    Abstract Numerical simulations are commonly used to understand the parameter dependence of given spatio-temporal phenomena. Sampling a multi-dimensional parameter space and running the respective simulations leads to an ensemble of a large number of spatio-temporal simulation runs. A main objective for analyzing the ensemble is to partition (or segment) the multi-dimensional parameter space into connected regions of simulation runs with similar behavior. To facilitate such an analysis, we propose a novel visualization method for multi-dimensional parameter-space partitions. Our visualization is based on the concept of a hyper-slicer, which allows for undistorted views of the parameter-space segments' extent and transitions. For navigation within the parameter space, interactions with a 2D embedding of the parameter-space samples, including their segment memberships, are supported. Parameter-space partitions are generated in a semi-automatic fashion by analyzing the similarity space of the ensemble's simulation runs. Clusters of similar simulation runs induce the segments of the parameter-space partition. We link the parameter-space partitioning visualizations to similarity-space visualizations of the ensemble's simulation runs and embed them into an interactive visual analysis tool that supports the analysis of all facets of the spatio-temporal simulation ensemble targeted at the overarching goal of analyzing the parameter-space partitioning. The partitioning can then be visually analyzed and interactively refined. We evaluated our approach with experts within case studies from three different domains.

    Abstract Oberservation of particle movement during wet granulation is crucial for identifying granulation mechanisms, process optimizations and troubleshooting. An example of such a process is spheronization, which is commonly used as a wet granulation technique to produce spherical, pharmaceutical pellets. Due to the dense particle bed, measurements of poloidal pellet velocities remain a challenge and have not been yet been reported in literature. In this work, a modelling approach was used in combination with particle image velocimetry (PIV) and mixing progress as a surrogate parameter to obtain information about the movement inside the pellet bed, including those parts not visible from outside the pellet bed. A good agreement between the experiment and the simulation was found. Regions of low poloidal velocities were found in both experiments and simulations. The initial pellet shape was not identified as a significant influence on the mixing progress.

    Abstract Macropinocytosis is one of the major mechanisms by which cancer cells uptake extracellular nutrients from tumor microenvironment (TME) and plays very important roles in various steps of tumorigenesis. We previously reported the unexpected finding that intratumoral and extracellular ATP (eATP), as one of the major drastically upregulated extracellular nutrients and messengers in tumors, is taken up by cancer cells through macropinocytosis in large quantities and significantly contributing to cancer cell growth, survival, and increased resistance to chemo and target drugs. Inhibition of macropinocytosis substantially reduced eATP uptake by cancer cells and slowed down tumor growth in vivo. More recently, we have found the eATP also plays a very important role in inducing epithelial-to-mesenchymal transition (EMT), and that macropinocytosis is an essential facilitator in the induction. Thus, macropinocytosis and eATP, working in coordination, appear to play some previously unrecognized but very important roles in EMT and metastasis. As a result, they are likely to be interactive and communicative with each other, regulating each other's activity for various needs of host tumor cells. They are also likely to be an integral part of the future new anticancer therapeutic strategies. Moreover, it is undoubted that we have not identified all the important activities coordinated by ATP and macropinocytosis. This review describes our findings in how eATP and macropinocytosis work together to promote cancer cell growth, resistance, and EMT. We also list scientific challenges facing eATP research and propose to target macropinocytosis and eATP to reduce drug resistance and slow down metastasis.

    Abstract Integrated river basin management (IRBM) is recently fostered in the European Union mainly by two framework directives which were established in order to realise sustainable and effective river basin management and aiming for integrated approaches on a river basin scale. One is the water framework directive which objective is to assess water quality and achieve a good status for all water bodies. The other one is the flood risk management directive on the assessment and management of flood risks. This paper discusses the potential synergies of the two directives against IRBM in general and describes European experts’ views which were formulated as recommendations. The status of the water bodies and water governance system in Germany are described and critically reflected against the experts’ recommendations. Potential methodological approaches which were developed and tested in German case studies are presented and discussed in the light of IRBM with focus on identifying and using cross-sectoral synergies. The analysis reveals shortcomings in IRBM approaches in Germany and potentials for identification and use of synergies if certain framework, concept approaches and methodological approaches would be used.
