    Abstract Genetic counseling for hearing loss today originated from decoding the genetic code of hereditary hearing loss, which serves as an effective strategy for preventing hearing loss and constitutes a crucial component of the diagnostic and therapeutic framework. This paper described the main principles and contents of genetic counseling for hearing loss, the key points of counseling across various genetic models and its application in tertiary prevention strategies targeting hearing impairment. The prospects of an AI-assisted genetic counseling decision system and the envisions of genetic counseling in preventing hereditary hearing loss were introduced. Genetic counseling for hearing loss today embodies the hallmark of a new era, which is inseparable from the advancements in science and technology, and will undoubtedly contribute to precise gene intervention!

    Abstract Objective: To study the safety and efficacy of high-power ablation for atrial fibrillation (AF) guided by lesion size index (LSI) and impedance cutoff. Method: A total of 223 patients who underwent radiofrequency catheter ablation of atrial fibrillation (including paroxyparal atrial fibrillation and persistent atrial fibrillation) in the Department of Cardiology of Anhui Provincial Hospital from February 2019 to July 2020 were enrolled, and were divided into 123 patients in the high-power ablation group (HPAI) and 100 patients in the conventional power ablation group (CPAI). The HPAI group adopted high-power (40–50 W) ablation by impedance cutoff, and the CPAI group adopted conventional-power (30–35 W) ablation. Patients in both groups were ablated guided by the same LSI. For both groups, we analyzed the pulmonary vein single-circle isolation rate, ablation time, X-ray exposure, impedance drop value, incidence of complications, and recurrence rate within one year after operation. Results: There was no significant difference in the success rate of pulmonary vein single-circle isolation, X-ray perspective time, and X-ray exposure quantity between the HPAI group and the CPAI group (88.60% vs 82.00%, P = 0.161; 8.7 ± 3.74 min vs 7.82 ± 3.86 min, P = 0.067; 54.74 ± 28 min vs 52.78 ± 39.58 min, P = 0.139); the annular pulmonary vein ablation time and total ablation time were less in the HPAI group (35.74 ± 7.25 min vs 65.49 ± 7.34 min, P < 0.01; 55.42 ± 11.61 min vs 76.9 ± 6.79 min, P < 0.01); the impedance drop values at 10–15Ω and 15–20Ω were higher in the HPAI group (25.3% vs 19.1%, P < 0.05; 24.1% vs 19.1%, P < 0.05); there was no significant difference in the recurrence rate within one year after operation between the two groups; and no serious complications occurred in the two groups. Conclusion: High-power ablation guided by LSI and impedance cutoff could significantly shorten the AF ablation time and reduce complications.

    Abstract Epigenetic modifications, including DNA methylation and histone post-translational modifications, intricately regulate gene expression patterns by influencing DNA accessibility and chromatin structure in higher organisms. These modifications are heritable, are independent of primary DNA sequences, undergo dynamic changes during development and differentiation, and are frequently disrupted in human diseases. The reversibility of epigenetic modifications makes them promising targets for therapeutic intervention and drugs targeting epigenetic regulators (e.g., tazemetostat, targeting the H3K27 methyltransferase EZH2) have been applied in clinical therapy for multiple cancers. The NSD family of H3K36 methyltransferase enzymes-including NSD1 (KMT3B), NSD2 (MMSET/WHSC1), and NSD3 (WHSC1L1)-are now receiving drug development attention, with the exciting advent of an NSD2 inhibitor (KTX-1001) advancing to Phase I clinical trials for relapsed or refractory multiple myeloma. NSD proteins recognize and catalyze methylation of histone lysine marks, thereby regulating chromatin integrity and gene expression. Multiple studies have implicated NSD proteins in human disease, noting impacts from translocations, aberrant expression, and various dysfunctional somatic mutations. Here, we review the biological functions of NSD proteins, epigenetic cooperation related to NSD proteins, and the accumulating evidence linking these proteins to developmental disorders and tumorigenesis, while additionally considering prospects for the development of innovative epigenetic therapies.

    Abstract Brain-computer interfaces (BCIs) are anticipated to improve the efficacy of rehabilitation for people with motor disabilities. However, applying BCI in clinical practice is still a challenge due to the great diversity of patients. In the current study, a novel action observation (AO) based BCI was proposed and tested on stroke patients. Ten non-hemineglect patients and ten hemineglect patients were recruited. Four AO stimuli were designed, each presenting a decomposed action to complete the reach-and-grasp task. EEG data and eye movement data were collected. Eye movement data was utilized to analyze the reasons for individual differences in BCI performance. Task discriminative component analysis was utilized to perform online target detection. The results showed that the designed AO-based BCI could simultaneously induce steady state motion visual evoked potential (SSMVEP) from the occipital region and sensory motor rhythm from the sensorimotor region in stroke patients. The average online detection accuracy among the four AO stimuli reached 67% within 3 s in the non-hemineglect group, while the accuracy only reached 35% in the hemineglect group. Gaze metrics showed that the average total duration of fixations during the stimulus phase in the hemineglect group was only 1.31 s ± 0.532 s which was significantly lower than that in the non-hemineglect group. The results indicated that hemineglect patients have difficulty gazing at the AO stimulus, resulting in poor detection performance and weak desynchronization in the sensorimotor region. Furthermore, the degree of neglect is inversely proportional to the target detection accuracy in hemineglect stroke patients. In addition, the gaze metrics associated with cognitive load were significantly correlated with the accuracy in non-hemineglect patients. It indicated the cognitive load may affect the AO-based BCI. The current study will expedite the clinical application of AO-based BCI.

    Abstract Cu/CHA and sodium-promoted Cu/CHA catalysts were prepared and applied to catalyze NH3-SCR reaction. Also, the effect of phosphorus poisoning on the catalytic performance of these two catalysts were explored. Compared with Cu/CHA catalyst, sodium-promoted Cu/CHA catalyst presented better catalytic performance, which was attributed to the optimized distribution of copper active sites. Moreover, it was proved that sodium ions as sacrificial sites could effectively protect copper active ions and improve the phosphorus poisoning resistance of the sodium-promoted Cu/CHA catalyst.
