    Abstract The widespread distribution of nanoplastics and nanomaterials in aquatic environments is of great concern. Nanoplastics have been found to modulate the toxicity of other environmental pollutants in organisms, while few studies have focused on their influences on nanomaterials. Thus, this study evaluated the influences of polystyrene (PS) nanoplastics on the toxicity of silver nanoparticles (AgNPs) to zebrafish (Danio rerio) embryos, including acute toxicity, oxidative stress, apoptosis, immunotoxicity, and metabolic capability. The results showed that the presence of PS nanoplastics could act as a carrier of the co-existing AgNPs in waters. The release ratio of Ag+ from AgNPs was up to 4.23%. The lethal effects of AgNPs on zebrafish embryos were not significantly changed by the co-added PS nanoplastics. Whereas, the alterations in gene expression related to antioxidant and metabolic capability in zebrafish (sod1, cat, mt2, mtf-1, and cox1) caused by AgNPs were significantly enhanced by the presence of PS nanoplastics, which simultaneously lowered the apoptosis and immunotoxicity (caspase9, nfkβ, cebp, and il-1β) induced by AgNPs. It suggests the presence of PS nanoplastics suppressed the AgNPs-induced genotoxicity in zebrafish. The released Ag+ from AgNPs may be responsible for the toxicity of AgNPs in zebrafish, while the subsequent absorption and agglomeration of AgNPs and the released Ag+ on PS nanoplastics may alleviate the toxicity.

    Abstract Abstract Proprotein convertase subtilisin/kexin type 9 (PCSK9) has evolved as a pivotal enzyme in lipid metabolism and a revolutionary therapeutic target for hypercholesterolemia and its related cardiovascular diseases (CVD). This comprehensive review delineates the intricate roles and wide-ranging implications of PCSK9, extending beyond CVD to emphasize its significance in diverse physiological and pathological states, including liver diseases, infectious diseases, autoimmune disorders, and notably, cancer. Our exploration offers insights into the interaction between PCSK9 and low-density lipoprotein receptors (LDLRs), elucidating its substantial impact on cholesterol homeostasis and cardiovascular health. It also details the evolution of PCSK9-targeted therapies, translating foundational bench discoveries into bedside applications for optimized patient care. The advent and clinical approval of innovative PCSK9 inhibitory therapies (PCSK9-iTs), including three monoclonal antibodies (Evolocumab, Alirocumab, and Tafolecimab) and one small interfering RNA (siRNA, Inclisiran), have marked a significant breakthrough in cardiovascular medicine. These therapies have demonstrated unparalleled efficacy in mitigating hypercholesterolemia, reducing cardiovascular risks, and have showcased profound value in clinical applications, offering novel therapeutic avenues and a promising future in personalized medicine for cardiovascular disorders. Furthermore, emerging research, inclusive of our findings, unveils PCSK9’s potential role as a pivotal indicator for cancer prognosis and its prospective application as a transformative target for cancer treatment. This review also highlights PCSK9’s aberrant expression in various cancer forms, its association with cancer prognosis, and its crucial roles in carcinogenesis and cancer immunity.     Abstract Post‐meiotic sperm ageing, both before ejaculation and after ejaculation, has been shown to negatively affect offspring fitness by lowering the rate of embryonic development, reducing embryonic viability and decreasing offspring condition. These negative effects are thought to be caused by intrinsic factors such as oxidative stress and ATP depletion or extrinsic factors such as temperature and osmosis. Effects of post‐ejaculation sperm ageing on offspring fitness have so far almost exclusively been tested in internal fertilizers. Here, we tested whether intrinsic post‐ejaculation sperm ageing affects offspring performance in an external fertilizer, the Atlantic salmon Salmo salar. We performed in vitro fertilizations with a split‐clutch design where sperm were subjected to four post‐ejaculation ageing treatments. We varied the duration between sperm activation and fertilization while minimizing extrinsic stress factors and tested how this affected offspring fitness. We found no evidence for an effect of our treatments on embryo survival, hatching time, larval standard length, early larval survival or larval growth rate, indicating that intrinsic post‐ejaculation sperm ageing may not occur in Atlantic salmon. One reason may be the short life span of salmon sperm after ejaculation. Whether our findings are true in other external fertilizers with extended sperm activity remains to be tested.

