    Abstract As a tumor suppressor protein, p53 is directly involved in the control of cell cycle arrest, which plays a crucial role in cell metabolism, growth, apoptosis and responses to different types of damage or stress processes. And since the tumor and cancer are usually related to the mutation or functional inactivation of the p53, it’s necessary to investigate the mechanism of p53 functioning the tumor and other disease, trying to figure out its drug design strategy. In this paper, a brief introduction of p53 is given, including the structure, function and properties. Subsequently, the relationship of p53 gene and cancer is also discussed. In the end, a new rational drug-design for disordered region of p53 was summarized

    Abstract Intratumoral hypoxia is widely associated with the development of malignancy, treatment resistance, and worse prognoses. This study aims to investigate the role of hypoxia-related genes (HRG) in the immune landscape, treatment response, and prognosis of head and neck squamous cell carcinoma (HNSCC). The transcriptome and clinical data of HNSCC were downloaded from TCGA and GEO databases, and HNSCC molecular subtypes were identified using non-negative matrix factorization (NMF) clustering. Prognostic models were constructed using univariate, Lasso, and multivariate Cox regression analyses. The relationship between HRGs and immune cell infiltration, immune therapy response, and drug sensitivity was evaluated, and a nomogram was constructed. 47 HRGs were differentially expressed in HNSCC, among which 10 genes were significantly associated with HNSCC prognosis. Based on these 10 genes, 2 HNSCC molecular subtypes were identified, which showed significant heterogeneity in terms of prognosis, immune infiltration, and treatment response. A total of 3280 differentially expressed genes were identified between the subtypes. After univariate, Lasso, and multivariate Cox regression analysis, 18 genes were selected to construct a novel prognostic model, which showed a significant correlation with B cells, T cells, and macrophages. Using this model, HNSCC was classified into high-risk and low-risk groups, which exhibited significant differences in terms of prognosis, immune cell infiltration, immune therapy response, and drug sensitivity. Finally, a nomogram based on this model and radiotherapy was constructed, which showed good performance in predicting HNSCC prognosis and guiding personalized treatment strategies. The decision curve analysis demonstrated its better clinical applicability compared to other strategies. HRGs can identify 2 HNSCC molecular subtypes with significant heterogeneity, and the HRG-derived risk model has the potential for prognostic prediction and guiding personalized treatment strategies.

    Abstract Telomere dysfunction has been identified as a biological marker of cancer progression in several types of cancer, including Head and Neck Squamous Cell Carcinoma (HNSCC). This study aimed to characterize the telomere maintenance genes (TMG)-related signature in prognosis and treatment response in HNSCC. The transcriptome and clinical data of HNSCC were obtained from the Cancer Genome Atlas (TCGA) and Gene Expression Omnibus databases, respectively. Non-negative matrix factorization (NMF) was used to identify molecular subtypes derived from TMG. Gene set enrichment analysis (GSEA) was performed to analyze the differentially expressed pathways between subtypes, and a risk score model derived from TMG was established. Kaplan-Meier survival analysis was used to evaluate inter-group prognostic features, and the correlation between TMG-derived molecular subtypes and risk score model with immune infiltration, immunotherapy, and chemosensitivity was assessed. Two HNSCC subtypes were identified based on 59 TMG-related genes, which exhibit significant heterogeneity in prognosis, immune cell infiltration, and treatment response. Additionally, a TMG-derived risk signature containing 9 genes was developed to assess the prognosis of HNSCC patients. The signature had significant predictive ability for HNSCC prognosis and was significantly correlated with immune cell infiltration and immunotherapy response. A nomogram integrating the risk signature, N stage and radiotherapy was constructed to predict 1-, 3-, and 5-year overall survival (OS) of HNSCC patients, which had better performance than other prognostic models and included TMG-derived risk score, radiotherapy, and N stage. This study identified TMG-derived molecular subtypes in HNSCC and developed a novel prognostic score model, highlighting the potential value of TMG in HNSCC prognosis and immunotherapy.

