Abstract Fibroblast growth factor 21 (FGF21), a member of the FGF subfamily, is produced primarily in the liver and adipose tissue. The main function of FGF21 is to regulate energy metabolism of carbohydrates and lipids in the body through endocrine and other means, making FGF21 have potential clinical value in the treatment of metabolic disorders. Although FGF21 and its receptors play a role in the regulation of bone homeostasis through a variety of signaling pathways, a large number of studies have reported that the abuse of FGF21 and its analogues and the abnormal expression of FGF21 in vivo may be associated with bone abnormalities. Due to limited research information on the effect of FGF21 on bone metabolism regulation, the role of FGF21 in the process of bone homeostasis regulation and the mechanism of its occurrence and development have not been fully clarified. Certainly, the various roles played by FGF21 in the regulation of bone homeostasis deserve increasing attention. In this review, we summarize the basic physiological knowledge of FGF21 and the effects of FGF21 on metabolic homeostasis of the skeletal system in animal and human studies. The information provided in this review may prove beneficial for the intervention of bone diseases.

Abstract A key challenge confronting any sizeable country is how to build a unified nation while avoiding the trap of relying on a centralized government and an ossified bureaucracy that fail to address multi-faceted governance issues at varying levels and scales. Using Alexis de Tocqueville's concept of “centralized government, decentralized administration” as the starting point, this paper examines continuing institutional challenges in contemporary China, focusing on urban governance issues related to government accountability, fitting institutions to types of problems, and encouraging cross-sectoral collaboration. China's patriarchal and authoritarian legacies have shaped the nature of these challenges. To address these challenges, researchers must recognize how these legacies shape contemporary practices and the limits for reform.

Abstract In order to ensure the quality of resource recommendation and solve the problems of low recommendation accuracy, long recommendation time, and high data loss rate in the process of resource recommendation in traditional methods, a personalized recommendation system of English teaching resources based on the multi-K nearest neighbor regression algorithm is designed. According to the overall architecture of the personalized recommendation system of teaching resources, this study designs the resource browsing function module, teaching resource detailed page recommendation module, and teaching resource database. Based on the basic idea of the multi-K nearest neighbor regression algorithm, in order to avoid the loss of important data in English teaching resource recommendation and reduce the data loss rate, a missing data reconstruction algorithm of English teaching resources is proposed. Finally, the path interest of student users is considered from the selection of browsing path and access time to realize the personalized recommendation of English teaching resources. The experimental results show that the system has high resource recommendation accuracy, short recommendation time, and low data loss rate.

Abstract Ear photosynthesis plays a key role in wheat photosynthesis during the grain filling stage, particularly under drought stress. Thus, dissecting the responsibilities of the glume and awn in photosynthetic carbon fixation and assimilates transportation during the grain filling stage in spikes is imperative. In this study, the detachment of the glume (DG) and awn (DA) of a wheat variety (Pubing143) was used to estimate their influences on ear photosynthesis and dry matter distribution. Radioactive carbon-14 (14C) isotope was detected by a multifunctional liquid scintillation counting system. The accumulation of 14C assimilates and their contributions to grain weight were then calculated. Under well-watered conditions, ear photosynthesis was reduced by 16.8 % and 46.2 % 25 d after anthesis (DAA) in the de-glumed control (DGC) and de-awned control (DAC) treatments, respectively, compared with the intact ear control (IEC). Under drought stress, ear photosynthesis was reduced by 46 % and 74.2 % at 25 DAA after removing the glume and awn, respectively. Under normal conditions, the number of 14C assimilates of DGC, and DAC was reduced by 14.6 % and 20.9 % in grains at 25 DAA, respectively, compared with the IEC. Compared with IED, the 14C assimilates of DGD, and DAD declined by 17.2 % and 27 %, respectively, in grains at 25 DAA under drought conditions. Under well-watered conditions, the grain weight per pot was reduced by 11.2 % and 25.4 % in the de-glumed control (DGC) and de-awned control (DAC) treatments, respectively, compared with the intact ear control (IEC). The grain weight per pot was further reduced after removing the glume and awn (16 % and 32.2 %, respectively) under drought stress. Furthermore, the awn contribution to grain weight was twice that of the glume. Our results suggest that the glume and awn of ears play prominent roles during grain filling in wheat, especially under drought stress, and that the awn is more crucial than the glume.

Abstract The implementation of measures such as check dams and terraces in the Loess Plateau of China has had a groundbreaking impact on water and sediment conditions. The question of how to accurately simulate the runoff–sediment process under complex underlying surface conditions has become key to clarifying the water cycle law. This study focused on the Chenggou River basin, a small watershed located in the Loess Plateau, to examine the effect of the underlying surface characteristics on the runoff production process, and the spatial distribution of the dominant runoff process in the runoff generation mechanism was determined according to the land application, slope and vegetation coverage of the watershed. A runoff–sediment model was constructed that was combined with the traditional hydrological physical mechanism and a deep learning algorithm. Different parameters were calibrated depending on the spatial distribution of the dominant runoff process and we then ran the runoff–sediment simulation model to very its serviceability in the typical watershed of the Loess Plateau. Different parameters were calibrated for each type of hydrological response unit (HRU), according to the division of each HRU and the actual flood process, to calculate the runoff yield of each HRU. An LSTM algorithm was used for flow routing and a CSLE algorithm was used to simulate soil erosion. The results show that there were 29 flood events in the Chenggou River basin from 2013 to 2017. The average runoff depth had an 8.86% margin of error, while the peak flow had a slightly higher 9.44% deviation. The Nash efficiency coefficient was 0.84, and the relative error of soil erosion was 14.45%. The model simulation effect is good and can be applied to the typical watershed of the Loess Plateau. The model can provide a scientific basis for the highly efficient and sustainable utilization of water resources, ecological environment construction and the sustainable development of agriculture.

