    Abstract External lubrication of tooling with magnesium stearate (MgSt) is a common strategy to eliminate punch sticking when compressing powders with a high sticking propensity, such as many pure active pharmaceutical ingredients (APIs). We found that it actually led to aggravated punch sticking at low compaction pressures. This counterintuitive phenomenon was explained based on interplay of forces among the punch tip, MgSt, and API. The explanation is supported by the observed effects of pressure and mechanical properties of APIs on this phenomenon.

    Abstract Pipe jacking is a trenchless construction method to achieve forward tunneling and efficient construction of underground structure simultaneously without extensive surface excavation. In the process of pipe jacking construction, the jacking force provided by the hydraulic jacking equipment must overcome the frontal resistance of the cutter head and the frictional resistance between the pipe sections and formation at the same time. In particular, the pipe-soil frictional resistance increases with the increases of jacking distance, buried depth, pipe diameter and the complexity of jacking trajectory. Therefore, it is very important to correctly estimate jacking force in trenchless jacking engineering practice for the smooth implementation of pipe jacking, operation risk and comprehensive cost control. Firstly, the stress states of jacking circular and rectangular pipe sections in the soil are analyzed, and the key influencing factors of their pipe-soil frictional resistance are obtained respectively. Then, the pipe-soil frictional resistance of jacking the circular and rectangular pipe sections with the same external surface area in the dry sandy soil and coal granular layer are tested separately by using the self-developed multifunctional experimental apparatus during trenchless pipe jacking. The results show that the pipe-soil frictional resistances of jacking circular and rectangular pipe sections in the coal granular layer are always smaller than that in the sandy soil under the same experimental conditions, and the corresponding fitting calculation equation of pipe-soil frictional resistances are obtained respectively. Meanwhile, the modified calculation methods of the above pipe-soil frictional resistances are proposed respectively based on the relationship between the lateral pressure coefficient K and the buried depth of pipe section H. Moreover, the disturbed area of soil in the upper part of jacking circular pipe section presents an arc distribution, while the disturbed area of soil in the upper part of jacking rectangular pipe section presents a slightly concave distribution. Due to the different disturbance conditions of soil around the pipe section, the lateral pressure coefficient K should be corrected in the calculation equations of pipe-soil frictional resistance of jacking circular and rectangular pipe sections based on the discrete element numerical simulation analysis by EDEM software. Finally, the pipe-soil frictional resistances obtained by different methods in the sandy soil are compared and analyzed. The calculated values of the modified theoretical calculation method are very close to the experimental test values, while the other methods are smaller than the experimental test values, which makes the rationality of the modified theoretical calculation method of pipe-soil frictional resistance is verified, and some suggestions are also put forward for the value of some coefficients in the relevant empirical estimation equations. The above research achievements systematically compared the states of pipe-soil frictional resistances of jacking circular and rectangular pipe sections based on different research methods, especially for the correct evaluation of jacking force during trenchless pipe jacking, they could provide some valuable references and effective guidance for the subsequent research, engineering practice and further development of trenchless pipe jacking technology.

    Title translation PD-1单抗治疗一例dMMR/MSI-H/TMB-H型结肠癌伴颅内转移瘤患者临床完全缓解.
    Abstract A 70-year-old man had radical surgery for colon cancer one year before the symptoms of memory loss and decreasing cognitive function. Subsequent magnetic resonance imaging revealed a brain mass, which was surgically resected and confirmed to be metastatic intestinal adenocarcinoma. Immunohistochemistry of the primary tumor and brain metastasis showed mismatch repair deficiency. The patient received adjuvant chemotherapy after surgery. However, the brain metastasis relapsed one month after the last chemotherapy. Genetic testing on the resected colon tumor samples confirmed microsatellite instability-high with a high tumor mutation burden by 77.7 muts/Mb. The patient was subsequently treated with programmed death-1 (PD-1) monoclonal antibody pembrolizumab (keytruda). The brain metastatic lesions were completely shrunk, and a complete clinical response was achieved.

    Abstract Background: The survival rate of hepatocellular carcinoma (HCC) is low and the prognosis is poor. Metabolic reprogramming is still an emerging hallmark of cancer, and reprogramming of cholesterol metabolism plays a crucial action in tumor pathogenesis. Increasing evidence suggests that cholesterol metabolism affects the cell proliferation, invasion, migration, and resistance to chemotherapy of HCC. To date, no long noncoding RNA (lncRNA) signature associated with cholesterol metabolism has been developed to predict the outcome of patients with HCC.
    Methods: The RNA-seq data as well as the prognostic and clinical data were obtained from The Cancer Genome Atlas (TCGA) database. We conducted univariate and multivariate analyses to assess cholesterol metabolism-related lncRNAs correlated with the prognosis of patients with HCC in order to construct a prognostic signature. Functional differences between low- and high-risk groups were investigated using genomic enrichment analysis (GSEA). Kaplan-Meier (KM) curves were applied to explore the overall survival (OS) of the low- and high-risk groups. Single-sample genomic enrichment analysis (ssGSEA) was applied to investigate the association between this predictive signature and immune function. We subsequently examined how this signature relates to treatment response in HCC patients.
    Results: A prognostic signature comprising six lncRNAs related to cholesterol metabolism was constructed (
    Conclusions: This cholesterol metabolism-related lncRNA signature can predict the prognosis of HCC patients and guide the clinical management of HCC patients, including immunotherapy.

    Abstract The sea anemone Exaiptasia diaphana (Aiptasia) is a versatile model in studying cellular mechanisms that govern cnidarian-Symbiodiniaceae symbiosis, the foundation of coral reef ecosystems. Here, we provide a protocol to efficiently dissociate adult Aiptasia tissue into a single-cell suspension using enzymatic digestion. We detail steps including washing animals, dissociating tissue with pronase digestion, and evaluating dissociated single cells using fluorescence imaging. This procedure can be applied to other cnidarians, including coral polyps. For complete details on the use and execution of this protocol, please refer to Jinkerson et al. (2022).

    Abstract Alzheimer's disease is the most common neurodegenerative disease. Acanthopanax senticosus, also known as Ciwujia or Siberian ginseng in Chinese, has a wide range of antioxidant and anti-inflammatory activities. The study aims to explore the action mechanism of A. senticosus against Alzheimer's disease using network pharmacology and molecular docking. The active ingredients and targets of A. senticosus were searched through the ETCM database, and Alzheimer's disease-related targets were obtained through the OMIM and GeneCards databases. The Cytoscape 3.7.2 software was used to construct a "drug-component-target" relationship network, and the target genes of A. senticosus against Alzheimer's disease were imported into the String database to establish a protein interaction (PPI) network. The Gene Ontology and Kyoto Encyclopedia of Genes and Genomes gene enrichment analyses were performed through the Metascape database to obtain potential pathways of action of A. senticosus for the treatment of Alzheimer's disease, and the ability of these active ingredients to bind to core targets was then verified by molecular docking. 51 active ingredients were screened from A. senticosus, and 88 effective targets for Alzheimer's disease were screened. Topological and pathway-enrichment analyses revealed that A. senticosus could play a beneficial role in the treatment of Alzheimer's disease by regulating apoptosis and inflammation. Molecular docking results showed that Ciwujianoside B, Chiisanoside, and Ciwujianoside D1 had strong binding abilities to key target proteins (TNFα, IL1β, and CASP3). Collectively, A. senticosus is feasible in the treatment of Alzheimer's disease.
