    Abstract In order to study the influence of sudden change of loading rate on crack propagation and failure mode of rock bridge of fractured rock mass, the specimens used in this paper were rock-like materials containing two prefabricated fractures. The mechanical properties and the failure mode of the specimens under different constant loading rates and sudden changes in loading rate were tested. The photographic monitoring and acoustic emission were carried out at the same time. The results show that: (1) In the process of sudden change loading, the specimen shows the characteristics of approximate elastic deformation, and the stress-strain curve after sudden loading is similar to the corresponding stress-strain curve at the corresponding constant loading rate. (2) Combined with acoustic emission detection, it is found that when the loading rate is not much different before the mutation, the loading rates is the same after the mutation and the stress-strain curve of the specimen is similar. In the low-speed loading stage, the acoustic emission count is generally low, while in the high-speed loading stage, the acoustic emission count is generally high. The sudden change from low-speed loading to high-speed loading easily induces stress drop, resulting in crack generation and increase of acoustic emission count. (3) The rate of the specimen during the crack development period plays a decisive role in the failure mode of the specimen. Before the sudden change of loading rate, the low-speed loading within a certain range has little effect on the specimen. When the high-speed loading is carried out when the stress is low in the early stage of the mutation, the acoustic emission count of the specimen is high, which will cause some damage inside the specimen. As a result, even if the rate is the same after the mutation, the final peak stress and failure mode may be different.

    Abstract Abstract Chemical substitution is a promising route for the exploration of a rich variety of doping- and/or disorder-dependent collective phenomena in low-dimensional quantum materials. Here we show that transition metal dichalcogenide alloys are ideal platforms to this purpose. In particular, we demonstrate the emergence of superconductivity in the otherwise metallic single-layer TaSe2 by minute electron doping provided by substitutional W atoms. We investigate the temperature and magnetic field dependence of the superconducting state of Ta1-δWδSe2 with electron doping (δ) using variable temperature (0.34–4.2 K) scanning tunneling spectroscopy (STS). We unveil the emergence of a superconducting dome spanning 0.003 < δ < 0.03 with a maximized critical temperature of 0.9 K, a significant increase from that of bulk TaSe2 (T C = 0.14 K). Superconductivity emerges from an increase of the density of states (DOS) as the Fermi surface approaches a van Hove singularity due to doping. Once the singularity is reached, however, the DOS decreases with δ, which gradually weakens the superconducting state, thus shaping the superconducting dome. Lastly, our doping-dependent measurements suggest the development of a Coulomb glass phase triggered by disorder due to W dopants.

    Abstract Background. Older and sicker adults with type 2 diabetes (T2D) were underrepresented in randomized trials of glucagon-like peptide 1 receptor-agonist (GLP1RA) and sodium-glucose cotransporter 2 inhibitors (SGLT2I), and thus, health benefits are uncertain in this population. Objective. To assess the impact of age, health status, and life expectancy in older adults with T2D on health benefits of GLP1RA and SGLT2I. Design. We used the United Kingdom Prospective Diabetes Study (UKPDS) model to simulate lifetime health outcomes. We calibrated the UKPDS model to improve mortality prediction in older adults using a common geriatric prognostic index. Participants. National Health and Nutrition Examination Survey 2013–2018 participants 65 y and older with T2D, eligible for GLP1RA or SGLT2I according to American Diabetes Association guidelines. Interventions. GLP1RA or SGLT2I use versus no additional medication. Main Measures. Lifetime complications and weighted life-years (LYs) and quality-adjusted life-years (QALYs) across overall treatment arms and life expectancies. Key Results. The overall older adult population was predicted to experience significant health benefits from GLP1RA (+0.29 LY [95% confidence interval: 0.27, 0.31], +0.15 QALYs [0.14, 0.16]) and SGLT2I (+0.26 LY [0.24, 0.28], +0.13 QALYs [0.12, 0.14]) as compared with no added medication. However, expected benefits declined in subgroups with shorter life expectancies. Participants with <4 y of life expectancy had minimal gains of <0.05 LY and <0.03 QALYs from added medication. Accounting for injection-related disutility, GLP1RA use reduced QALYs (−0.03 QALYs [−0.04, −0.02]). Conclusions. While GLP1RA and SGLT2I have substantial health benefits for many older adults with type 2 diabetes, benefits are not clinically significant in patients with <4 y of life expectancy.     Abstract Milk is widely recognized as an important food source with health benefits. Different consumer groups have different requirements for the content and proportion of milk fat; therefore, it is necessary to investigate the differential metabolites and their regulatory mechanisms in milk with high and low milk fat percentages (MFP). In this study, untargeted metabolomics was performed on milk samples from 13 cows with high milk fat percentage (HF) and 13 cows with low milk fat percentage (LF) using ultra-high performance liquid chromatography coupled with mass spectrometry (UHPLC-MS/MS). Forty-eight potential differentially labeled compounds were screened using the orthogonal partial least squares-discriminant analysis (OPLS-DA) combined with the weighted gene co-expression network analysis (WGCNA) method. Amino acid metabolism was the key metabolic pathway with significant enrichment of L-histidine, 5-oxoproline, L-aspartic acid, and L-glutamic acid. The negative correlation with MFP differentiated the HF and LF groups. To further determine the potential regulatory role of these amino acids on milk fat metabolism, the expression levels of marker genes in the milk fat synthesis pathway were explored. It was noticed that L-histidine reduced milk fat concentration primarily by inhibiting the triglycerides (TAG) synthesis pathway. L-aspartic acid and L-glutamic acid inhibited milk fat synthesis through the fatty acid de novo and TAG synthesis pathways. This study provides new insights into the mechanism underlying milk fat synthesis and milk quality improvement.