Abstract IntroductionAlthough there is growing evidence supporting the effectiveness of Janus kinase (JAK) inhibitors in treating alopecia areata, the high rate of recurrence following drug discontinuation has led to prolonged treatment courses and raised concerns about long-term safety. In clinical practice, caution should be exercised while using JAK inhibitors for various indications, and a comprehensive pre-treatment screening.MethodsThis study presents an analysis of screening data collected from real-world settings before the initiation of Janus kinase inhibitors in patients with alopecia areata. Investigators collected retrospective medical data characterizing patients’ screening data. Data on demographic and clinical data, including age, sex, disease duration, severity of alopecia tool scale, history of prior treatment, and treatment regimen were recorded.ResultsIn this cohort (N = 218), JAK inhibitors were initiated for 163 of 218 (74.8%) alopecia areata patients. The numbers of patients positive for antinuclear antibodies, hepatitis B surface antigen, hepatitis C virus antibodies, human immunodeficiency virus antibody, treponema pallidum hemagglutination assay, and thyroid-stimulating hormone were 32 (32/176), 10(10/218), 0 (0/218), 0 (0/218), 3 (3/218) and 9 (9/176), respectively. The number of patients with T-cell spot positive or imaging of the chest indicating tuberculosis was 37 (37/218).DisccusionOur data provide additional information on the safety profile of JAK inhibitors in patients with alopecia areata. As such, it is necessary and crucial to screen for JAK inhibitors before it is used, particularly for individuals with a high risk of tuberculosis, hepatitis B, and other infections.

Abstract To enhance the deflagration efficiency and protection level of combustible mixed gases in narrow spaces, a small square experimental pipeline system was designed. Experiments were conducted to investigate the effects of lateral vent pipes and metal wire meshes on the explosion characteristics of methane gas. This study examined the influence of changing the positions of the lateral vent pipes and metal wire meshes in the pipeline on the variation of parameters such as the flame shape, leading speed, and pressure of the methane/air premixed gas in the pipeline. The results indicated that the lateral vent pipes could effectively release part of the energy from the methane explosion, and the release effect was stronger the closer they were to the ignition end. This was significantly more effective in releasing the flame and pressure than when the vent pipes were placed in the middle or at the end of the pipeline. For lateral vent pipes close to the ignition source, their effective release of the not yet fully developed premixed flame allowed the heat absorption, wave absorption, and quenching performance of the installed metal wire mesh in the pipeline to fully exert their effects on the slow-spreading premixed flame. Furthermore, when a metal mesh was installed in the pipeline and the flame could not be extinguished, the flame penetrated the mesh structure, causing the flame front to become unstable and exhibit “irregular wrinkles”. That is, the flame front was no longer smooth, the wrinkles became more pronounced, and the degree of turbulence was enhanced.

Abstract Lung adenocarcinoma (LUAD) is the most prevalent histologic type of lung cancer, associated with a high incidence rate and substantial mortality rate worldwide. Accumulating evidence shows that the aberrant expression of neuromedin U (NMU) contributes to the initiation and progression of cancer. Herein, we explored whether NMU could be adopted as a new diagnostic and therapeutic marker in LUAD. The UALCAN and GEPIA web resources were employed to assess data on the NMU expression in LUAD. The STRING web resource was used to develop the PPI (protein-protein interaction) network of NMU, whereas Cytoscape was applied for module analysis. The Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses of NMU and the interacting proteins were examined using the WebGestalt tool. Survival analysis was performed with the Kaplan-Meier plotter tool. Results revealed that the NMU expression in LUAD was significantly higher than in the nonmalignant tissues. Moreover, higher NMU levels were dramatically related to shorter overall survival, first progression survival, and postprogression survival. The specific gene mutations G45V, R143T, and F152L of NMU occurred in LUAD samples and were associated with a worse prognosis in patients. KEGG and western blot analyses demonstrated an association of NMU with the cell cycle and the cAMP signaling cascade. Bioinformatic analysis and the in vitro experiments implicated NMU as a promising prognostic signature and treatment target for LUAD.

Abstract In order to explore the accuracy of computed tomography (CT) and type-B ultrasonography in the examinations infection of ovarian cyst, the computed tomography and type-B ultrasonography were used for the examinations of 50 patients with infection of ovarian cysts, whose results were analyzed to determine the accuracy. Results have shown that the misdiagnosis rates of CT for patients with infection of ovarian cyst reached 18%, in which the misdiagnosis rate of small lesion 2.1 × 1.5 × 1.8 cm was relatively higher; in addition, the misdiagnosis rates of type-B ultrasonography reached 26%, in which the misdiagnosis rate of large lesion 2.6 × 1.6 × 1.5 cm was relatively higher; however, the misdiagnosis rates of the joint examination of these methods were only 2%. In summary, the deficiencies of CT and type-B ultrasonography may lead to misdiagnosis; therefore, in the clinical practices, the examinations of patients with infection of ovarian cyst should combine CT and type-B ultrasonography together to avoid the misdiagnosis and obtain the most accurate results as much as possible, which is important to both the patients and the development of medical examinations.

Abstract Post-transplant anemia is one of the most common complications in kidney transplant recipients, severely affecting patient prognosis and quality of life, and is an independent predictor of graft kidney loss and patient mortality. However, our clinical understanding and the attention given to post-transplant anemia are currently insufficient. This paper reviews the current status, risk factors, and therapeutic progress in anemia after transplantation in kidney transplant recipients. We recommend that clinical staff pay attention to anemia and its complications in kidney transplant recipients and intervene early for anemia.
