    Abstract Background: The use of high-throughput genotyping techniques has enabled us to identify the rare germline genetic variants with different pathogenicity and penetrance, and understand their role in cancer predisposition. We report here a familial cancer case, a study from Western Indian.
    Methods: NGS-WES was carried out in a lung cancer patient who has a family history of multiple cancers across generations, including tongue, lung, brain, cervical, urothelial, and esophageal cancer. The results were validated by data mining from available data bases. I-TASSER, RasMol and PyMol were used for protein structure modelling.
    Results: The sequencing by NGS-WES revealed PPM1D c.1654C>T (p.Arg552Ter) mutation in hotspot region exon 6 leading to sudden protein truncation and loss of the C-terminal, due to the substitution of C>T. This mutation was classified as a variant of uncertain significance (VUS), due to limited data on lung cancer, The three unaffected siblings of proband did not show any pathogenic variants and comparative analysis of the four siblings indicate 9 shared genetic variants, classified as benign as per ClinVar.
    Conclusion: PPM1D constitutional genetic alterations are rare and uncommon in different ethnic populations. This gene encodes a phosphatase playing role in regulating the P53 tumor suppressor pathway and DNA damage response. Genetic alterations in the PPM1D gene maybe linked to history of gliomas, breast cancer, and ovarian cancer onset in the proband's family.

    Abstract Huntington's disease (HD) is a complex progressive neurodegenerative disorder affected by genetic, environmental, and metabolic factors contributing to its pathogenesis. Gut dysbiosis is termed as the alterations of intestinal microbial profile. Emerging research has highlighted the pivotal role of gut dysbiosis in HD, focusing on the gut-brain axis as a novel research parameter in science. This review article provides a comprehensive overview of gut microbiota dysbiosis and its relationship with HD and its pathogenesis along with the future challenges and opportunities. The focuses on the essential mechanisms which link gut dysbiosis to HD pathophysiology including neuroinflammation, immune system dysregulation, altered metabolites composition, and neurotransmitter imbalances. We also explored the impacts of gut dysbiosis on HD onset, severity, and symptoms such as cognitive decline, motor dysfunction, and psychiatric symptoms. Furthermore, we highlight recent advances in therapeutics including microbiota-based therapeutic approaches, including dietary interventions, prebiotics, probiotics, fecal microbiota transplantation, and combination therapies with conventional HD treatments and their applications in managing HD. The future challenges are also highlighted as the heterogeneity of gut microbiota, interindividual variability, establishing causality between gut dysbiosis and HD, identifying optimal therapeutic targets and strategies, and ensuring the long-term safety and efficacy of microbiota-based interventions. This review provides a better understanding of the potential role of gut microbiota in HD pathogenesis and guides the development of novel therapeutic approaches.

    Hybrid Approach to Learning Segmentation by Simultaneously Exploiting Image and Event Modalities

    Abstract Event cameras detect changes in per-pixel intensity to generate asynchronous `event streams'. They offer great potential for accurate semantic map retrieval in real-time autonomous systems owing to their much higher temporal resolution and high dynamic range (HDR) compared to conventional cameras. However, existing implementations for event-based segmentation suffer from sub-optimal performance since these temporally dense events only measure the varying component of a visual signal, limiting their ability to encode dense spatial context compared to frames. To address this issue, we propose a hybrid end-to-end learning framework HALSIE, utilizing three key concepts to reduce inference cost by up to $20\times$ versus prior art while retaining similar performance: First, a simple and efficient cross-domain learning scheme to extract complementary spatio-temporal embeddings from both frames and events. Second, a specially designed dual-encoder scheme with Spiking Neural Network (SNN) and Artificial Neural Network (ANN) branches to minimize latency while retaining cross-domain feature aggregation. Third, a multi-scale cue mixer to model rich representations of the fused embeddings. These qualities of HALSIE allow for a very lightweight architecture achieving state-of-the-art segmentation performance on DDD-17, MVSEC, and DSEC-Semantic datasets with up to $33\times$ higher parameter efficiency and favorable inference cost (17.9mJ per cycle). Our ablation study also brings new insights into effective design choices that can prove beneficial for research across other vision tasks.
