    Abstract Black tea, rich in various and complex polyphenols, is one of the most popular beverages in the world due to its pleasant aroma and health benefits, such as alleviate hyperglycemia and antioxidant. The extensive investigation on the chemical constituents of Yunnan black tea 'Jin-Ya', produced from the leaf buds of Camellia sinensis var. assamica, led to the isolation and identification of 16 flavonoids including three new flavan-3-ols, namely theaflavoids A-C (1–3). Most of the isolates displayed obvious inhibitory activity on α-glucosidase with IC₅₀ values ranging from 1.11 to 24.7 μM. Notably, the new theaflavoids 1–3 showed much stronger inhibitory activity (IC₅₀ = 1.34, 1.11, and 4.57 μM, respectively) than those of the positive controls (acarbose and quercetin, IC₅₀ = 228 and 9.38 μM, respectively). In addition, the isolated flavonol glycosides 5–16 were evaluated for their antioxidant activities by DPPH and ABTS ⁺ radical scavenging assays. Those with galloyl group in molecular exhibited comparable free radical scavenging activities to the positive controls (ascorbic acid and Trolox), while most of the flavonol glycosides showed only moderate activities. The results indicated that Yunnan black tea 'Jin-Ya' is an ideal drink with hypoglycemic and antioxidant property, and is worthy of regular consumption.

    Abstract Plant microRNAs (miRNAs) are a class of small non-coding RNAs which target and regulate the expression of genes involved in several growth, development, and metabolism processes. The miR169 family is the largest and most conserved miRNA family in plants. Recent researches have shown involvement of miR169 members in the regulation of plant responding to abiotic stress and development. Computational prediction and experimental analyses suggest that miR169 targets members of the transcription factor NF-YA gene family. In this review, we summarize the origins, evolution and diversity of miR169 family members. In addition, a comprehensive understanding the regulatory functions of the miR169/NF-YA module in plant with particular emphasis on stress-induced flowering is discussed.

    Abstract Construction of the 998.64-km Linzhi–Ya’an section of the Sichuan–Tibet Railway has been influenced by landslide disasters, threatening the safety of Sichuan–Tibet railway projects. Landslide identification and deformation analysis in this area are urgently needed. In this context, it was the first time that 164 advanced land-observing satellite-2 (ALOS-2) phased array type L-band synthetic aperture radar-2 (PALSAR-2) images were collected to detect landslide disasters along the entire Linzhi–Ya’an section. Interferogram stacking and small baseline interferometry methods were used to derive the deformation rate and time-series deformation from 2014–2020. After that, the hot spot analysis method was introduced to conduct spatial clustering analysis of the annual deformation rate, and the effective deformation area was quickly extracted. Finally, 517 landslide disasters along the Linzhi–Ya’an route were detected by integrating observed deformation, Google Earth optical images, and external geological data. The main factors controlling the spatial landslide distribution were analyzed. In the vertical direction, the spatial landslide distribution was mainly concentrated in the elevation range of 3000–5000 m, the slope range of 10–40°, and the aspect of northeast and east. In the horizontal direction, landslides were concentrated near rivers, and were also closely related to earthquake-prone areas, fault zones, and high-precipitation areas. In short, rainfall, freeze–thaw weathering, seismic activity, and fault zones are the main factors inducing landslides along this route. This research provides scientific support for the construction and operation of the Linzhi–Ya’an section of the Sichuan–Tibet Railway.

    Abstract As a subgroup of flavoalkaloids, N-ethyl-2-pyrrolidinone substituted flavan-3-ols are reported to possess various biological activities that may play important roles in the beneficial healthcare functions of tea. The HPLC and LC-MS analyses showed the existence of N-ethyl-2-pyrrolidinone substituted flavan-3-ols in ‘Jin-Ya’, which is a Yunnan black tea produced only from the buds of the tea plant Camellia sinensis var. assamica. Further phytochemical study on this precious black tea led to the identification of 8 flavoalkaloids, 1–8, along with 11 known flavan-3-ols (9–14) and flavonol glycosides (15–19). The new compounds, (−)-6-(5″S)-N-ethyl-2-pyrrolidinone-epiafzelechin (1), (−)-8-(5″R)-N-ethyl-2-pyrrolidinone-epiafzelechin-3-O-gallate (2a), and (−)-8-(5″S)-N-ethyl-2-pyrrolidinone-epiafzelechin-3-O-gallate (2b), were identified based on extensive spectroscopic analysis. Flavoalkaloids 2–6 showed inhibitory activity on α-glucosidase with IC₅₀ values ranging from 2.09 to 8.47 μM, compared to those of quercetin and acarbose (IC₅₀ = 6.87 and 228.9 μM, respectively). Moreover, compounds 2, 3, and 6 displayed an inhibitory effect on acetyl-cholinesterase with IC₅₀ values of 14.23, 33.79, and 34.82 μM, respectively, compared to tacrine (IC₅₀ = 0.223 μM).

    Abstract Hematopoietic stem cell (HSC) self-renewal and differentiation are influenced through multiple pathways, including homeobox transcription factors, signaling through beta-catenin and Notch-1, telomerase, and p27. How these multiple pathways interact and are orchestrated is currently unknown. We now report that NF-Ya, the regulatory and DNA-binding subunit of the trimeric transcription factor NF-Y, plays a central, integrating role in several of these HSC pathways. NF-Ya is preferentially expressed in HSC-enriched bone marrow subpopulations, and NF-Ya mRNA rapidly declines with HSC differentiation. Overexpression of NF-Ya in primitive hematopoietic cells activates the transcription of multiple HOX4 paralogs, as well as Notch-1, LEF-1, and telomerase RNA. HSCs overexpressing NF-Ya are biased toward primitive hematopoiesis in vitro and show strikingly increased in vivo repopulating abilities after single or sequential bone marrow transplantation. Thus, NF-Ya is a potent cellular regulator of HSC self-renewal.
