    Abstract Yeasts are widely used cell factories for commercial heterologous protein production, however, specific productivities are usually tightly coupled to biomass formation. This greatly impacts production processes, which are commonly not run at the maximum growth rate, thereby resulting in suboptimal productivities. To tackle this issue, we evaluated transcriptomics datasets of the yeast Pichia pastoris (syn. Komagataella phaffii), which is known for its high secretory efficiency and biomass yield. These showed a clear downregulation of genes related to protein translation with decreasing growth rates, thus revealing the yeast translation machinery as cellular engineering target. By overexpressing selected differentially expressed translation factors, translation initiation was identified to be the main rate-limiting step. Specifically, overexpression of factors associated with the closed-loop conformation, a structure that increases stability and rates of translation initiation before start codon scanning is initiated, showed the strongest effects. Overexpression of closed-loop factors alone or in combination increased titers of different heterologous proteins by up to 3-fold in fed-batch processes. Furthermore, translation activity, correlating to the obtained secreted recombinant protein yields, selected transcript levels and total protein content were higher in the engineered cells. Hence, translation factor overexpression, globally affects the cell. Together with the observed impact on the transcriptome and total protein content, our results indicate that the capacity of P. pastoris for protein production is not at its limit yet.

    Abstract We examine numerically vortex families near band edges of the Bloch wave spectrum for the Gross-Pitaevskii equation with two-dimensional periodic potentials and for the discrete nonlinear Schrödinger equation. We show that besides vortex families that terminate at a small distance from the band edges via fold bifurcations, there exist vortex families that are continued all the way to the band edges.

    Abstract We study the dynamics of 2D gap solitons (GSs) in Bragg resonant nonlinear (photonic) gratings in the presence of localized defects. Previous work [Stud. Appl. Math.115, 209 (2005)] explains the mechanism of trapping the GS-carried energy at a defect via a resonant energy transfer from the GS into defect modes. We derive a finite-dimensional model that describes the evolution of the defect-trapped state as an interaction of linear defect modes and show that this model approximates the full dynamics very well in the regime when moderate amounts of GS energy are trapped.

    Abstract Background and purpose: Emerging evidence indicates that hypertension is mediated by immune mechanisms. We hypothesized that exposure to Porphyromonas gingivalis antigens, commonly encountered in periodontal disease, can enhance immune activation in hypertension and exacerbate the elevation in BP, vascular inflammation and vascular dysfunction.
    Experimental approach: Th1 immune responses were elicited through immunizations using P. gingivalis lysate antigens (10 μg) conjugated with aluminium oxide (50 μg) and IL-12 (1 μg). The hypertension and vascular endothelial dysfunction evoked by subpressor doses of angiotensin II (0.25 mg·kg
    Key results: Systemic T-cell activation, a characteristic of hypertension, was exacerbated by P. gingivalis antigen stimulation. This translated into increased aortic vascular inflammation with enhanced leukocyte, in particular, T-cell and macrophage infiltration. The expression of the Th1 cytokines, IFN-γ and TNF-α, and the transcription factor, TBX21, was increased in aortas of P. gingivalis/IL-12/aluminium oxide-immunized mice, while IL-4 and TGF-β were unchanged. These immune changes in mice with induced T-helper-type 1 immune responses were associated with an enhanced elevation of BP and endothelial dysfunction compared with control mice in response to 2 week infusion of a subpressor dose of angiotensin II.
    Conclusions and implications: These results support the concept that Th1 immune responses induced by bacterial antigens such as P. gingivalis can increase sensitivity to subpressor pro-hypertensive insults such as low-dose angiotensin II, thus providing a mechanistic link between chronic infection, such as periodontitis, and hypertension.

    Abstract We investigate the statistics of flat-top solitary wave parameters in the presence of weak multiplicative dissipative disorder. We consider first propagation of solitary waves of the cubic-quintic nonlinear Schrödinger equation (CQNLSE) in the presence of disorder in the cubic nonlinear gain. We show by a perturbative analytic calculation and by Monte Carlo simulations that the probability-density function (PDF) of the amplitude eta exhibits loglognormal divergence near the maximum possible amplitude eta(m), a behavior that is similar to the one observed earlier for disorder in the linear gain [A. Peleg, Phys. Rev. E 72, 027203 (2005)]. We relate the loglognormal divergence of the amplitude PDF to the superexponential approach of eta to eta(m) in the corresponding deterministic model with linear/nonlinear gain. Furthermore, for solitary waves of the derivative CQNLSE with weak disorder in the linear gain both the amplitude and the group velocity beta become random. We therefore study analytically and by Monte Carlo simulations the PDF of the parameter p, where p = eta/(1-epsilon(s)beta/2) and epsilon(s) is the self-steepening coefficient. Our analytic calculations and numerical simulations show that the PDF of p is loglognormally divergent near the maximum p value.

    Abstract We study the effects of weak disorder in the linear gain coefficient on front formation in pattern forming systems described by the cubic-quintic nonlinear Schrödinger equation. We calculate the statistics of the front amplitude and position. We show that the distribution of the front amplitude has a loglognormal diverging form at the maximum possible amplitude and that the distribution of the front position has a lognormal tail. The theory is in good agreement with our numerical simulations. We show that these results are valid for other types of dissipative disorder and relate the loglognormal divergence of the amplitude distribution to the form of the emerging front tail.
