Abstract Background. The dysregulation of circular RNAs (circRNAs) participates in the malignant progression of multiple cancers, including osteosarcoma (OS). However, the role of circ_0097271 in OS development remains unclear. We thus aimed at unveiling the functional role and mechanism of circ_0097271 in OS. Methods. The expressions of circ_0097271, miR-640, and MCAM in OS were analyzed by qPCR. Cell proliferation and migration were inspected by CCK-8 assay, colony formation assay, and Transwell assay. Circ_0097271’s role in vivo was assayed by establishing animal models. The predicted binding relationship between miR-640 and circ_0097271 or MCAM was verified by dual-luciferase reporter or RIP assay. Results. Circ_0097271’s expression was enhanced in OS samples and cells. The knockdown of circ_0097271 restrained OS cell growth and migration, and its downregulation also blocked solid tumor growth in vivo. Circ_0097271 targeted miR-640 and negatively modulated miR-640 expression. MiR-640 was poorly expressed in OS, and its depletion recovered OS cell growth and migration that were repressed by circ_0097271 knockdown. MiR-640 bound to MCAM 3’UTR and thus suppressed MCAM expression. MCAM knockdown repressed OS cell growth and migration, while additional miR-640 depletion partially abolished the anticancer effects of MCAM knockdown in OS cells. Conclusion. Circ_0097271 is an oncogenic driver and contributes to OS development via targeting the miR-640/MCAM pathway, which provides a potential opinion for OS treatment.

Abstract Summary: RNA transcripts can anneal with template DNA strands to form RNA-DNA hybrids (or R-loops if the non-template DNA strands exist), which play a variety of roles in many physiological processes. Here, we provide an accessible and reproducible approach for immunofluorescent staining of RNA-DNA hybrids with the S9.6 antibody in spread meiotic nuclei of Saccharomyces cerevisiae. This protocol allows the examination of RNA-DNA hybrids as clearly distinguishable foci and the colocalizations of RNA-DNA hybrids with other proteins. For complete details on the use and execution of this protocol, please refer to Yang et al. (2021).

Abstract Corporate energy consumption and emissions are major contributors to pollution and greenhouse gas emissions. Therefore, corporate energy conservation and emission reduction are essential for sustainable development and low-carbon transformation in China. With pressing global environmental issues and the mounting pressure for carbon neutrality, it is increasingly important for firms to disclose environmental information. However, research on the relationship and mechanisms between corporate environmental information disclosure (EID) and corporate performance is insufficient. This study examines the effect of EID on corporate environmental performance (EP) and financial performance (FP) using a two-way fixed-effect model. For this purpose, this study considers the panel data of 1125 Chinese A-share listed companies from 2009 to 2021. Additionally, we explore the underlying mechanisms using mediation models. The results reveal that EID can significantly improve corporate performance through green innovation (GI). Moreover, EID can enhance corporate GI, which improves the financial performance of high-polluting firms and the environmental performance of non-high-polluting firms. According to these results, it is imperative to continuously improve corporate EID systems and encourage GI in enterprises. Thus, firms should take the initiative to disclose environmental information. There is a pressing need to enhance the effectiveness of the capital market, therefore, firms with higher levels of EID and better environmental performance can gain greater benefits.

Abstract Accurate surface water mapping is crucial for rationalizing water resource utilization and maintaining ecosystem sustainability. However, the diverse shapes and scales of water bodies pose challenges in automatically extracting them from remote sensing images. Existing methods suffer from inaccurate lake boundary extraction, inconsistent results, and failure to detect small rivers. In this study, we propose a dual-stream parallel feature aggregation network to address these limitations. Our network effectively combines global information interaction from the Swin Transformer network with deep local information integration from Convolutional Neural Networks (CNNs). Moreover, we introduce a deformable convolution-based attention mechanism module (D-CBAM) that adaptively adjusts receptive field size and shape, highlights important channels in feature maps automatically, and enhances the expressive ability of our network. Additionally, we incorporate a Feature Pyramid Attention (FPA) module during the advanced coding stage for multi-scale feature learning to improve segmentation accuracy for small water bodies. To verify the effectiveness of our method, we chose the Yellow River Basin in China as the research area and used Sentinel-2 and Sentinel-1 satellite images as well as manually labelling samples to construct a dataset. On this dataset, our method achieves a 93.7% F1 score, which is a significant improvement compared with other methods. Finally, we use the proposed method to map the seasonal and permanent water bodies in the Yellow River Basin in 2021 and compare it with existing water bodies. The results show that our method has certain advantages in mapping large-scale water bodies, which not only ensures the overall integrity but also retains local details.

