Abstract Background and objectives: Orthodontic treatment involves the application of mechanical force to induce periodontal tissue remodeling and ultimately promote tooth movement. It is essential to study the response mechanisms of human periodontal ligament stem cells (hPDLSCs) to improve orthodontic treatment.
Methods: In this study, hPDLSCs treated with compressive force were used to simulate orthodontic treatment. Cell viability and cell death were assessed using the CCK-8 assay and TUNEL staining. Alkaline phosphatase (ALP) and alizarin red staining were performed to evaluate osteogenic differentiation. The binding relationship between IGF1 and METTL14 was assessed using RIP and dual-luciferase reporter assays.
Results: The compressive force treatment promoted the viability and osteogenic differentiation of hPDLSCs. Additionally, m6A and METTL14 levels in hPDLSCs increased after compressive force treatment, whereas METTL14 knockdown decreased cell viability and inhibited the osteogenic differentiation of hPDLSCs treated with compressive force. Furthermore, the upregulation of METTL14 increased m6A levels, mRNA stability, and IGF1 expression. RIP and dual-luciferase reporter assays confirmed the interaction between METTL14 and IGF1. Furthermore, rescue experiments demonstrated that IGF1 overexpression reversed the effects of METTL14 knockdown in hPDLSCs treated with compressive force.
Conclusions: In conclusion, this study demonstrated that compressive force promotes cell viability and osteogenic differentiation of hPDLSCs by regulating IGF1 levels mediated by METTL14.

Abstract Currently, we are witnessing a trend toward subtle or absent hints of luxury, reflecting the rise of inconspicuousness. We seek to address why and how Chinese luxury brand managers, instead of matching conspicuous branding of many Western brands, develop inconspicuous strategies and craft authentic attributes in their brand communication. In the Chinese luxury brand context, we use the ethnographic research method with in-depth interviews, field visits, and photographs of eight Chinese luxury brands with inconspicuous preferences to reveal three main themes contributing to inconspicuousness. First, an inconspicuous approach of Chinese luxury brands is derived from the rise of inconspicuous consumption in China and a rejection of status brands due to being less famous than well-known Western brands, superficiality of status branding, and limited production capability. Second, we argue that inconspicuous branding can encompass developing luxury brands that avoid overtly displaying wealth and social status. Third, we identify three ways of crafting brand authenticity to build inconspicuous brands by using (a) nature to craft quality commitment dimension of authenticity (places and rare raw materials); (b) traditional Chinese craftsmanship and symbols to craft heritage dimension of authenticity; and (c) sincere stories (of how innovations are used in traditional craftsmanship), and the use of sustainability (sustainable raw materials, traditional craftsmanship, luxury production process, and saving resources) to craft sincerity dimension of authenticity in developing inconspicuous brands.

Abstract At present, the most important method for the detection of thyroid hormones in hospitals in China is radioimmunoassay. Besides radioimmunoassay, there are blood test and antibody test methods for thyroid hormone detection. However, after long-term clinical investigations, the accuracy of the results of thyroid hormone detection by radioimmunoassay has been affected by many factors. Possible influencing factors include inaccurate thyroid hormone test results due to improper way of blood collection by nurses and improper way of keeping and transporting blood samples by nurses. Therefore, this paper analyzes and discusses the influencing factors of the accuracy of thyroid hormone (T4) detection by radioimmunoassay technology. In this paper, we conducted research using statistical analysis of clinical data, improved separation methods for quality control in the laboratory, and blood specimen collection methods. Radioimmunoassay occurs with antibodies. Of the 10 batches of 964 cases in the improved separation methods for quality control in the laboratory, 154 mismatched items accounted for 16%, and the error of method and operation only accounted for 5.8% of unmatched specimens, most of which were the biochemical characteristics and clinical manifestations of thyroid hormones. The blood sample collection method research found that mild hemolysis had no significant effect on the measurement results, severe hemolysis had a tendency to affect the results, and blood collection tubes had no effect on the test results. Mild hemolysis refers to the increase in the rate of red blood cell destruction due to various internal and external factors in the body. The symptoms when mild hemolysis occurs are generally not obvious. Severe hemolysis refers to a disease caused by blood group incompatibility, mainly referring to immune hemolysis caused by blood group incompatibility between mother and baby, as well as severe jaundice or severe anemia. The statistical analysis of clinical data found that, among 160 patients, the reasons for the inaccuracy of T4 results using radioimmunoassay technology were as follows: 104 patients were inaccurate due to personal factors, and the results were due to hospital factors. A total of 56 patients had inaccurate results. During the measurement of thyroid hormone, it will be affected by many factors. For this reason, the influencing factors of the accuracy of radioimmunoassay should be clarified, and appropriate measures should be taken to deal with it, so as to give full play to the role of radioimmunoassay and improve the detection.

Abstract As the core of artificial intelligence applications, the research of convolution has become a hot topic in high performance computing. With the rapid development of the emerging SW26010 processor in artificial intelligence, there is an urgent need for high-performance convolution algorithms on the processor. However, the current support of convolution on SW26010 is still rudimentary. The only studies provide sufficient runtime peak performance but lack the adaptability to various convolution scenes. To perfect convolution algorithms on SW26010, we propose a multi-grained matrix-multiplication-mapping convolution algorithm called MG3MConv, which targets the architectural features of SW26010. MG3MConv supports diversified mapping schemes of convolution tasks based on the concept of the thread block proposed in this paper. All the architecture-oriented optimization methods are elaborately designed from four levels to fully exploit the hardware efficiency of SW26010. The experiments show that the hardware efficiency of MG3MConv can reach 84.78% in max, which is 1.75 times compared with that of cuDNN based on NVIDIA K80m GPU. Moreover, MG3MConv can overperform cuDNN in most convolution scenes. We also use six representative CNNs as real-world cases, and the hardware efficiency of MG3MConv reaches up to 67.04% on the VGG network model, which is 1.37 times and 1.96 times that of cuDNN and swDNN, respectively.

Abstract Background: Lenvatinib is an important molecular target drug for the treatment of advanced hepatocellular carcinoma (HCC). However, the application of molecular targeted therapies for HCC also faces some challenges. Cumulative evidence has also shown that curcumol is a potential anti-HCC drug. Curcumol can be used as a chemosensitizer to enhance the antitumor effect of chemotherapeutic drugs. The purpose of our study is to explore the effect of curcumol combined with lenvatinib on HCC.
Methods: The antitumor effects of curcumol or/and lenvatinib on Huh 7 cells of the HCC cell line were examined using the cell counting kit-8 (CCK-8) assay, colony formation assay, and transwell assay. For
Results: In this study, we found that curcumol or lenvatinib could promote HCC cell apoptosis
Conclusion: Curcumol enhances the antitumor effect of lenvatinib on hepatocellular carcinoma cells.
