Abstract This study aims to investigate the role and mechanism of ubiquitin-specific protease 3 (USP3) in cisplatin (DDP) in non-small cell lung cancer (NSCLC). USP3 expression in NSCLC cells was detected using reverse transcription quantitative PCR and Western blot. DDP-resistant cells were constructed and cell counting kit-8 assay determined the IC50 of cells to DDP. USP3 expression was silenced in DDP-resistant cells, followed by detection of cell proliferation by clone formation assay, iron ion contents, ROS, MDA, and GSH levels by kits, GPX4 and ACSL4 protein expressions by Western blot. The binding between USP3 and ACOT7 was analyzed using Co-IP, and the ubiquitination level of ACOT7 was measured. USP3 and ACOT7 were highly expressed in NSCLC cells and further increased in drug-resistant cells. USP3 silencing reduced the IC50 of cells to DDP and diminished the number of cell clones. Moreover, USP3 silencing suppressed GSH and GPX4 levels, upregulated iron ion contents, ROS, MDA, and ACSL4 levels, and facilitated ferroptosis. Mechanistically, USP3 upregulated ACOT7 protein expression through deubiquitination. ACOT7 overexpression alleviated the promoting effect of USP7 silencing on ferroptosis in NSCLC cells and enhanced DDP resistance. To conclude, USP3 upregulated ACOT7 protein expression through deubiquitination, thereby repressing ferroptosis in NSCLC cells and enhancing DDP resistance.

Abstract Cutaneous squamous cell carcinoma is a common malignant tumor. The aim of the present study was to examine the biological function of microRNA (miR)-27b-3p in cutaneous squamous cell carcinoma (CSCC) and its underlying mechanism. The relative expression levels of miR-27b-3p were determined in A-431, Colo-16 and NHEK/SVTERT3-5 cell lines. The regulatory effects of miR-27b-3p on the proliferation of CSCC cells were evaluated using MTT and colony formation assays. Transwell assays were conducted to examine the role of miR-27b-5p in the migratory and invasive abilities of CSCC cells. The levels of EGFR, MMP-13, Akt, phosphorylated (p)-Akt, cyclin D1, N-cadherin (CAD) and E-CAD were detected in CSCC cells using reverse transcription-quantitative PCR and western blot analysis. Binding between miR-27b-3p and the 3'-untranslated region (UTR) of EGFR or MMP-13 was assessed using a dual-luciferase reporter assay. miR-27b-3p was significantly downregulated in CSCC cell lines, compared with the skin keratinocyte cell line. Transfection with a miR-27b-3p mimic significantly reduced the proliferative, migratory and invasive abilities of CSCC cells

Abstract The current study aimed to investigate the potential function and mechanism of microRNA (miR)-216a-3p in the osteogenic differentiation of human adipose-derived stem cells (hADSCs). Dynamic expression changes of miR-216a-3p in the osteogenic differentiation of hADSCs were examined by reverse transcription-quantitative PCR (RT-qPCR). Regulatory effects of miR-216a-3p on the relative levels of osteogenesis-associated genes were also detected by RT-qPCR and western blotting. The relationship between miR-216a-3p and Wnt3a was verified through a dual-luciferase reporter assay. Furthermore, the influence of miR-216a-3p on the Wnt/β-catenin signaling pathway during the osteogenic differentiation of hADSCs was investigated by western blotting. The results revealed that during the osteogenic differentiation process of hADSCs, miR-216a-3p was downregulated and Wnt3a was upregulated. It was further verified that Wnt3a was the target of miR-216a-3p. Through inactivation of the Wnt/β-catenin signaling pathway, miR-216a-3p was able to mediate osteogenic differentiation of hADSCs. In conclusion, by targeting Wnt3a, miR-216a-3p mediated the osteogenic differentiation of hADSCs, which negatively regulated the Wnt/β-catenin signaling pathway.

Abstract This article investigates channel phase mismatch calibration during the application of displaced-phase-center antenna (DPCA) in dual-channel sliding spotlight synthetic aperture radar (SAR) for ground moving target indication (GMTI). In sliding spotlight SAR, the utilization of beam progressive sweeping in azimuth causes antenna phase centers to be misaligned from the sensor path, resulting in the phase mismatch between channels. Then, spatial channel co-registration required in the DPCA cannot be achieved directly by an azimuth time shift. In this study, a calibration method based on scanning geometry of the dual-channel sliding spotlight SAR is developed to address this issue. Moreover, the effect of the phase mismatch calibration on the estimation of azimuth time difference between the two channels is derived and analyzed in depth. The clutter suppression results processed from experimental data acquired by a C-band dual-channel SAR system (Gaofen-3) operated in sliding spotlight mode are shown for the first time to demonstrate the effective phase mismatch calibration.