The Netherlands Organization for Applied Scientific Research (TNO) has been committed to developing a quantitative risk assessment methodology for the unintentional intake of allergens.

    Abstract Numerous studies have explored the correlation between non-symbolic number comparison and mathematical abilities in children, but the results have been inconsistent. The underlying mental processing featuring fluency may affect the correlation. The current study tested the fluency hypothesis that non-symbolic number comparison is associated with mathematical fluency in the development of mathematical ability. Non-symbolic number comparison, arithmetic computation, mathematical reasoning, non-symbolic number estimation, symbolic number comparison, and a series of basic cognitive processing tasks, including mental rotation, non-verbal matrix reasoning, and choice reaction time, were administered to 1072 first- to fourth-grade children. The results show that non-symbolic number comparison (measured via numerosity comparison) was the only independent predictor of arithmetic computation in higher grades, even after controlled for age, gender, basic cognitive processing, non-symbolic number estimation (measured via numerosity estimation), and symbolic number comparison (measured via digit comparison). However, it did not correlate with mathematical reasoning in any grade. These findings support the fluency hypothesis for developmental correlation between non-symbolic number comparison and mathematical abilities. That is, non-symbolic number comparison correlates with mathematical ability featuring fluency.

    Abstract Previous studies have found a correlation between numerosity processing and arithmetical performance. Visual perception has already been indicated as the shared cognitive mechanism between these two; however, these studies mostly focused on children. It is not clear whether the association between numerosity processing and arithmetical performance still existed following the development of individual arithmetical performance. Consequently, the underlying role of visual perception in numerosity processing and arithmetical performance has not been sufficiently studied in adults. For this study, researchers selected a total of 205 adult participants with an average age of 22years. The adults were administered arithmetic tests, numerosity comparison, and visual figure matching. Mental rotation, choice reaction time, and nonverbal intelligence were used as cognitive covariates. Results showed that numerosity comparison of adults correlated with their arithmetical performance, even after controlling for age and gender differences as well as general cognitive processing. However, after controlled for visual figure matching, the well-established association between numerosity comparison and arithmetic performance disappeared. These results supported the visual perception hypothesis, that visual perception measured by visual figure matching can account for the correlation between numerosity comparison and arithmetic performance. This indicated that even for adult populations, visual perceptual ability was the underlying component of numerosity processing and arithmetic performance.

    Abstract This study aims to investigate the status and influencing factors of two-way social support among old adults. A cross-sectional study of 408 convenient samples of old adults was conducted using socio-demographic questionnaire, Brief 2-Way Social Support Scale, Modified Barthel index, General Well-being Schedule, Family APGAR Index, and Lubben Social Network Scale 6. The two-way social support score for old adults in China was (43.74±7.86), with the receiving and giving social support scoring (22.80±4.06) and (20.94±4.52), respectively. The multiple linear regression analysis revealed that family care, residence place, socioeconomic status, and social network were associated with both receiving and giving social support. Chronic diseases and religious beliefs were related to receiving social support, while gender, general well-being, and residence form were related to giving social support. Tailored interventions based on the distinct influencing factors are needed to enhance old adults' social support both as recipients and providers.

    Abstract Although a great deal of research has shown a relationship between numerosity sense and mathematical ability, some studies have failed to do so. The main source of this inconsistency could be the varied ways of measuring mathematical abilities. The current investigation explored several types of mathematical ability, from basic number processing and arithmetic computation to numerical reasoning and arithmetic learning. We hypothesized that the correlation between numerosity sense and mathematical ability depends on mathematical fluency. A total of 415 college students (178 males and 237 females, mean age = 20.42 years, range = 18.58-22.92 years) were recruited to complete seven mathematical tasks and two numerosity tasks, as well as other tasks that measured cognitive covariates. The results showed that after controlling for age, gender, and related general cognitive factors, numerosity sense still predicted substantial variation in parity judgment, visual digit comparison, and computation, but it did not predict variation in numerosity estimation, auditory digit comparison, number series completion, or digit associate learning. The results suggest that numerosity sense correlates with mathematical abilities that accompany fluency.
