    Abstract Abstract Background Meningiomas are common primary brain tumours, with macrophages playing a crucial role in their development and progression. This study aims to identify module genes correlated with meningioma‐associated macrophages and analyse their correlation with clinical features and immune infiltration. Methods We analysed single‐cell RNA sequencing (scRNA‐seq) data from two paired meningioma and normal meninges to identify meningioma‐associated macrophages. High‐dimensional weighted gene co‐expression network analysis (hdWGCNA) was employed to identify module genes linked to these macrophages, followed by functional enrichment and pseudotime trajectory analyses. A machine learning‐based model using the module genes was developed to predict tumour grades. Finally, meningiomas were classified into two molecular subtypes based on the module genes, followed by a comparison of clinical characteristics and immune cell infiltration. Results Meningiomas exhibited a significantly higher proportion of macrophages than normal meninges, including novel macrophage clusters referred to as meningioma‐associated macrophages. The hdWGCNA analysis of macrophages within meningiomas unveiled 12 distinct modules, with the blue, black, and turquoise modules closely correlated with the meningioma‐associated macrophages. Hub genes within these modules were enriched in immune regulation, cellular communication, and metabolism pathways. Machine learning analysis identified 13 module genes (RSBN1, TIPRL, ATIC, SPP1, MALSU1, CDK1, MGP, DDIT3, SUPT16H, NFKBIA, SRSF5, ATXN2L, and UBB) strongly correlated with meningioma grade and constructed a predictive model with high accuracy and robustness.     Abstract Vaccines are the most promising tools for maintaining public health. Most emerging human infectious diseases are caused by viruses originating from an animal reservoir via zoonotic transmission. Therefore, zoonotic virus spillover and spread in humans have become global health threats. Nanoparticle-based vaccines are ideal for antigen delivery, as adjuvants, and as viral structure mimics. Nanoparticles benefit vaccine design and are utilized to protect the antigen cargo, and increase the immunogenicity and efficacy. Therefore, nanoparticle vaccines are a novel method of immunization by which optimal immune responses are elicited. Herein we review current approaches in the development of nanoparticle vaccines and highlight the role of nanoparticle vaccines against zoonotic viral diseases.

    Abstract Abstract Comprehensive and accurate assessments of fish composition and diversity are essential for understanding fish ecology and resource management. Traditional fish surveys generally involve capturing organisms, which is invasive for the biological community under study and conflicts with the original intention of biodiversity conservation. Environmental DNA (eDNA) metabarcoding has become an integrated method for monitoring fish species without disturbing ecosystems. However, due to the serious nonspecific amplification of primers in eDNA‐based monitoring, many non‐fish sequences, usually human sequences, are also amplified, which results in serious data wastage and an increase in monitoring costs. We designed new universal primers for freshwater fish by analyzing the whole mitochondrial genome of Chinese freshwater fish. The performance of the primers was compared using an in silico polymerase chain reaction, followed by an in vitro metabarcoding analysis using eDNA from the Yangtze River, which is the third largest river in the world and harbors many freshwater fish species. We found that the mitochondrial 12S region is the most suitable metabarcoding gene marker for both Chinese and other freshwater fish. The minor change at the 3′‐end of the primer can greatly reduce the nonspecific amplification and improve the effectiveness of eDNA metabarcoding. Even small changes in primers may have qualitative and quantitative effects on the detected biodiversity, which should be considered in experimental design and data interpretation. These results will help with primer design and selection for eDNA‐based fish surveys, and consequently support the conservation of freshwater biodiversity.

    Abstract Characteristic Chinese towns are “green ecology” innovation space units based on the background of the ‘Beautiful China Initiative’ (BCI), new urbanization, supply-side structural reform and the implementation of rural revitalization strategies. In this paper, spatial analysis models such as kernel density analysis, spatial autocorrelation analysis, the local correlation index and ArcGIS 10.5 are used to analyze the spatial layout and structural characteristics of the green development evolution of characteristic towns and to explore their spatial differentiation characteristics and laws, internal influencing factors and mechanisms. The analysis of the spatial distribution kernel density shows that regional economic development is an important influencing factor that affects the layout of the characteristic towns. Spatial autocorrelation analysis indicates that the spatial distribution of the characteristic towns does not have random distribution characteristics but is clustered in areas of similar scale. The results show the following: (1) The spatial layout of characteristic towns is generally a cohesive distribution with obvious agglomeration trends and differences, showing the characteristics of “dense in the southeast, sparse in the northwest” and “overall agglomeration, relying on economy, along the coast and along traffic arteries, spreading around cities, and differentiating by scenery”. (2) The high-density core area and sub-high-density area of characteristic towns are situated along the coast and along traffic arteries, are characterized by resource endowments and economic development, and are distributed along urban agglomerations and metropolitan areas. (3) The spatial pattern of green development evolution is organically coupled across three dimensions: location, industry and community.     Abstract IntroductionA superficial cervical vaginal myofibroblastoma is a rare mesenchymal tumor, originating from the superficial stroma of the vagina and cervix. This study reports a patient, who was diagnosed with endometrioid carcinoma and a concomitant benign superficial cervicovaginal myofibroblastoma.Case presentationA 53-year-old female with endometrial carcinoma was admitted to the Department of Gynecology of our hospital. She had a history of breast cancer on May 23, 2010, and took toremifene citrate for 41 months. Radical resection of the endometrial carcinoma was performed at our hospital. Based on the pathological findings, she was postoperatively diagnosed with endometrial adenocarcinoma with superficial cervical vaginal myofibroblastoma. The patient continued receiving postoperative breast cancer treatment. She underwent follow-up for 23 months. No recurrence or metastasis of the endometrial cancer or superficial cervical vaginal myofibroblastoma was observed.ConclusionThere were similarities between superficial cervical vaginal myofibroblastoma and other mesenchymal tumors of the female genital tract. Superficial cervical vaginal myofibroblastomas have a good prognosis, and the combination of tissue morphology and immunohistochemistry helped establish a definitive diagnosis.

