct unexplored research prospects in biomedical and orthopedics by adapting mechanical and biomedical properties and pioneering implant designs with predictable features. This review primarily examines the current advancements in the additive manufacturing of tantalum and related alloys using the PBF-LB/M process. The analysis encompasses the evaluation of process parameters, mechanical properties, and potential biological applications. This will offer the reader valuable insights into the present state of PBF-LB/M for tantalum alloys.

    Abstract Objective: To review the research progress of natural biomaterial polyhydroxyalkanoate (PHA) in orthopedics.
    Methods: The literature concerning PHA devices for bone defects, bone repair, and bone neoplasms, respectively, in recent years was extensively consulted. The three aspects of the advantages of PHA in bone repair, the preparation of PHA medical devices for bone repair and their application in orthopedics were discussed.
    Results: Due to excellent biodegradability, biocompatibility, and potential osteoinduction, PHA is a kind of good bone repair material. In addition to the traditional PHA medical implants, the use of electrostatic spinning and three-dimensional printing can be designed to various functional PHA medical devices, in order to meet the orthopedic clinical demands, including the bone regeneration, minimally invasive bone tissue repair by injection, antibacterial bone repair, auxiliary establishment of three-dimensional bone tumor model, directed osteogenic differentiation of stem cells,
    Conclusion: At present, PHA is a hotspot of biomaterials for translational medicine in orthopedics. Although they have not completely applied in the clinic, the advantages of repair in bone defects have been gradually reflected in tissue engineering, showing an application prospect in orthopedics.

    Abstract This was a descriptive study carried out from January to December 2021, at Quanzhou First Hospital, an affiliated hospital of Fujian Medical University, to investigate the efficacy of the urea breath test in detecting Helicobacter pylori infection in patients with peptic ulcer bleeding affected with proton pump inhibitors. A total of 77 patients with peptic ulcer bleeding, who underwent urea breath testing after active bleeding, were divided into two groups. The Helicobacter pylori infection positivity rate in patients with peptic ulcer bleeding was 66.2%. The time from bleeding to detection and from admission to detection was not significantly different between the Helicobacter pylori-positive and -negative groups (p=0.840 and 0.285, respectively). Even with high-dose proton pump inhibitor treatment, a urea breath test can be performed after peptic ulcer bleeding ceases and results in an acceptable positivity rate. There was no significant difference in the accuracy of Helicobacter pylori detection between the time from bleeding to testing and from admission to testing.
