    Abstract Monoclonal antibodies (mAbs) and their derivatives have emerged as one of the most important classes of biotherapeutics in recent decades. The success of mAb is due to their high versatility, high target specificity, excellent clinical safety profile, and efficacy. Antibody discovery, the most upstream stage of the antibody development pipeline, plays a pivotal role in determination of the clinical outcome of an mAb product. Phage display technology, originally developed for peptide directed evolution, has been extensively applied to discovery of fully human antibodies due to its unprecedented advantages. The value of phage display technology has been proven by a number of approved mAbs, including several top-selling mAb drugs, derived from the technology. Since antibody phage display was first established over 30 years ago, phage display platforms have been developed to generate mAbs targeting difficult-to-target antigens and tackle the drawbacks present in in vivo antibody discovery approaches. More recently, the new generation of phage display libraries have been optimized for discovery of mAbs with "drug-like" properties. This review will summarize the principles of antibody phage display and design of three generations of antibody phage display libraries.

    Abstract In view of the problems existing in the current exam system, such as separation from the command information system and difficulty in carrying out remote group training, a scalable and extensible command training exam system architecture is proposed to provide technical support for integrating and invoking the relevant components of the command information system and carrying out large-scale group training. Firstly, the component integration framework of multi-system integration and the integrated exam course evaluation mode are proposed to realize the assessment of each course including command skills. Then, a multi-level server exam structure is designed. On this basis, through the application of virtualization cluster and other technologies, the rapid deployment of the system under diverse conditions and the large-scale remote group training application is realized.

    Abstract Image-text retrieval is a widely studied topic in the field of computer vision due to the exponential growth of multimedia data, whose core concept is to measure the similarity between images and text. However, most existing retrieval methods heavily rely on cross-attention mechanisms for cross-modal fine-grained alignment, which takes into account excessive irrelevant regions and treats prominent and non-significant words equally, thereby limiting retrieval accuracy. This paper aims to investigate an alignment approach that reduces the involvement of non-significant fragments in images and text while enhancing the alignment of prominent segments. For this purpose, we introduce the Cross-Modal Prominent Fragments Enhancement Aligning Network(CPFEAN), which achieves improved retrieval accuracy by diminishing the participation of irrelevant regions during alignment and relatively increasing the alignment similarity of prominent words. Additionally, we incorporate prior textual information into image regions to reduce misalignment occurrences. In practice, we first design a novel intra-modal fragments relationship reasoning method, and subsequently employ our proposed alignment mechanism to compute the similarity between images and text. Extensive quantitative comparative experiments on MS-COCO and Flickr30K datasets demonstrate that our approach outperforms state-of-the-art methods by about 5% to 10% in the rSum metric.

    Abstract Spectroscopic nanoscopy (SN) has been recognized as a key functional imaging tool in cell biology and chemistry because it offers the unique capability to simultaneously obtain the spatial and spectral information for single molecules. However, it has an intrinsic issue in using the limited photon budget from single emitters divided into two imaging channels to concurrently acquire spatial and spectral images. Accordingly, this issue lowers the spatial localization and spectral precision. Although several techniques have been introduced to improve the spatial precision in SN, improving the spectral precision has been overlooked so far. Here we propose a method to improve the spectral precision by optically manipulating the width of the spectroscopic signatures using a demagnifier. We evaluate its performance using numerical simulations with systematic investigations of several underlying optimal parameters such as the demagnification factor and the integration width in the proposed configuration. We also present achievable spectral precision values with different signal and background levels. Compared to the existing SN system, the 3× demagnifier-based configuration shows an approximate 35% improvement, from 2.9 nm to 1.9 nm, in the spectral precision at the 1000 photons signal level.

    Abstract Background: To investigate the involvement of LINC02605 in the progression of paediatric Mycoplasma pneumoniae pneumonia (MPP).
    Methods: One hundred and thirty-two children with MPP (90 simple MPP and 42 MPP + diarrhoea) were enrolled, and their plasma was collected for detection of LINC026505 expression. CCK-8 kit and commercial apoptosis kit were introduced to determine cell growth and apoptosis. In silico prediction analyses were conducted to predict the downstream miRNA for LINC02605, following verification by dual luciferase reporter assay. The lipid-associated membrane proteins (LAMPs) were used to treat A549 and Coca-2 cells.
    Results: LIN02605 was highly expressed in the MPP, especially in MPP complicated with diarrhoea. LINC02605 downregulation in A549 cells correlated with significant suppression of cell apoptosis rate and growth inhibition rate in vitro. Introduction of miR-539-5p inhibited luciferase activity in a reporter system containing the wild-type LINC02605 and CXCL1. After stimulation with LAMPs, overexpression of LINC02605 and CXCL1 and inhibition of miR-539-5p were found. miR-539-5p and CXCL1 knockdown resulted in a rescue effect on the LINC02605-inhibited cell apoptosis. LAMPs induced IL-1β in intestinal epithelial cells and IL-1β induced LINC02605 expression in A549 cells.
    Conclusions: LINC02605 was upregulated in MPP and miR-539-5p was a target for LINC02605. LINC02605 may be involved in the crosstalk between the gastrointestinal tract and the respiratory tract.

    Abstract The rapid development of robotics has benefited by more and more people putting their attention to it. With the demand for robots is growing for the purpose of fulfilling tasks instead of humans, how to control the robot better is becoming a hot topic. For obstacle avoidance, we proposed algorithms for both 2D planar environments and 3D space environments. The example cases we raise are those that need to be addressed but have always been ignored. In addition, we put efforts into trajectory planning for robots. The two scenarios we set are self-driving cars on the road and reconnaissance and surveillance of drones. For future expectations, there are some possible directions. How to combine traditional navigation algorithms and high-tech algorithms together so as to fulfill the tasks perfectly while the computational efficiency is not too high is a worthy topic. In addition, extending the obstacle avoidance algorithms to more competitive situations. Moreover, cooperation among multi robots are worth attention by researchers. All in all, there is still a long way to go for the development of navigation and control of mobile robots. Despite this, we believe we do not need to wait for too long time to see the revolution of robots.
