    Abstract Objective: To investigate secondary adrenal insufficiency post varying traumatic brain injuries' and its impact on prognosis.
    Methods: 120 traumatic brain injury patients were categorized into mild, moderate and severe groups based on Glasgow Coma Scale. Adrenal function was evaluated through testing.
    Results: Secondary adrenal insufficiency rates were 0% (mild), 22.85% (moderate) and 44.82% (severe). Hypothalamus-pituitary-adrenal axis dysfunction rates were 14.81% (mild), 42.85% (moderate) and 63.79% (severe). Differences among groups were significant (
    Conclusion: As traumatic brain injury severity increases, secondary adrenal insufficiency incidence rises. The low-dose adrenocorticotropic hormone test is promising for hypothalamus-pituitary-adrenal axis evaluation. Patients with hypothalamus-pituitary-adrenal dysfunction experience prolonged hospitalization and worse prognosis.

    Abstract Background: Portwine stain (PWS) birthmarks are congenital vascular malformations. The quantification of PWS area is an important step in lesion classification and treatment evaluation.
    Aims: The aim of this study was to evaluate the combination of 3D scan with deep learning for automated PWS area quantization.
    Materials and methods: PWS color was measured using a portable spectrophotometer. PWS patches (29.26-45.82 cm
    Results: IDeepLabV3+ (Xception) showed the best results in PWS lesion extraction and area quantification. Its mean Intersection over Union (MIou) was 0.9797, Mean Pixel Accuracy (MPA) 0.9908, Accuracy 0.9989, Recall 0.9886 and F1-score 0.9897, respectively. In PWS area quantization, the mean value of the area error rate of this scheme was 2.61 ± 2.33.
    Conclusions: The new 3D method developed in this study was able to achieve accurate quantification of PWS lesion area and has potentials for clinical applications.

    Abstract The influence of alloying element Mg on Na and Sr modifying Al-7Si hypoeutectic alloys was investigated. The residual content of Na and the morphology of modified eutectic silicon were characterized. It was found that the alloying element Mg had an enhanced effect on the uptake of sodium in the Al-7Si hypoeutectic alloy modified by the Na-contained modifier. Moreover, the morphology of eutectic silicon of the modified Al-7Si alloys was significantly different from that of Al-7Si-0.4Mg alloys in the present research. When the addition of the modifier is enough, both modifiers could entirely modify the eutectic silicon phase of Al-7Si alloys, while incompletely modified eutectic silicon was observed in both Na-modified and Sr-modified Al-7Si-0.4Mg alloy. It was observed that there was an adhering relationship between the partially modified eutectic silicon with Mg-rich phases. According to the results, it can be proposed that the addition of Mg will affect the solidification behavior of alloys, thereby, leading to the incomplete modification of eutectic silicon phases.

    Abstract Side reaction pathways are observed to occur between antioxidants and peroxide during the processing of multiple resin grades mediated by controlled peroxide-induced degradation of impact polypropylene. In the present work, the reaction mechanism and main by-products between antioxidants and peroxides were investigated. The results demonstrate that peroxides greatly accelerate the decomposition reactions, and the free radicals formed from peroxide decomposition react with, for example, the antioxidant AO1010 to produce dehydrogenation of phenyl propionic unit(s) at the α position (cinnamic acid ester moiety), which generates a conjugated system leading to the increased color of the product. It is the first time to confirm the cinnamic acid moiety's existence and report its NMR data. Further, this work confirms the dehydrogenation mechanism by comparison with different sterically hindered phenolic antioxidants. It also systematically summarizes the oxidation and degradation mechanism of AO168 and AO1010 under air and peroxide environments, respectively. Based on present study, clear guidelines are obtained to improve the quality of polymeric products, especially the appearance and stability, during product development.

