    Abstract Tetramethylpyrazine (TMP) is one of the active ingredients of Chuan Xiong that has been reported to have effects on numerous diseases, including diabetic nephropathy (DN). Whereas, related molecular mechanisms are not fully elucidated. We aimed to explore circACTR2's role in TMP-mediated protective effects on DN. In vitro DN condition was established in human kidney cells (HK-2) by treating high glucose (HG). CCK-8 assay and flow cytometry assay were used to observe cell viability and survival. Oxidative stress was determined by the associated markers using kits. The release of inflammatory factors was detected using ELISA kits. Quantitative real-time PCR (qPCR) and western blot were utilized for expression analysis of cricACTR2, miR-140-5p, and GLI pathogenesis-related 2 (GLIPR2). The binding between miR-140-5p and circACTR2 or GLIPR2 was confirmed by dual-luciferase, RIP, and pull-down studies. HG largely induced HK-2 cell apoptosis, oxidative stress, and inflammation, which were alleviated by TMP. CircACTR2's expression was enhanced in HG-treated HK-2 cells but attenuated in HG + TMP-treated HK-2 cells. CircACTR2 overexpression attenuated the functional effects of TMP and thus restored HG-induced cell apoptosis, oxidative stress, and inflammation. CircACTR2 bound to miR-140-5p to enhance the expression of GLIPR2. MiR-140-5p restoration or GLIPR2 inhibition reversed the role of circACTR2 overexpression. CircACTR2 attenuated the protective effects of TMP on HG-induced HK-2 cell damages by regulating the miR-140-5p/GLIPR2 network, indicating that circACTR2 was involved in the functional network of TMP in DN.

    Abstract As a classical local weighting least-square method, the geographic weighted regression (GWR) model always suffers from the space sparsity of samples and the local multicollinearity of predictors, which results in the uncertainties of the model results show spatial heterogeneity. By constructing accuracy evaluation metric of posterior standard error based on the covariance propagation law, this study proposed an uncertainty measuring and constraining method for geographic weighted regression model and validated this method using the instance of ground PM2.5 concentration remote sensing mapping. After uncertainty constraint, the results show the fitted accuracy and sample-based/site-based/regional-based cross validation accuracy for GWR model with different parameters are all improved; the sign error of regression coefficients caused by local multicollinearity are also corrected; the outlier and negative values in the GWR predicted values can also be effectively detected which improve the reliability of the ground PM2.5 concentration mapping results. The proposed method can effectively guarantee the stability and effectiveness of GWR results.

    Abstract The characteristics of synthetic aperture radar (SAR) images are easily affected by factors such as sensor parameters, imaging scenes, etc., which may lead to data distributional discrepancies between the ideal training phase and the practical application phase, resulting in generalization performance degradation of deep learning based detectors in practice. In this paper, an accurate and fast domain adaptive detection method is proposed to address the performance degradation problem of cross-scene detection without requiring additional manual annotations. The cross-scene detection performance is gradually improved by confidence-induced unsupervised feature adaptation (CUFA) and uncertainty-aware adaptive pseudo-label learning (UAPL). In CUFA, image-level and instance-level feature adaptation are progressively implemented to bridge the distribution discrepancies between the source and the target domain in the latent feature space. Then, UAPL is performed to adaptively introduce accurate and stable supervisions of target domain according to the learning status of the detector, avoiding error accumulations and further improving the cross-scene target detection performance. The proposed cross-scene detection method is validated on airborne and spaceborne SAR image datasets of different imaging scenes. The experimental results show that by combining both CUFA and UAPL, compared with the second-best comparison method, the proposed method significantly improves the cross-scene target detection performance in SAR images (at least + 6.5% AP50 and F1 in miniSAR and FARAD cross-scene tasks, and at least + 7.5% AP50 and F1 in Gaofen-3 and Sentinel-1 cross-scene tasks), which facilitates application of the deep learning based detectors in the realistic scenarios.

