Abstract Human immunodeficiency virus (HIV) infection poses substantial medical risks to global public health. An essential strategy to combat the HIV epidemic is timely and effective virus testing. CRISPR-based assays combine the highly compatible CRISPR system with different elements, yielding portability, digitization capabilities, low economic burden and low operational thresholds. The application of CRISPR-based assays has demonstrated rapid, accurate, and accessible means of pathogen testing, suggesting great potential as point-of-care (POC) assays. This review outlines the different types of CRISPR/Cas systems based on Cas proteins and their applications for the detection of HIV. Additionally, we also offer an overview of future perspectives on CRISPR-based methods for HIV detection, including advances in nucleic acid amplification-free testing, improved personal testing, and refined testing for HIV genotypes and drug-resistant strains.

Abstract The purpose of this study was to investigate the parenchymal changes in idiopathic pulmonary fibrosis (IPF) caused by massive fibroblastic infiltration and proliferation in lung tissue. Galectin-1 (Gal-1) has been reported to be involved in angiogenesis and fibrosis via modification of TGF-b receptor signaling pathways. However, it remains unknown whether Galectin-1 plays a critical role in IPF. In the current study, we aimed to identify Gal-1 as a crucial fibrotic protein in IPF process. Murine lung fibroblast was pre-treated using Gal-1 inhibitor OTX-008 or overexpression of Gal-1 and then activated using transforming growth factor-beta (TGF-β). Adult male C57BL/6J mice were conducted intratracheal injection of bleomycin (BLM) for lung fibrosis. Mice were conducted OTX-008 administration. Gal-1 expression, fibroblast activation and proliferation, extracellular matrix (ECM), lung fibrosis, lung histology and pulmonary function were investigated respectively. We demonstrated that Gal-1, as a positive pro-fibrotic marker, could promote lung fibroblast activation and proliferation. Inhibition of Gal-1 reduced fibroblast activation and proliferation through negative regulation of TGF-β/Erk1/2 and AKT pathway. In vivo, Gal-1 inhibition ameliorates lung fibroblast accumulation and protects lung histology and function. Gal-1 is verified to be a pro-fibrotic gene in IPF pathogenesis, which promotes fibroblast activation and proliferation via TGF-β/Erk1/2 and AKT pathway. Moreover, inhibition of Gal-1 in lung fibrosis model attenuates lung fibroblast bioactivity and reduces ECM, leading to improved pulmonary histology and function. Hence, knockdown of Gal-1 in IPF may be a promising target therapy.

Abstract This article proposes a survival model based on graph convolutional networks (GCNs) with geometric graphs directly constructed from high-dimensional features. First, we clarify that the graphs used in GCNs play an important role in processing the relational information of samples, and the graphs that align well with the underlying data structure could be beneficial for survival analysis. Second, we show that sparse geometric graphs derived from high-dimensional data are more favorable compared with dense graphs when used in GCNs for survival analysis. Third, from this insight, we propose a model for survival analysis based on GCNs. By using multiple sparse geometric graphs and a proposed sequential forward floating selection algorithm, the new model is able to simultaneously perform survival analysis and unveil the local neighborhoods of samples. The experimental results on real-world datasets show that the proposed survival analysis approach based on GCNs outperforms a variety of existing methods and indicate that geometric graphs can aid survival analysis of high-dimensional data.

Abstract Poly(ionic liquid) (PIL)-based porous membranes are extensively investigated as soft polymer actuators. While PILs have shown significant advancements in membrane fabrication and stabilization of metal nanoparticles (MNPs), research on integrating MNPs into porous membranes to achieve actuation behavior under multiple stimuli is limited. Herein, this work presents a new paradigm for designing a porous PIL-polyacrylic acid (PAA) membrane with a distinct MNP gradient via a top-bottom diffusion approach involving a metal salt precursor solution and NaBH

Abstract BACKGROUND As an important aluminosilicate, trace element contents affect the performance of the product. The kaolin standard substances GBW03121, GBW03122, GBW03122a lack the recommended values of ten elements such as arsenic and antimony. Similar rock standard materials were used for monitoring the determination procedure, but the accurate results of the kaolin samples may be affected. OBJECTIVES To establish a method for accurate determination of 10 trace elements in kaolin sample such as arsenic and antimony. METHODS With microwave digestion technology, the nitrate-hydrofluoric acid system and nitrate-hydrofluoric-peroxide system were compared for the procedural heating conditions and dissolution time of microwave digestion. Interference factors of the inductively coupled plasma mass spectrum were also investigated. RESULTS The detection limits of 10 elements were 0.01-0.09mg/kg, and measurement limits were 0.03-0.30mg/kg. The accuracy and precision of the method were directly verified by using rock reference materials. The element recovery was between 90.9% and 103.2%, and the relative standard deviation (RSD) was between 1.2% and 5.8%. At the same time, a comprehensive evaluation of the uncertainty of the method was carried out, and the method was proved to be accurate and reliable. CONCLUSIONS This method has little acid dosage, shortens the measurement time, and reduces the damage on the environment and human, making it suitable for the batch analysis of 10 trace elements in kaolin samples. The method also provides a reference for the determination of the certified values of 10 trace elements such as As and Sb in the national standard material of kaolin.

Abstract The research around the public9s usage of masks to prevent the spread of COVID-19 is developing quickly. In this work, we analyzed data from 50 nations to assess the long-term effectiveness of mask policies with different levels using the Poisson regression model and generalized linear mixed model. Over the long term, stricter obligatory mask regulations were linked to more stable patterns and slower increases in Covid-19 case occurrences. The mitigation of disease transmission by mask policies was shown to have substantial major impacts throughout the entire year of 2020, whereas the incidence of illness displays increasing trends over time under various policies. When compared to no mask policy deployment, mask policies might reduce incidence growth by 13.5% to 17.8%, although the incidence under every policy climbed 1.5% to 1.9% on average every ten days. The mask policy is effective in controlling illness, according to the bulk of the data shown above. This result confirms the mask policy9s importance as a governing approach in the context of the worldwide pandemic.
