    Abstract The p6 domain of the Gag precursors (Gag p6) in human immunodeficiency virus type 1 (HIV-1) plays multifunctional roles in the viral life cycle. It utilizes the endosomal sorting complex required for transport (ESCRT) system to facilitate viral budding and release from the plasma membrane through the interactions with the ESCRT-I component tumor susceptibility gene 101 (TSG101) and with the ALG-2 interacting protein X (ALIX). Moreover, Gag p6 contributes to viral replication by a range of posttranslational modifications such as SUMOylation, ubiquitination and phosphorylation. Additionally, Gag p6 also mediates the incorporation of the accessory protein Vpr into virions, thereby promoting Vpr-induced viral replication. However, less attention is focused on Gag p6 as therapeutic intervention. This review focuses on the structures and diverse functions of Gag p6 in viral replication, host cells, and pathogenesis. Additionally, several challenges were also discussed in studying the structure of Gag p6 and its interactions with partners. Consequently, it concludes that the Gag p6 represents an attractive target for the development of antiretroviral drugs, and efforts to develop p6-targeted antiretrovirals are expected to undergo significant growth in the forthcoming years.

    Abstract To support indoor scene understanding, room layouts have been recently introduced that define a few typical space configurations according to junctions and boundary lines. In this paper, we study camera pose estimation from eight common room layouts with at least two boundary lines that is cast as a PnL (Perspective-n-Line) problem. Specifically, the intersecting points between image borders and room layout boundaries, named image outer corners (IOCs), are introduced to create additional auxiliary lines for PnL optimization. Therefore, a new PnL-IOC algorithm is proposed which has two implementations according to the room layout types. The first one considers six layouts with more than two boundary lines where 3D correspondence estimation of IOCs creates sufficient line correspondences for camera pose estimation. The second one is an extended version to handle two challenging layouts with only two coplanar boundaries where correspondence estimation of IOCs is ill-posed due to insufficient conditions. Thus the powerful NSGA-II algorithm is embedded in PnL-IOC to estimate the correspondences of IOCs. At the last step, the camera pose is jointly optimized with 3D correspondence refinement of IOCs in the iterative Gauss-Newton algorithm. Experiment results on both simulated and real images show the advantages of the proposed PnL-IOC method on the accuracy and robustness of camera pose estimation from eight different room layouts over the existing PnL methods.     Abstract APOB-containing lipoproteins are large, complex lipid carriers that ferry bulk lipids into the circulation via the secretory pathway, originating from the endoplasmic reticulum of specialized cells in the liver or the gut. Elevation of APOB-containing lipoproteins in the plasma represents a major risk factor for cardiovascular diseases. The production of these lipoproteins requires enzyme-catalyzed, cross-membrane transfer of neutral lipids and phospholipids to lipoproteins, in particular onto the structural component APOB. Transport of these lipid-bearing cargos relies on the COPII machinery and employs the transmembrane cargo receptor SURF4 and the small GTPase SAR1B, together constituting a selective transport program. Intriguingly, a number of factors implicated in lipoprotein production are also packaged into COPII vesicles and may be cotransported with APOB. These observations therefore point to a specialized produce-and-export itinerary during the secretion of these lipid-bearing cargos, warranting future investigations into this unique yet pivotal process at the crossroad of cell biology and physiology.

    Abstract Generalized zero-shot learning (GZSL) is one of the most realistic but challenging problems due to the partiality of the classifier to supervised classes, especially under the class-inductive instance-inductive (CIII) training setting, where testing data are not available. Instance-borrowing methods and synthesizing methods solve it to some extent with the help of testing semantics, but therefore neither can be used under CIII. Besides, the latter require the training process of a classifier after generating examples. In contrast, a novel non-transductive regularization under CIII called Semantic Borrowing (SB) for improving GZSL methods with compatibility metric learning is proposed in this paper, which not only can be used for training linear models, but also nonlinear ones such as artificial neural networks. This regularization item in the loss function borrows similar semantics in the training set, so that the classifier can model the relationship between the semantics of zero-shot and supervised classes more accurately during training. In practice, the information of semantics of unknown classes would not be available for training while this approach does NOT need it. Extensive experiments on GZSL benchmark datasets show that SB can reduce the partiality of the classifier to supervised classes and improve the performance of generalized zero-shot classification, surpassing inductive GZSL state of the arts.

    Abstract Animal behavior analysis plays a crucial role in contemporary neuroscience research. However, the performance of the frame-by-frame approach may degrade in scenarios with occlusions or motion blur. In this study, we propose a spatiotemporal network model based on YOLOv8 to enhance the accuracy of key-point detection in mouse behavioral experimental videos. This model integrates a time-domain tracking strategy comprising two components: the first part utilizes key-point detection results from the previous frame to detect potential target locations in the subsequent frame; the second part employs Kalman filtering to analyze key-point changes prior to detection, allowing for the estimation of missing key-points. In the comparison of pose estimation results between our approach, YOLOv8, DeepLabCut and SLEAP on videos of three mouse behavioral experiments, our approach demonstrated significantly superior performance. This suggests that our method offers a new and effective means of accurately tracking and estimating pose in mice through spatiotemporal processing.

    Abstract Crowd evacuation has gained increasing attention due to its importance in the day-to-day management of public areas. During an emergency evacuation, there are a variety of factors that need to be considered when designing a practical evacuation model. For example, relatives tend to move together or look for each other. These behaviors undoubtedly aggravate the chaos degree of evacuating crowds and make evacuations hard to model. In this paper, we propose an entropy-based combined behavior model to better analyze the influence of these behaviors on the evacuation process. Specifically, we utilize the Boltzmann entropy to quantitatively denote the degree of chaos in the crowd. The evacuation behavior of heterogeneous people is simulated through a series of behavior rules. Moreover, we devise a velocity adjustment method to ensure the evacuees follow a more orderly direction. Extensive simulation results demonstrate the effectiveness of the proposed evacuation model and provide useful insights into the design of practical evacuation strategies.
