    Abstract Resveratrol plays a crucial phytoalexin role in the grapevine and is beneficial to human health. However, the molecular mechanism of resveratrol accumulation in the enhancement of disease resistance is unclear. Here, we report that the transcription factor VqMYB154 from Vitis quinquangularis accession Danfeng-2 is strongly expressed under artificial inoculation with Uncinula necator and regulates resveratrol accumulation. Unlike its homolog, VqMYB154 has a pathogen-induced promoter and responds to stimulation by U. necator, Pseudomonas syringae, and other treatments. Yeast one-hybrid and GUS activity assays confirmed that VqMYB154 can activate the stilbene synthase genes VqSTS9, VqSTS32, and VqSTS42 by directly binding to their promoters. Overexpression of VqMYB154 in grape leaves resulted in activation of the stilbene pathway, upregulation of STS genes, and accumulation of stilbenoids. In addition, heterologous overexpression of VqMYB154 in Arabidopsis activated resistance-related genes and resulted in greater programmed cell death and accumulation of reactive oxygen species, which led to resistance against P. syringae. These results suggest that the transcription factor VqMYB154 from V. quinquangularis accession Danfeng-2 participates in the regulatory mechanism that improves the biosynthesis and accumulation of stilbenes and enhances resistance to disease in grapevine.

    Abstract Objective: This study aimed to clone the whole open reading frame (ORF) of VviSEP2 from *Vitis vinifera* and verify the relationship between VviSEP2 and agronomic traits of stenospermocarpic (seedless) grapevines. Method: The full-length ORF sequence of VviSEP2 was isolated from *V. vinifera* cv. ‘Thompson Seedless’ and analyzed with bioinformatics methods. Semi-quantitative real-time PCR (sqRT-PCR) and quantitative real-time PCR (qRT-PCR) were also used to detect the expression pattern of VviSEP2. Result: A gene related to ovule development of stenospermocarpic grapevine was obtained. The cDNA sequence length of this gene was 1 132 bp and the ORF was 741 bp, encoding 246 amino acids. It was confirmed that the gene was a member of the Class E MADS-box family by amino acid sequence alignment and phylogenetic tree analysis, and it was designated as VviSEP2.The VviSEP2 expressed in flower bud, flower and ovule, but not in root, stem and leaf. The expression levels of this gene in flower and flower bud of ‘Pinot Noir’ were significantly higher than that of ‘Thompson Seedless’. The relative expression levels of the VviSEP2 gene in ‘Pinot Noir’ in each development period were 3-5 times of those in ‘Thompson Seedless’. Conclusion: The VviSEP2 gene was associated with stenospermocarpy.

    Abstract Vacuolar processing enzyme (γVPE) was cloned from Chinese wild *Vitis pseudoreticulata* accession. ‘Baihe-35-1’ and *Vitis vinifera* cv. ‘Carinena’ by RT-PCR. They were named *VpγVPE* and *VvCγVPE*. The full length of *VpγVPE* and *VvCγVPE* cDNA are 1624 bp, and the open reading frame are 1482 bp, encoding 493 amino acid. The alignment of the amino acid sequence revealed that Ser 395, one key amino acid in substrate pocket, was replaced by Ala in *Vitis pseudoreticulata* accession. ‘Baihe-35-1’, *Vitis vinifera* cv. ‘Carinena’, *Vitis vinifera* cv. ‘Thompson seedless’, and *Vitis vinifera* cv. ‘Pinot Noir’. The different expression profile of *VpγVPE* gene and *VvCγVPE* gene in different periods after powdery mildew induced were analyzed by Real-time fluorescent quantitative PCR. After induction the expression of *VpγVPE* gene increase slightly in 4 h, 48 h, 168 h and the *VvCγVPE* gene shows predominant expression in 4 h after that it decreases slowly. The study shows that *γVPE* is related to resistant to powdery mildew. The study provides reference value to the molecular mechanism of *γVPE* gene in disease resistance.
