    Abstract Background Thyroid-associated ophthalmopathy (TAO) is a common orbital inflammatory disease, but the abnormal expression of proteins in tears of TAO patients has not been systematically studied. The purpose of this study is to compare and analyze the total tear protein profile of TAO patients and to provide protein cues for TAO pathogenesis. Methods Tear samples were isolated from 30 TAO patients with obvious ocular surface damage and 30 healthy control subjects. Tear samples from 30 individuals were mixed and divided into three sample pools. Easy nano-scale LC-MS/MS based on labeling-free quantitative technology was utilized to profile tear proteome. Results Here, electrospray ionization mass spectra and SDS-PAGE results confirmed the good parallelisms among samples. A total of 313 proteins were obtained from six tear pools, among them, 103 differential abundance proteins (DAPs) were identified, including 99 up-regulated DAPs (including APOA1, HV103, IGH, and Transferrin variant) and four down-regulated DAPs (including FABA, VCC1, NUCB2, and E-cadherin) in the TAO group compared with the control group. GO analysis showed that up-regulated DAPs were mainly enriched in lipid metabolism and platelet molecular function, and down-regulated DAPs were involved in binding, cell junction, and cellular process. KEGG results indicated that DAPs were involved in 117 kinds of signal transduction pathways, among which the immune-related pathway of complement and coagulation cascades had the greatest relevance. Conclusion In conclusion, label-free LC-MS/MS is an effective strategy for profiling tear proteins component. Our study provides proteins and pathways altered in TAO and provides protein cues for further study on the precise mechanism of TAO pathogenesis.

    Abstract Pavements are closely related to the livelihoods of urban residents as an important part of the urban road system. Based on the early field measurements of our team, we found that plant spacing, pavement orientation, and pavement material influence the pavement thermal environment. Our objective was to quantitatively evaluate the influence of various factors on pavements in Guangzhou along West Zhonghuan Road, Guangzhou University. Based on our team’s previous research, environment-met (ENVI-MET) numerical simulation software was used to simulate pavements under different road orientations, underlying surface materials, and plant distances. Quantitative analysis of the effects of different factor combinations on the temperature, humidity, wind speed, physiological equivalent temperature (PET), and radiant temperature (Tmrt) was performed. The results show that among the various factors affecting the thermal environment of pavement, street tree spacing was found to have the greatest effect on the thermal environment of pavement. The effect of direction is negligible; however, it has a significant effect on the wind speed. There was no significant difference in the air temperature and relative humidity between concrete and asphalt. The average air temperature of red floor tile was slightly lower than that of asphalt and concrete. Reducing the spacing of street trees can effectively reduce the penetration of direct solar radiation, physiological equivalent temperature (PET), and temperature (Tmrt) and improve pedestrian walking comfort. The results can serve as a reference for pavement design in Guangzhou and improve the wellbeing of citizens and promote environmental sustainability.

    Abstract The Angelman Syndrome (AS) is an extreme neurodevelopmental disorder without effective treatments. While most patients with this disease can be diagnosed by genetic testing, there are still a handful of patients have an unrecognized genetic cause for their illness. Thus, novel approaches to clinical diagnosis and treatment are urgently needed. The aim of this study was to identify and characterize differentially expressed genes involved in AS and built potential diagnostic panel for AS by NGS sequencing. A multi-cohort analysis framework was used to analyze stem cell-derived neurons from AS patients in GSE160747 dataset. We identified three differentially expressed genes (ACTN1, ADAMTS2, SLC30A8) differentiates AS patients from controls. Moreover, we validated the expression patterns of these genes in GSE146640, GSE120225. Receiver operating characteristic (ROC) curves analysis demonstrated that these genes could function as potential diagnostic biomarkers [AUC = 1 (95% CI 1–1)]. This study may provide new approach for diagnosing patients with AS and helping to develop novel therapies in treating AS patients.

    Abstract Cataract, an eye disease that threatens the health of millions of people, brings about severe economic burden for patients and society. MicroRNA (miR)-378a-5p and miR-630 were recognized as essential regulators in multiple cancers. However, the exact functions of miR-378a-5p and miR-630 in cataract are still unclear. The expression of miR-378a-5p, miR-630, and E2F transcription factor 3 (E2F3) in tissues and cells was measured by quantitative real-time polymerase chain reaction. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay was used to evaluate cell viability. Flow cytometry was conducted to analyze cell apoptosis. The interaction between E2F3 and miR-378a-5p or miR-630 was confirmed by dual-luciferase reporter assay. The expression of proteins E2F3, B cell lymphoma (Bcl-2), Bcl-2 associated X (Bax), and cleaved caspase 3 was detected by western blot assay. The expression of miR-378a-5p and miR-630 was up-regulated whereas E2F3 was down-regulated in human cataract lens tissues compared with normal lens tissues. Depletion of miR-378a-5p or miR-630 enhanced proliferation and reduced apoptosis of human lens epithelial cells. Interestingly, up-regulation of E2F3 exhibited the same trend. Next, dual-luciferase reporter assay validated the interaction between E2F3 and miR-378a-5p or miR-630. The rescue experiments further revealed that E2F3 knockdown could recover miR-378a-5p, and miR-630 inhibitor induced promotion of cell proliferation and inhibition of apoptosis in cataract. miR-378a-5p and miR-630 repressed proliferation and induced apoptosis of lens epithelial cells by targeting E2F3 in cataract, representing a prospective alternative therapy for cataract.

