    Abstract Abstract The Yangjiacun coal mine, located in western China, exhibits distinctive characteristics, such as its shallow depth, thick bedrock, thin loose layer, and unique roof strata breaking movement. In this research, a comprehensive analysis of previous exploration data, subsequent mechanical testing, key stratum theory, natural equilibrium arch theory, and simulation tests and numerical simulations has been conducted to investigate the failure behavior of the overlying rock strata in the Yangjiacun Coal Mine. Specifically, the load‐bearing capacity of the key stratum structure in the bedrock and the morphological properties of the arch‐beam structure have been thoroughly examined. A mechanical model of the roof rock arch‐beam structure has been established, and an analytical formula for the limit size and stability criterion of the arch‐beam structure has been derived. The mining‐induced rock arch‐beam structure plays a crucial role in transferring the load of the upper strata to the surrounding goaf, thereby preventing subsidence and deformation of the overlying strata. However, the arch‐beam structure periodically experiences instability during the advancement of the working face, gradually progressing towards the upper layer until the main key stratum is breached and the arch‐beam structure collapses. Based on the characteristics of weathered rock and the arch‐beam failure model in the Yangjiacun Coal Mine, quantitative backfilling mining methods have been proposed to either compensate for the mining void created by coal extraction or limit the size of exposed key strata through strip mining, ensuring the stability of the R6 key stratum. This study represents a significant contribution to understanding the intricate failure behavior of overlying rock strata in coal mines and provides a valuable framework for the responsible and efficient extraction of coal resources in similar environments.

    Abstract The degeneration of midbrain dopaminergic (mDA) neurons, particularly in the substantia nigra pars compacta (SNc), is one of the most prominent pathological hallmarks of Parkinson's disease (PD). To uncover the pathogenic mechanisms of mDA neuronal death during PD may provide therapeutic targets to prevent mDA neuronal loss and slow down the disease's progression. Paired-like homeodomain transcription factor 3 (Pitx3) is selectively expressed in the mDA neurons as early as embryonic day 11.5 and plays a critical role in mDA neuron terminal differentiation and subset specification. Moreover, Pitx3-deficient mice exhibit some canonical PD-related features, including the profound loss of SNc mDA neurons, a dramatic decrease in striatal dopamine (DA) levels, and motor abnormalities. However, the precise role of Pitx3 in progressive PD and how this gene contributes to mDA neuronal specification during early stages remains unclear. In this review, we updated the latest findings on Pitx3 by summarizing the crosstalk between Pitx3 and its associated transcription factors in mDA neuron development. We further explored the potential benefits of Pitx3 as a therapeutic target for PD in the future. To better understand the transcriptional network of Pitx3 in mDA neuron development may provide insights into Pitx3-related clinical drug-targeting research and therapeutic approaches.

    Abstract Objective: To systematically evaluate the efficacy of valproic acid (VPA) in rats with spinal cord injury (SCI) to reduce the risk of clinical conversion and provide a valuable reference for future animal and clinical studies.
    Methods: We searched scientific databases, including PubMed, Ovid-Embase, Web of Science, and Scopus databases. The relevant literature was searched from the establishment date of the database to June 28, 2023. The search results were screened, data were extracted, and the quality of the literature was evaluated independently by 2 reviewers.
    Results: Among 656 nonduplicated references, 14 articles were included for meta-analysis. The summary results showed that the overall Basso, Beattie and Bresnahan scores of the VPA intervention group were significantly higher than those in the control group at 1-6 weeks after VPA intervention. Subgroup analysis showed that the injury model, administration dose, rat strain, country of study, or follow-up duration had no significant effect on the efficacy of VPA on rats with SCI. In addition, mesh analysis showed that high doses of the VPA group had a better effect on SCI rats, compared with the low dose group and the medium dose group.
    Conclusions: To date, this is the first systematic evaluation of the potential effects of VPA on motor recovery in rats with SCI. We concluded that VPA can promote motor recovery in rats with SCI, and higher doses of VPA seem to be more effective in rats with SCI. However, the limited quality and sample of included studies reduced the application of this meta-analysis. In the future, more high-quality, direct comparative studies are needed to explore this issue in depth.

    Abstract Acute myocardial infarction is one of the leading causes of morbidity worldwide, but the underlying mechanism responsible for myocardial ischemia–reperfusion (I/R) injury remains elusive. lncRNA plays roles in inflammatory response, cell apoptosis and regulation of myocardial ischemia. However, whether lncRNA HAGLR could regulate myocardial I/R injury and the molecular mechanism need to be further investigated. lncRNA has been shown to bind to miRNAs and compete with endogenous RNAs. miR-133a-3p has been shown to regulate cardiomyocyte apoptosis and ischemic myocardial injury. In this work, it has shown that knockdown of HAGLR could suppress inflammatory response and cell apoptosis induced by I/R and, thus, alleviate myocardial I/R injury. HAGLR promoted myocardial I/R injury by inhibiting miR-133a-3p to promote MAPK1 expression.

    Abstract Selective labeling of the protein of interest (POI) in genetically unmodified live cells is crucial for understanding protein functions and kinetics in their natural habitat. In particular, spatiotemporally controlled installation of the labels on a POI under light control without affecting their original activity is in high demand but is a tremendous challenge. Here, we describe a novel ligand-directed photoclick strategy for spatiotemporally controlled labeling of endogenous proteins in live cells.