Abstract Bundle recommendation is an emerging research direction in the recommender system with the focus on recommending customized bundles of items for users. Although Graph Neural Networks (GNNs) have been applied in this problem and achieve superior performance, existing methods underexplore the graph-level GNN methods, which exhibit great potential in traditional recommender system. Furthermore, they usually lack the transferability from one domain with sufficient supervision to another domain which might suffer from the label scarcity issue. In this work, we propose a subgraph-based Graph Neural Network model, SUGER, for bundle recommendation to handle these limitations. SUGER generates heterogeneous subgraphs around the user-bundle pairs, and then maps those subgraphs to the users' preference predictions via neural relational graph propagation. Experimental results show that SUGER significantly outperforms the state-of-the-art baselines in both the basic and the transfer bundle recommendation problems.

Abstract Hepatic endoplasmic reticulum (ER) stress is a hallmark of obesity-induced liver steatosis and contributes to the progress of steatosis and insulin resistance in liver. However, its influence on adipose function is still unclear. Here, we identify a hepatic ER stress-induced activating transcription factor 4 (ATF4)-cold-inducible RNA-binding protein (CIRP)-angiopoietin-related protein3 (ANGPTL3) cascade critical for the regulation of adipose browning. We find that obesity increases CIRP expression in liver through ER stress-induced ATF4. CIRP in turn binds to the 3' UTR and increases mRNA stability of ANGPTL3. ANGPTL3 secreted from liver suppresses uncoupling protein 1 expression through integrin α

Abstract Background: This study aimed to compare the postoperative quality of life (PQOL) between non-small-cell lung cancer (NSCLC) patients who underwent video-assisted thoracoscopic sublobar resection (subsegment, segment, or wedge) and lobectomy. Meanwhile, we developed a PQOL scale for patients with NSCLC after optimization.
Methods: Developing and evaluating the postoperative quality-of-life scale of non-small-cell lung cancer (NSCLC-PQOL) followed by the international principles for developing quality-of-life scale. Therefore, we used the NSCLC-PQOL scale to evaluate the PQOL of patients who underwent different surgeries.
Results: The overall PQOL of patients who underwent video-assisted thoracoscopic lobectomy and sublobar resection gradually worsened from discharge to 3 months postoperatively and progressively improved from three to 6 months postoperatively. And the sublobar resection group showed better PQOL in chest tightness, breath shortness, breathlessness, cough and expectoration than the lobectomy group, and the differences were statistically significant (P < 0.05). The final version of the NSCLC-PQOL contained three dimensions: "signs-symptoms", "psychological and psychiatric", and "social-life" dimensions.
Conclusions: The sublobar resection group showed better PQOL in "chest tightness", "breath shortness", "breathlessness", "cough", and "expectoration" than the lobectomy group. Twenty-two items formed a well-behaved PQOL scale after being validated satisfactorily. The scale was a suitable rating tool for evaluating the NSCLC-PQOL of patients.
Trial registration: As this study was a retrospective study and not a clinical trial, we did not register this study in the Chinese Clinical Trial Registry.

Abstract A narrow linewidth optical frequency comb (OFC) based on a directly modulated microcavity laser with external optical feedback is investigated numerically and demonstrated experimentally. Based on the numerical simulations with rate equations, the evolution of the optical and electrical spectra is presented for the direct-modulated microcavity laser with increased feedback strength, and the linewidth property is improved at suitable feedback conditions. The simulation results also show good robustness for the generated OFC in terms of feedback strength and phase. Moreover, the OFC generation experiment is performed by combining with the dual-loop feedback structure to suppress the side mode, and an OFC with a side-mode suppression ratio of 31 dB is realized. Thanks to the high electro-optical response of the microcavity laser, a 15-tone OFC with a frequency interval of 10 GHz is obtained. Finally, the linewidth of each comb tooth is measured to be around 7 kHz under the feedback power of 47 µW, which indicates an enormous compression of approximately 2000 times compared with the free-running continuous-wave microcavity laser.