    Abstract Abstract Limited mitochondria outer membrane permeability (MOMP) is a novel biological process where mammalian cells initiate the intrinsic apoptosis pathway with increased mitochondrial permeability but survive. One of the major consequences of limited MOMP is apoptotic endonuclease-induced DNA double strand breaks. Recent studies indicate that these DNA double stand breaks and ensuing activation of DNA damage response factors such as ATM play important but previously underappreciated roles in carcinogenesis and tumor growth. Furthermore, novel non-canonical roles of DNA repair factors such as ATM in tumor growth and treatment are also emerging. In this review, we try to summarize recent findings on this newly revealed link between DNA double strand break repair and cell death pathways.

    Abstract Abstract Background Splenectomy may lead to severe postoperative complications, including sepsis and cancers. A possible solution to this problem is heterotopic autotransplantation of the spleen. Splenic autografts rapidly restore the regular splenic microanatomy in model animals. However, the functional competence of such regenerated autografts in terms of lympho- and hematopoietic capacity remains uncertain. Therefore, this study aimed to monitor the dynamics of B and T lymphocyte populations, the monocyte-macrophage system, and megakaryocytopoiesis in murine splenic autografts. Methods The model of subcutaneous splenic engraftment was implemented in C57Bl male mice. Cell sources of functional recovery were studied using heterotopic transplantations from B10-GFP donors to C57Bl recipients. The cellular composition dynamics were studied by immunohistochemistry and flow cytometry. Expression of regulatory genes at mRNA and protein levels was assessed by real-time PCR and Western blot, respectively. Results Characteristic splenic architecture is restored within 30 days post-transplantation, consistent with other studies. The monocyte-macrophage system, megakaryocytes, and B lymphocytes show the highest rates, whereas the functional recovery of T cells takes longer. Cross-strain splenic engraftments using B10-GFP donors indicate the recipient-derived cell sources of the recovery. Transplantations of scaffolds populated with splenic stromal cells or without them afforded no restoration of the characteristic splenic architecture. Conclusions Allogeneic subcutaneous transplantation of splenic fragments in a mouse model leads to their structural recovery within 30 days, with full reconstitution of the monocyte-macrophage, megakaryocyte and B lymphocyte populations. The circulating hematopoietic cells provide the likely source for the cell composition recovery.

    Abstract Ethylhexyl methoxy cinnamate (EHMC) is one of the major organic ultraviolet (UV) filter pollutants in the environment. The purpose of this study was to investigate the parental transfer of EHMC and induced biochemical responses in zebrafish (Danio rerio). Zebrafish embryos were exposed to EHMC solution (1, 10, and 100 μg/L) for 4 months until sexual maturation. Then male and female parents were paired to lay eggs. F1 generations were divided into 2 categories: with and without continued EHMC exposure. EHMC was detected in both F0 parents and F1 eggs, indicating that EHMC can accumulate in zebrafish and transfer to offspring through reproduction. The hatching rate decreased and malformation rate increased significantly among parents and progeny embryos in the high concentration exposure group. For 40 dpf (days post-fertilisation) F0 generations, estradiol hormone and vitellogenin (Vtg) contents, the expression levels of Vtg1, P450 aromatase (Cyp19a and Cyp19b), 17β-hydroxysteroid dehydrogenase (Hsd17b1, Hsd17b3), estrogen receptor-alpha and progesterone receptor in all concentration groups decreased significantly, while androgen receptor increased significantly in 10 and 100 μg/L exposure groups compared with the corresponding control group, showing anti-estrogen and androgen effects. For 120 dpf F0 generations, acetylcholinesterase activity was significantly decreased and glutathione and malondialdehyde levels, superoxide dismutase, catalase and glutathione reductase activities were significantly increased in all treatment groups compared with the corresponding control group. In addition, F1 offspring with or without continued exposure to EHMC suffered similar or stronger oxidative stress compared with their parents. DNA breakage and apoptosis also occurred in 120 dpf parental liver cells in all treatment groups as a result of oxidative damage. Results suggested that EHMC have transfer effects between parents and offspring, which may cause negative effects on growth and development of zebrafish and induce biochemical responses in both parents and offspring.