    Abstract Abstract Despite continued advances in prevention and treatment strategies, cardiovascular diseases (CVDs) remain the leading cause of death worldwide, and more effective therapeutic methods are urgently needed. Polygonatum is a traditional Chinese herbal medicine with a variety of pharmacological applications and biological activities, such as antioxidant activity, anti-inflammation, antibacterial effect, immune-enhancing effect, glucose regulation, lipid-lowering and anti-atherosclerotic effects, treatment of diabetes and anticancer effect. There has also been more and more evidence to support the cardioprotective effect of Polygonatum in recent years. However, up to now, there has been a lack of comprehensive studies on the active ingredients and their pharmacotoxicological effects related to cardiovascular diseases. Therefore, the main active components of Polygonatum (including Polysaccharides, Flavonoids, Saponins) and their biological activities were firstly reviewed in this paper. Furthermore, we summarized the pharmacological effects of Polygonatum’s active components in preventing and treating CVDs, and its relevant toxicological investigations. Finally, we emphasize the potential of Polygonatum in the prevention and treatment of CVDs.

    Abstract Background and Objectives: The physiological phenomenon peculiar to women, namely menopause, makes the occurrence of left ventricular hypertrophy (LVH) in postmenopausal hypertensive women more characteristic. Less is known about the risk of developing LVH in Chinese postmenopausal hypertensive women. Thus, the present study was intended to design a nomogram for predicting the risk of developing LVH in Chinese postmenopausal hypertensive women. Materials and Methods: Postmenopausal hypertensive women aged between 49 and 68 years were divided into either the training set ( n = 550) or the validation set ( n = 284) in a 2:1 ratio. Patients in the validation set were followed up for one year. A stepwise multivariable logistic regression model was used to assess the predictors of LVH in postmenopausal women with hypertension. The best-fit nomogram was executed using R software. The calibration and decision curve were employed to verify the predictive accuracy of the nomogram. The results were evaluated in the validation set. Results : Menopause age (OR = 0.929, 95% CI 0.866–0.998, p = 0.044), BMI (OR = 1.067, 95% CI 1.019–1.116, p = 0.005), morning systolic blood pressure (SBP: OR = 1.050, 95% CI 1.032–1.069, p = 0.000), morning diastolic BP (DBP OR = 1.055, 95% CI 1.028–1.083, p = 0.003), angiotensin II receptor blocker (ARB) utilization rate (OR = 0.219, 95% CI 0.131–0.365, p = 0.000), LDL-C (OR = 1.460, 95% CI 1.090–1.954, p = 0.011) and cardio-ankle vascular index (CAVI) (OR = 1.415, 95% CI 1.139–1.757, p = 0.028) were associated with LVH in postmenopausal hypertension patients. The nomogram model was then developed using these variables. The internal validation trial showed that the nomogram model described herein had good performance in discriminating a C-index of 0.881 (95% CI: 0.837–0.924) and high quality of calibration plots.     Abstract Introduction: Aiming at the problem of structural damage of top beam and column in the actual bearing process of top coal caving hydraulic support caused by uneven roof pressure in the process of fully mechanized caving mining in thick coal seam coal mine, this paper analyzes the dynamic bearing performance of ZF20000/26/48 top coal caving hydraulic support based on the working conditions given by the 101 working face of Shaanxi Zheng-tong Coal Mine.Methods: Genetic algorithm was used to optimize the top beam of hydraulic support.Results: Under the same loading conditions, the maximum stress of the optimized support is reduced by 22.28MPa, and the stress of the top beam is more uniform, and the maximum stress is reduced by 24.53MPa.Discussion: The purpose of optimization design is achieved, and the bearing performance of hydraulic support is improved,A new method using genetic algorithm to optimize the top beam of hydraulic support is developed.

    Abstract IntroductionThe design of high-pressure optical windows is one of the key tasks in deep-sea exploration, as it directly determines the reliability and maximum operating depth of underwater optical equipment.MethodIn this paper, mechanical modeling and finite element analysis methods are employed to study optical windows.Results and discussionResults show that changes in boundary conditions during loading significantly affect the stress distribution of the window. Based on this conclusion, an optimization design method is proposed, which involves the use of transition materials to reduce the impact of base deformation on the window. The study further analyzes the effects of the transition material’s Young’s modulus, Poisson’s ratio, thickness, and friction coefficient on the stress of the optical window. A calculation method for material selection criteria is proposed to select appropriate transition materials, and actual materials are used for verification. Finally, the reliability of the optimization design scheme based on transition materials is confirmed through high-pressure experiments.