    Abstract Background: Thromboangiitis obliterans (TAO), also known as Buerger's disease, is an occlusive arterial disease; however, the pathogenesis of TAO is still unclear. Research has shown that traditional Chinese medicine (TCM) has significant advantages in the treatment of TAO. Our purpose was to explore the underlying roles of TCM in combination with nibble debridement and dressing method (NDDM) in a TAO rat model.
    Methods: We administered rats with 10 mg/mL sodium laurate to establish a TAO model, and then the TAO model rats were treated with notoginseng powder (NP), maifusheng (MFS), or the combination of NP or MFS and NDDM. Gangrene classification and blood rheology were evaluated; the pathological characteristics of rat limbs were examined by hematoxylin and eosin (H&E) staining and Masson staining; and cluster of differentiation 3+ (CD3+) and cluster of differentiation 20+ (CD20+) levels were measured by immunohistochemistry (IHC) and flow cytometry. In addition, inflammation-associated cytokines were analyzed by quantitative reverse transcription polymerase chain reaction (RT-qPCR), western blot, and enzyme-linked immunosorbent assay (ELISA).
    Results: Integration of NP or MFS and NDDM dramatically reduced the gangrene classification and affected blood rheology parameters of TAO model rats compared with NP and MFS alone. Meanwhile, NP or MFS in combination with NDDM decreased CD3+CD20+ T cells, reduced thrombosis and inflammatory cell infiltration, and dramatically decreased the levels of inflammation-associated cytokines.
    Conclusions: Our results suggested that integration of NP or MFS and NDDM could relieve the symptoms of TAO model rats induced by sodium laurate, which might provide a new management strategy for TAO.

    Abstract Checkpoint inhibitor therapy has become increasingly important and has been endorsed as a treatment regimen in breast cancer. But benefits were limited to a small proportion of patients. We aimed to develop an improved signature on the basis of immune genes for detection of potential benefit from immunotherapy. Gene expression data of patients with breast cancer initially extracted from The Cancer Genome Atlas were analyzed. Ten genes were selected from the interaction of differentially expressed genes as well as immune-related genes to develop a survival signature. We compared the high-risk and low-risk groups by gene set enrichment analysis, immune infiltration, checkpoint molecule expression and immunophenoscore. Ten genes were extracted from interactions of differentially expressed and immune-related genes. The immune risk score was determined on the basis of the Cox regression coefficient of hub genes and validated with the GSE96058 dataset. Immune cell infiltrates, including CD8 + T cells, plasma cells, follicular helper T cells, CD4 + memory T cells, M1 macrophages, regulatory T cells and resting NK cells, were more highly infiltrated in the high-risk group as compared to the low-risk group. Checkpoint molecules, including CTLA-4, PD-L1, TIM-3, VISTA, ICOS, PD-1, and PD-L2, were expressed at markedly lower levels in the high-risk group as compared to the low-risk group. Immunophenoscores, as a surrogate of response to immune checkpoint therapy, was observed significant lower in the high-risk group. The 10-gene prognostic signature could identify patients' survival and was correlated with the biomarkers of immune checkpoint inhibitor therapy, which may guide precise therapeutic decisions in clinical practice.

    Abstract Acute lung injury (ALI) is an acute inflammatory process that can result in life-threatening consequences. Programmable DNA nanostructures have emerged as excellent nanoplatforms for microRNA-based therapeutics, offering potential nanomedicines for ALI treatment. Nonetheless, the traditional systematic administration of nanomedicines is constrained by low delivery efficiency, poor pharmacokinetics, and nonspecific side effects. Here, we identify macrophage microRNA-155 as a novel therapeutic target using the magnetic bead sorting technique. We further construct a DNA nanotubular nucleic acid drug antagonizing microRNA-155 (NT-155) for ALI treatment through intratracheal administration. Flow cytometry results demonstrate that NT-155, when inhaled, is taken up much more effectively by macrophages and dendritic cells in the bronchoalveolar lavage fluid of ALI mice. Furthermore, NT-155 effectively silences the overexpressed microRNA-155 in macrophages and exerts excellent inflammation inhibition effects in vitro and ALI mouse models. Mechanistically, NT-155 suppresses microRNA-155 expression and activates its target gene SOCS1, inhibiting the p-P65 signaling pathway and suppressing proinflammatory cytokine secretion. The current study suggests that deliberately designed nucleic acid drugs are promising nanomedicines for ALI treatment and the local administration may open up new practical applications of DNA in the future.