Abstract Objective: To study the modulation effect of blu-ray irradiation combined with comprehensive therapy on serum indexes of neonatal erythrocyte glucose-6-phosphate dehydrogenase deficiency-induced hyperbilirubinemia. Methods: A total of 42 cases of neonates with erythrocyte glucose-6-phosphate dehydrogenase deficiency-induced hyperbilirubinemia were chosen for study and randomly divided into observation group (n=21) and control group (n=21). Observation group received blu-ray irradiation combined with comprehensive treatment and control group only received routine treatment. Then bilirubin levels, bilirubin encephalopathy condition, anemia condition and oxidative stress degree of two groups were compared. Results: 12 h, 24 h and 48 h after treatment, serum TBIL, DBIL, IBIL, Hb, GSH and CAT contents of both groups showed decreasing trend and MDA contents showed increasing trend; serum TBIL, DBIL, IBIL, Hb, GSH and CAT contents of observation group were lower than those of control group and MDA contents were higher than those of control group. 6 d, 7 d and 8 d after treatment, serum S100 β and NSE contents of both groups showed decreasing trend and serum S100 β and NSE contents of observation group were lower than those of control group. Conclusion: Blu-ray irradiation combined with comprehensive therapy helps to reduce bilirubin levels of neonatal erythrocyte glucose-6-phosphate dehydrogenase deficiency-induced hyperbilirubinemia and protect nerve function, but it will aggravate anemia condition and oxidative stress degree, and needs attention and intervention in clinical practice.

Abstract Nanoparticles (NPs) can be extremely effective in the early diagnosis and treatment of cancer due to their properties. The nanotechnology industry is developing rapidly. The number of multifunctional NPs has increased in the market and hundreds of NPs are in various stages of preclinical and clinical development. Thus, the mechanism underlying the effects of NPs on biological systems has received much attention. After NPs enter the body, they interact with plasma proteins, tumour cell receptors, and small biological molecules. This interaction is closely related to the size, shape, chemical composition and surface modification properties of NPs. In this review, the effects of the size, shape, chemical composition and surface modification of NPs on the biological effects of NPs were summarised, including the mechanism through which NPs enter cells, the resulting oxidative stress response, and the interaction with proteins. This review of the biological effects of NPs can not only provide theoretical support for the preparation of safer and more efficient NPs but also lay the foundation for their clinical application.

Abstract Effects of heat moisture treatment (HMT), extrusion treatment (ET), and the combination treatment (HMT-ET) on microstructural, physicochemical properties, and starch digestibility of brown rice flour (BRF) were investigated. With a rise in resistant starch (RS), melting temperature, and a decrease in swelling capacity (SC), peak viscosity, and apparent amylose content (AAC), the HMT-ET BRF showed a significant lower expected glycemic index (eGI) than HMT and ET. XRD and FTIR results showed ET, HMT-ET caused the transition of starch crystals from amorphous to crystalline region, suggesting the formation of the starch-lipid complex. The analysis of DSC and RVA proved HMT-ET flours induced starch gelatinization and inhibited the starch retrogradation of BRF compared with the other three flours. Correlation analysis suggested that the combined effect of HMT and ET was response for the changes in physicochemical properties and reduction of in vitro starch digestibility. Overall, the BRF after HMT-ET with improved physicochemical properties and starch digestibility could be better utilized as a good substitute for carbohydrate sources.
