    Abstract To investigate the effects of crude β-glucosidases from Issatchenkia terricola SLY-4 (SLY-4E), Pichia kudriavzevii F2-24 (F2-24E), and Metschnikowia pulcherrima HX-13 (HX-13E) on flavor complexity and characteristics of wines, grape juice was fermented by Saccharomyces cerevisiae with the addition of SLY-4E, F2-24E and HX-13E, respectively. The growth and sugar consumption kinetics of S. cerevisiae, the physicochemical characteristics, the volatile compounds, and the sensory dimensions of wines were analyzed. Results showed that adding SLY-4E, F2-24E, and HX-13E into must had no negative effect on the fermentation and physicochemical characteristics of wines, but increased the content of terpenes, esters, and fatty acids, while decreased the C6 compound content. Each wine had its typical volatile compound profiles. Adding SLY-4E or F2-24E into must could significantly improve the flavor complexity and characteristics of wines. These results would provide not only an approach to improve flavor complexity and characteristics of wines, but also references for application of β-glucosidases from other sources.

    Abstract ’Xueyuan Hong’ is a new seeded table grape cultivar bred through crossing 'Rousalka’ (female parent) and ’Adominique’ (male parent), both of which are of vinifera genetic background. The full expanded leaf of the cultivar is very large. The fruit cluster shape is conical, medium in size and high compactness. The berry is very large, with an average weight of 12.9 g, elliptical in shape. The berry skin is red, thin and with astringency in taste. Two seeds per berry on average, and neutral in flavor. The cultivar has good potential for delayed harvest to extend the harvest season and it has a high storability. In Beijing area, ’Xueyuan Hong’ bud breaks in late March, flowers in mid-May, reaches verasion in mid-to-late July, and ripens in mid-August.

    Abstract In insects, metamorphosis and reproduction are controlled by juvenile hormone (JH) and 20-hydroxyecdysone (20E). Krüppel homolog 1 (Kr-h1), a transcription factor, is regarded as a JH-early inducible gene responsible for the repression of metamorphosis. However, the role of Kr-h1 in reproduction of holometabolic insects is relatively less understood. In this study, we studied the role of Kr-h1 in larvae-pupae transition and female reproduction in the major agricultural pest Helicoverpa armigera. Two HaKr-h1 isoforms (HaKr-h1α and HaKr-h1ß) were identified, with HaKr-h1α predominant in the cotton bollworm. In larvae, HaKr-h1 was predominately expressed in the epidermis and markedly up-regulated during the moult stage, whereas in adults HaKr-h1 was mainly expressed in females and the highest transcription was detected in the ovaries. Considering the function of hormones in larval metamorphosis, we examined the modulation of gene expression in response to hormones, which showed that HaKr-h1 was significantly induced by both JH analogue (JHA) and 20E. Knockdown of HaKr-h1 in fifth-instar larvae resulted in precocious metamorphosis from larvae to pupae. Moreover, a fluorescence immunoassay coupled with heterologous expression revealed that HaKr-h1 was localized in the nucleus of oocyte membrane. In female adults, depletion of HaKr-h1 severely repressed the transcription of vitellogenin, disrupted oocyte maturation and reduced the number of eggs laid, suggesting that HaKr-h1 is required for vitellogenesis and egg production in H. armigera. The present study provides insight into the roles of HaKr-h1 in JH-mediated reproduction and highlights HaKr-h1 as a target for suppression of lepidopteran pests.

    Abstract ‘Scholar Red’ is a new seeded table grape cultivar bred through crossing between ‘Rousalka’ and ‘Adominique’, both are of *vinifera* genetic background. Fully developed seeds were collected from ripe berries from artificial hybridization pollination in 2005, the seeds were sowed in the spring of2006 after stratification and then the plantlets were moved from seed trays to soil in a greenhouse. In 2010, all plants had fructified. After fruit quality test using berry size and sugar content as the major criteria over five years from 2011 to 2015, ‘Scholar Red’ was finally selected. The variety is of middle size in cluster with medium compact appearance, the average cluster weight is 500 g at harvest. The berries are oval shape, with 1-2 seeds per berry, weighing averagely 8.9 g, of attractive blue-purple color, it is a variety easy to get skin color, the skin is thin and closely attached to the flesh, not easy to peeling. The flesh is crunchy, juicy and delicate in texture, and no anthocyanin accumulation in the flesh. Berries are of neutral flavor at ripening, the soluble solid content of grapes at harvest grown in the high tunnel is 20.1%, and the overall consumption quality is high. In growth habit, ‘Scholar Red’ is a vigorous variety, the vine bud breaks in late March, almost all the new shoots from winter buds bear 1-2 flower clusters, flowers are observed as imperfect flower type, other variety for pollination is required. The flowers bloom in the mid May, the same period as the major table grape varieties, and the berries ripen around middle August in Beijing area. The berries have long storage-life, and there is no problem of falling berries during transportation. The grape can also be kept on the vine for delayed harvest. The variety is suitable for outdoor cultivation in areas where the rainfall is not more than 600 mm in summer, and protected cultivation is required in areas with higher rainfall. ‘Scholar Red’ is of regular cold resistance as other *vinifera* cultivars, during the winter burying the vine or using other protected methods are suggested for areas that other *vinifera* varieties are required.

