    Abstract Precision metrology is important for understanding and monitoring various phenomena, especially in special and scientific applications. The subject of the article covers selected aspects of the study of high-resolution measurements of the absorption spectra and their impact on practical issues. This research is crucial for the development of new systems based on laser absorption spectroscopy methods that can detect trace amounts of matter. Some of the most important parameters describing measuring instruments are also defined and illustrated. Specific examples of measurement results are presented, taking into account the need for the highest possible resolution, accuracy and precision.

    Abstract This paper presents the description of the wavelength modulation spectroscopy (WMS) experiment, the parameters of which were established by use of the Artificial Intelligence (AI) algorithm. As a result, a significant improvement in the signal power to noise power ratio (SNR) was achieved, ranging from 1.6 to 6.5 times, depending on the harmonic. Typically, optimizing the operation conditions of WMS-based gas sensors is based on long-term simulations, complex mathematical model analysis, and iterative experimental trials. An innovative approach based on a biological-inspired genetic algorithm (GA) and custom-made electronics for laser control is proposed. The experimental setup was equipped with a 31.23 m Heriott multipass cell, software lock-in, and algorithms to control the modulation process of the quantum cascade laser (QCL) operating in the long-wavelength-infrared (LWIR) spectral range. The research results show that the applied evolutionary approach can efficiently and precisely explore a wide range of WMS parameter combinations, enabling researchers to dramatically reduce the time needed to identify optimal settings.     Abstract The paper presents one of the laser absorption spectroscopy techniques as an effective tool for sensitive analysis of trace gas species in human breath. Characterization of nitric oxide, carbonyl sulphide and ethane, and the selection of their absorption lines are described. Experiments with some biomarkers showed that detection of pathogenic changes at the molecular level is possible using this technique. Thanks to cavity enhanced spectroscopy application, detection limits at the ppb-level and short measurements time (<3 s) were achieved. Absorption lines of reference samples of the selected volatile biomarkers were probed using a distributed feedback quantum cascade laser and a tunable laser system consisting of an optical parametric oscillator and difference frequency generator. Setup using the first source provided a detection limit of 30 ppb for nitric oxide and 250 ppb for carbonyl sulphide. During experiments employing a second laser, detection limits of 0.9 ppb and 0.3 ppb were obtained for carbonyl sulphide and ethane, respectively. The conducted experiments show that this type of diagnosis would significantly increase chances for effective therapy of some diseases. Additionally, it offers non-invasive and real time measurements, high sensitivity and selectivity as well as minimizing discomfort for patients. For that reason, such sensors can be used in screening for early detection of serious diseases.

    Abstract High precision and fast measurement of gas concentrations is important for both understanding and monitoring various phenomena, from industrial and environmental to medical and scientific applications. This article deals with the recent progress in ammonia detection using in-situ solid-state and optical methods. Due to the continuous progress in material engineering and optoelectronic technologies, these methods are among the most perceptive because of their advantages in a specific application. We present the basics of each technique, their performance limits, and the possibility of further development. The practical implementations of representative examples are described in detail. Finally, we present a performance comparison of selected practical application, accumulating data reported over the preceding decade, and conclude from this comparison.

    Abstract In neurons, stromal interaction molecule (STIM) proteins regulate store-operated Ca 2+ entry (SOCE) and are involved in calcium signaling pathways. However, STIM activity in neurological diseases is unclear and should be clarified by studies that are performed in vivo rather than in cultured cells in vitro. The present study investigated the role of neuronal Stim2b protein in zebrafish. We generated stim2b knockout zebrafish, which were fertile and had a regular lifespan. Using various behavioral tests, we found that stim2b −/− zebrafish larvae were hyperactive compared with wild-type fish. The mutants exhibited increases in mobility and thigmotaxis and disruptions of phototaxis. They were also more sensitive to pentylenetetrazol and glutamate treatments. Using lightsheet microscopy, a higher average oscillation frequency and higher average amplitude of neuronal Ca 2+ oscillations were observed in stim2b −/− larvae. RNA sequencing detected upregulation of the annexin 3a and gpr39 genes and downregulation of the rrm2 , neuroguidin , and homer2 genes. The latter gene encodes a protein that is involved in several processes that are involved in Ca 2+ homeostasis in neurons, including metabotropic glutamate receptors. We propose that Stim2b deficiency in neurons dysregulates SOCE and triggers changes in gene expression, thereby causing abnormal behavior, such as hyperactivity and susceptibility to seizures.
