    Abstract PIWI-interacting RNAs (piRNAs) are important for ensuring the integrity of the germline. 3'-terminal 2'-O-methylation is essential for piRNA maturation and to protect them from degradation. HENMT1 (HEN Methyltransferase 1) carries out the 2'-O-methylation, which is of key importance for piRNA stability and functionality. However, neither the structure nor the catalytic mechanism of mammalian HENMT1 have been studied. We have constructed a catalytic-competent HENMT1 complex using computational approaches, in which Mg2+ is primarily coordinated by four evolutionary conserved residues, and is further auxiliary coordinated by the 3'-O and 2'-O on the 3'-terminal nucleotide of the piRNA. Our study suggests that metal has limited effects on substrate and cofactor binding but is essential for catalysis. The reaction consists of deprotonation of the 2'-OH to 2'-O and a methyl transfer from SAM to the 2'-O. The methyl transfer is spontaneous and fast. Our in-depth analysis suggests that the 2'-OH may be deprotonated before entering the active site or it may be partially deprotonated at the active site by His800 and Asp859, which are in a special alignment that facilitates the proton transfer out of the active site. Furthermore, we have developed a detailed potential reaction scenario indicating that HENMT1 is Mg2+ utilizing but is not a Mg2+ dependent enzyme.

    Abstract Background: Lenvatinib is an important molecular target drug for the treatment of advanced hepatocellular carcinoma (HCC). However, the application of molecular targeted therapies for HCC also faces some challenges. Cumulative evidence has also shown that curcumol is a potential anti-HCC drug. Curcumol can be used as a chemosensitizer to enhance the antitumor effect of chemotherapeutic drugs. The purpose of our study is to explore the effect of curcumol combined with lenvatinib on HCC.
    Methods: The antitumor effects of curcumol or/and lenvatinib on Huh 7 cells of the HCC cell line were examined using the cell counting kit-8 (CCK-8) assay, colony formation assay, and transwell assay. For

    Abstract Recent studies reveal that de novo gene origination from previously non-genic sequences is a common mechanism for gene innovation. These young genes provide an opportunity to study the structural and functional origins of proteins. Here, we combine high-quality base-level whole-genome alignments and computational structural modeling to study the origination, evolution, and protein structures of lineage-specific de novo genes. We identify 555 de novo gene candidates in D. melanogaster that originated within the Drosophilinae lineage. Sequence composition, evolutionary rates, and expression patterns indicate possible gradual functional or adaptive shifts with their gene ages. Surprisingly, we find little overall protein structural changes in candidates from the Drosophilinae lineage. We identify several candidates with potentially well-folded protein structures. Ancestral sequence reconstruction analysis reveals that most potentially well-folded candidates are often born well-folded. Single-cell RNA-seq analysis in testis shows that although most de novo gene candidates are enriched in spermatocytes, several young candidates are biased towards the early spermatogenesis stage, indicating potentially important but less emphasized roles of early germline cells in the de novo gene origination in testis. This study provides a systematic overview of the origin, evolution, and protein structural changes of Drosophilinae-specific de novo genes.

    Abstract Sodium-dependent glucose transporters 2 (SGLT2) inhibitors are a class of small-molecule drugs that have gained significant attention in recent years for their potential clinical applications in the treatment of type 2 diabetes mellitus (T2DM). These inhibitors function by obstructing the kidneys' ability to reabsorb glucose, resulting in a rise in the excretion of glucose in urine (UGE) and subsequently lowering blood glucose levels. Several SGLT2 inhibitors, such as Dapagliflozin, Canagliflozin, and Empagliflozin, have been approved by regulatory authorities and are currently available for clinical use. These inhibitors have shown notable enhancements in managing blood sugar levels, reducing body weight, and lowering blood pressure in individuals with T2DM. Additionally, they have exhibited potential advantages in decreasing the likelihood of cardiovascular incidents and renal complications among this group of patients. This review article focuses on the synthesis and clinical application of small-molecule SGLT2 inhibitors, which have provided a new therapeutic approach for the management of T2DM.

    Abstract In the information age, with the continuous development of Internet technology, information data occupies every field of contemporary society. The development of the big data age makes these data more and more prominent. While users read the information they need from these massive data, data quality has also become a concern of users. A large number of data are preprocessed before data analysis, such as some duplicate values, missing values deal with inaccurate and other abnormal data, and filter the data through the data cleaning system to improve the standardization of the data, so as to improve the analysis efficiency of the data, reduce some unnecessary expenses, and save time and effort. The data cleaning system in this paper is implemented based on flash framework. Taking Python as the main language for data cleaning, technical cleaning and standard integration are carried out for some structural problems, duplication problems and missing problems of some different source data. Through the processing of abnormal data, the data quality and data analysis efficiency are greatly improved.

    Abstract With the changing behavior of land use by explosive population growth, the ecological environment is constantly disturbed and destroyed by humans, and the value of ecosystem services is obviously degraded. After the introduction of the national new-type urbanization plan, counties have become the key areas to promote new-type urbanization, and county land use is exceptionally active. Therefore, it is especially urgent to grasp the impact of county land use changes on ecosystem service values. Based on remote sensing satellite image interpretation to obtain county land use classification data, the changes of ecosystem service values in County between 1995 and 2015 were thoroughly explored. The research results show that the total value of ecosystem services has increased in most regions during the 20 years, among which, the service value of Shunchang County has increased most significantly. In terms of the service values of different land use types in each region, woodland is the main contributor to ecological services in different regions. From different time periods, Jian’ou City had the highest value of woodland ecosystem services in 1995 and 2015. In contrast, Songxi County had a lower value of services for each ecosystem type during 20 years.