    Abstract Objective: This study explored the drought resistance of wild *Vitis* seedlings in Karst region to provide reference for the utilization of wild grape germplasm resources and the mining of drought resistant grape genes. Method: Seven individuals of four wild *Vitis* verities originated in Guizhou including ‘Huaxi-9’, ‘Huaxi-4’, ‘Nongyuan-11’, ‘Fuquan-1’ (*Vitis quinquangularis* Rehd.), ‘Maolan-11’ (*Vitis adenoclada* Hand-Mazz), ‘Xishui-4’ (*Vitis flexuosa* Thunb.) and ‘Nongyuan-1’ (*Vitis davidii* Foex) were selected and ‘Red globe’ (*V. vinifera* L.) and ‘Crystal’ (Hybrid of *V. vinifera* × *V. labrusca*) were used as control. Five stress treatments of 8, 16, 24, 32 and 40 d were set up for pot experiments to study morphology, and morphological and physiological characteristics related to water stress. Then, the drought resistance was evaluated by comprehensive membership function and principal component analyses. Result: Drought stress caused losing of green color and folding of leaves, significantly increased the symptoms of drought and electrical conductivity (REC), and significantly decreased the leaf water potential and relative water content (RWC). At 8 d without watering, all grapes showed no drought symptoms, while two materials (‘Huaxi-9’ and ‘Xishui-4’) showed grade drought damage symptoms and the rest showed grade 3 drought damage symptoms 40 d after stress. With the extension of stress time, the relative water content of leaves decreased gradually. ‘Xishui-4’ and ‘Huaxi-9’ decreased the least by 9.82% and 9.63% compared to 8 d after stress, while ‘Maolan-11’ and ‘Red globe’ decreased the most by 13.60% and 12.94%. With the extension of stress time, the leaf water potential gradually decreased. Compared with 8 d drought stress, the leaf water potentials of ‘Maolan-11’ and ‘Red globe’ declined the most by 2.10 and 2.05 MPa, while those of ‘Nongyuan-11’ and ‘Huaxi-9’ decreased the smallest by 1.18 and 1.19 MPa, respectively. Leaf conductivity of grape (REC) was 13.19%-21.75% under 8 d drought stress, and it increased continuously by 38.28%-42.09% under 40 d drought stress. ‘Huaxi-9’ had the lowest REC of 51.80%, while ‘Red globe’ had the largest of 62.73%, with the difference of 10.93%. Comprehensive evaluation showed that the drought resistance was in the order of ‘Huaxi-9’ > ‘Xishui-4’ > ‘Nongyuan-11’ > ‘Fuquan-1’ > ‘Nongyuan-1’ > ‘Crystal’ > ‘Huaxi-4’ > ‘Maolan-11’ > ‘Red globe’. Conclusion: *V. quinquangularis* ‘Huaxi-9’ had the strongest drought resistance, followed by *V. flexuosa* ‘Xishui-4’and *V. quinquangularis* ‘Fuquan-1’ and ‘Nongyuan-11’. They all can be used as drought grape breeding germplasm resources.

    Abstract In order to solve the problems of traditional aqueous enzymatic extraction of grape seed oil, such as complex pretreatment, large amounts of enzyme and water consumption and long extraction time, grape seed oil was extracted by aqueous ethanol method. The results showed that grape seed could be directly used to extract oil after pulverization without pretreatment. By studying the factors affecting the extraction of grape seed oil by aqueous ethanol, the optimal extraction conditions were determined as follows: volume fraction of ethanol 45%, solid-liquid ratio 1∶3 ,extraction temperature 60 °C, pH 9, extraction time 1 h, and residue was further extracted by aqueous phase under the same conditions. Under the optimal conditions, the extraction rate of the free oil reached 86. 57%. The obtained grape seed oil was simply refined, and the indexes reached the national standard of first-grade grape seed oil. The contents of γ-tocopherols, sterols and total phenols reached 52.70, 2120.23, 195.5 mg/kg, respectively, and the trans fatty acid content was 0. 763 g/kg. Compared with traditional aqueous enzymatic extraction, the raw materials in aqueous ethanol method need no pretreatment nor enzyme, and the extraction time shortened from above 3 h to 2 h, and the water consumption decreased from 6 t/t (based on the mass of grape seed) to 3 t/t (based on the mass of grape seed). The water phase could be recycled and the quality of the oil was high, which provided a new idea for the efficient extraction of grape seed oil.