    Abstract Objective To evaluate the effectiveness of Ommaya sac combined with external ventricular drainage in improving neurological function after periventricular-intraventricular hemorrhage (PIVH) in neonates. Methods A retrospective cohort study was carried out on the neonates hospitalized due to PIVH in the neonate and neurosurgery departments in our hospital from January 2015 to July 2020. They were divided into Ommaya sac combined with external ventricular drainage surgery group (surgery group) and non-surgery group. All of them were followed up until they were 12 months old. The concentration of inflammatory factors in cerebrospinal fluid in the early stage, the incidence of complications (epilepsy, hydrocephalus, cerebral palsy, and death) during follow-up, and neurodevelopmental outcomes were observed and analyzed in the 2 groups. Results A total of 71 PIVH neonates were enrolled, including 35 cases treated with Ommaya capsule combined with external ventricular drainage, and 36 cases treated without surgery. The levels of neuron enolase (NSE), nerve growth factor (NGF) and neutrophil-to-lymphocyte ratio (NLR) in the cerebrospinal fluid in 3 d after surgery were significantly decreased than the levels before surgery (P < 0.05). In the non-surgery group, no such differences in the above indexes were seen in 3 d after treatment (P>0.05). During the follow-up to 12 months of corrected age, 1 neonate (2.9%) died in the surgery group and 6 neonates (16.7%) died in the non-surgery group. The incidences of complications in the surgery group and non-surgery group were compared as follows: epilepsy (0 vs 2 cases), hydrocephalus (4 vs 9 cases) and cerebral palsy (1 vs 3 cases), and statistical differences were seen in the incidences (P < 0.05). Gesell Developmental Scale score indicated that the developmental of the surviving children in the surgery group were significantly better than those in the non-surgery group in terms of adaptability, fine motor, language and personal social interaction (all P < 0.05). ...

    Abstract Intraventricular hemorrhage results in posthemorrhagic hydrocephalus (PHH). Neonatal hydrocephalus remains a challenging disease due to the high failure rate of all management strategies. We evaluated long noncoding RNA growth arrest-specific 5 (GAS5)-mediated network in neonatal hydrocephalus, providing a new direction for the treatment of hydrocephalus. The PHH model was constructed in neonatal rats after intracerebroventricular injection with GAS5, miR-325-3p, and chaperonin containing T-complex protein 1, subunit 8 (CCT8) plasmids, or oligonucleotides. Next, behavioral tests, measurement of serum inflammation, observation of brain tissue pathology, and calculation of hemoglobin and brain water contents were implemented. GAS5, miR-325-3p, and CCT8 expression, in combination with their interactions, was checked. As the results reported, collagenase infusion induced hydrocephalus, impairing neurological function, enhancing inflammation and neuronal apoptosis, and increasing hemoglobin and brain water contents. GAS5 and CCT8 were up-regulated, while miR-325-3p was down-regulated in hydrocephalic rats. Downregulating GAS5/CCT8 or upregulating miR-325-3p could inhibit inflammatory response and improve neurological function in young hydrocephalic rats. GAS5 promotes CCT8 expression through sponge adsorption of miR-325-3p. GAS5 silencing-mediated protections against hydrocephalus were counteracted by CCT8 overexpression. In summary, GAS5 aggravates neonatal hydrocephalus and inflammatory responses in a way of leasing miR-325-3p-involved regulation of CCT8.

    Abstract Developing a gradient porous scaffold similar to bone structure is gaining increasing attention in bone tissue engineering. The GelMA/HAP hydrogel has demonstrated potential in bone repair. Although 3D printing can build GelMA/HAP with porous structure, fabricating porous GelMA/HAP with gradient porosity and pore size in one step remains challenging. In this paper, a gradient porous structure with controllable pore size, based on gelatin methacryloyl (GelMA) and hydxroxyapatite (HAP), was engineered and printed using stereolithography. Firstly, the GelMA and HAP were mixed to prepare a hydrogel with a solid content ranging from 10 wt% to 50 wt% for stereolithography. Taking advantage of the sol-gel characteristics of GelMA/HAP hydrogel, GelMA/HAP was fed on the workbench through a combination of extrusion and paving to form a thin layer. During the curing of each layer, the hydrogel exposed to the curing of a single UV beam immediately solidified, forming a highly interconnected porous structure. Additionally, the hydrogel outside the scanning range could be further polymerized to form a relatively dense structure due to the residual laser energy. Finally, without gradient structural design or changing printing parameters, the gradient porous structure of bone-like could be printed in a single-step process. By adjusting the curing parameters of the single UV beam and the concentration and size of ceramic in the hydrogel, the printed pore diameter of the spongy structure could be controlled within the range of 50-260 μm, while the thickness of the compact area could be adjusted within 130-670 μm.