    Abstract The GF-7 satellite is China’s first civil sub-meter resolution stereo mapping satellite, aiming at 1:10,000-scale mapping. To achieve this goal, apart from the stereo optical cameras that reach sub-meter resolution, the GF-7 satellite is equipped with a laser altimetry system capable of obtaining three-dimensional laser points (LPs) with high elevation accuracy. However, the combination of laser altimetry data and optical stereo images has not been thoroughly studied. In this paper, we exploit the images recorded by the highly integrated laser footprint cameras and propose a hierarchical phase correlation method based on a geographic pyramid for the registration of laser altimetry data and high-resolution optical stereo images, which lays a solid foundation for the following combined adjustment. Experiments show that the proposed registration method can automatically locate the LPs on high-resolution stereo images and meet the requirements of bundle adjustment. A series of bundle adjustment experiments were carried out, showing that laser altimetry data can significantly enhance the vertical accuracy of optical image stereo mapping and that elevation accuracy can reach roughly 1.0 m (RSME) without ground control points. Therefore, this study could be a good guide for global high-precision DSM acquisition with the GF-7 satellite.

    Abstract The Gaofen-7 (GF-7) satellite uses a two-beam laser altimetry system in which each beam is equipped with a laser footprint camera (LFC) to provide geometric processing of the laser footprint images that assist in optical image stereo mapping. Because of the violent vibrations during launch and the difference in the environment before and after entering orbit, the key parameters for geometric processing of the laser footprint images may change, which will cause large geolocation errors. Therefore, it is essential to carry out on-orbit calibration and validation for the laser footprint cameras. This study first constructs a rigorous geometric positioning model for the LFC of the GF-7 satellite and analyses various error sources that affect the geometric positioning accuracy of laser footprint images. Then, a comprehensive calibration method, which effectively eliminates the distortion of the LFC optical system, and the positioning error caused by the long-period jitter of the satellite platform, is proposed based on the multi-scene images combined with image simulation. The proposed method can effectively eliminate various errors that affect the geometric positioning accuracy of the GF-7 laser footprint image. The internal geometric positioning accuracy of the calibrated LFC is better than 0.7 pixels, and the absolute geometric positioning accuracy is within 6.0 m after using precise post-processing orbital and attitude data. Our study will contribute to the processing and application of laser altimetry data from the GF-7 satellite.

    Abstract Rock glaciers are typical periglacial landforms with tongue or lobate morphological shapes and characterized by the distinct front, lateral margins, and often by ridge-and-furrow surface topography textures as well as kinematic characteristics, widely distributed in alpine environments. Multitemporal Synthetic aperture radar interferometry (MT-InSAR), is a remote sensing technique with demonstrated effectiveness for detecting landform kinematics. However, its application to rock glaciers is challenged by temporal decorrelation and atmospheric phase noises due to complex topography and snow cover. We designed a quadtree segmentation and parallel computing-based MT-InSAR method to improve the quality and efficiency of deformation measurement of rock glaciers. We applied the method to a rock glacier inventory of the Nyainqêntanglha Range, China, derived from high-resolution Gaofen-2 images, to quantify the activity rate of each rock glacier. Results showed that 32.1% (6,389) of the identified rock glaciers exhibited slope-parallel deformation rates exceeding 100 mm/y. The activities of the rock glaciers exhibited strong correlations with their distance to glaciers, precipitation, freeze–thaw magnitude, and permafrost occurrence probability. The results demonstrate the effectiveness of the developed segmentation-parallel MT-InSAR method for monitoring rock glacier deformation over a large region.

    Abstract The RAN Guanine Nucleotide Release Factor (RANGRF) gene encodes the protein MOG1, which binds to Nav1.5 and facilitates its transport to the cell membrane. Nav1.5 mutations have been linked to various cardiac arrhythmias and cardiomyopathy. To investigate the role of RANGRF in this process, we utilized the CRISPR/Cas9 gene editing system to generate a homozygous RANGRF knockout hiPSC line. The availability of the cell line will prove to be an invaluable asset in the study of disease mechanisms and the testing of gene therapies for cardiomyopathy.

    Abstract Gushukang (GSK) is a traditional herbal compound used in Chinese medicine for the treatment of osteoporosis. Numerous studies have been conducted to elucidate the effects of GSK, but the mechanisms underlying these effects remain unclear. In the present study, we cultured osteoblasts and osteoclasts with low and high doses of GSK, and also administered 3-month-old mice with 4 and 8 g/kg/day of GSK solution. Gushukang was found to promote osteoblast differentiation and inhibit osteoclast differentiation in vitro. In vivo, mice in the GSK treatment groups showed an increase in bone mass, as measured by micro-computed tomography (Micro-CT). Tartrate resistant acid phosphatase (TRAP) staining and osteocalcin (OCN) staining experiments revealed decreased bone resorption and increased bone formation in the GSK treatment groups. In addition, we found a novel effect of GSK—it could induce type-H vessel formation in mice. The underlying mechanisms of these actions were further explored at the molecular level to investigate whether these effects were due to an overexpression of the hypoxia inducible factor-1 (HIF-1α). Our findings indicate the utility of GSK as a therapeutic for the prevention of osteoporosis.