    Abstract The common deleterious genetic defects in Holstein cattle include haplotypes 1-6 (HH1-HH6), haplotypes for cholesterol deficiency (HCD), bovine leukocyte adhesion deficiency (BLAD), complex vertebral malformation (CVM) and brachyspina syndrome (BS). Recessive inheritance patterns of these genetic defects permit the carriers to function normally, but homozygous recessive genotypes cause embryo loss or neonatal death. Therefore, rapid detection of the carriers is essential to manage these genetic defects. This study was conducted to develop a single-tube multiplex fluorescent amplification-refractory mutation system (mf-ARMS) PCR method for efficient genotyping of these 10 genetic defects and to compare its efficiency with the kompetitive allele specific PCR (KASP) genotyping assay. The mf-ARMS PCR method introduced 10 sets of tri-primers optimized with additional mismatches in the 3' end of wild and mutant-specific primers, size differentiation between wild and mutant-specific primers, fluorescent labeling of universal primers, adjustment of annealing temperatures and optimization of primer concentrations. The genotyping of 484 Holstein cows resulted in 16.12% carriers with at least one genetic defect, while no homozygous recessive genotype was detected. This study found carrier frequencies ranging from 0.0% (HH6) to 3.72% (HH3) for individual defects. The mf-ARMS PCR method demonstrated improved detection, time and cost efficiency compared with the KASP method for these defects. Therefore, the application of mf-ARMS PCR for genotyping Holstein cattle is anticipated to decrease the frequency of lethal alleles and limit the transmission of these genetic defects.

    Abstract Fractured rock mass is a relatively complex medium in nature. It plays a key role in various projects, such as geotechnical engineering, mining engineering and tunnel engineering. Especially, the interaction between fissures has a practical function in the guidance of safe production. This paper takes its research object as rock-like material which contains prefabricated parallel double fissures. It studies how the fissures’ length difference and spacing influence the failure of specimens under uniaxial compression, and analyzes them with fracture mechanics theory. The results include two aspects. Firstly, no matter how the length difference and spacing change, the upper fissure always generates new cracks. Secondly, the length difference and spacing produce three effects on the lower fissure. (1) The fissure propagates less obviously as the length difference increases. With the increase to 40mm, the propagation does not occur at all. (2) The decrease of spacing weakens the propagation. As it is reduced to 5 mm, the propagation stops. (3) The crack propagation is more sensitive to length difference than spacing. Regardless of spacing changes, if a length difference is large enough (40 mm or more), the new crack does not expand, while if it is small enough (10 mm or less), propagation always appears.

    Abstract Beyond the macroscopic perspective, this study microscopically investigates Si 1−x Ge x (001)-2×1 samples that were grown on the epi Ge(001) and epi Si(001) substrates via molecular-beam epitaxy, using the high-resolution synchrotron radiation photoelectron spectroscopy (SRPES) as a probe. The low-energy electron diffraction equipped in the SRPES chamber showed 2×1 double-domain reconstruction. Analyses of the Ge 3d core-level spectra acquired using different photon energies and emission angles consistently reveal the ordered spots to be in a Ge–Ge tilted configuration, which is similar to that in epi Ge(001)-2×1. It was further found that the subsurface layer was actually dominated by Ge, which supported the buckled configuration. The Si atoms were first found in the third surface layer. These Si atoms were further divided into two parts, one underneath the Ge–Ge dimer and one between the dimer row. The distinct energy positions of the Si 2p core-level spectrum were caused by stresses, not by charge alternations.
