    Abstract Background: Spasmolytic polypeptide-expressing metaplasia (SPEM) is a potential preneoplastic lesion.
    Aim: To elucidate the microRNA (miR)-7-mediated preventive and inhibitive effects of Yiwei Xiaoyu granules (YWXY) in SPEM lesions.
    Methods: Gastric mucosa biopsies were collected from chronic atrophic gastritis patients and healthy people with signed informed consent. YWXY was administered to the mice with induced SPEM by tamoxifen, and the gastric mucosa was harvested on the tenth day of the experiment. Then immunohistochemistry and immunofluorescence were performed to validate the SPEM, lesions and the potential mechanism was investigated. RNA transcripts were detected with reverse transcription-quantitative polymerase chain reaction.
    Results: The expression of miR-7 was downregulated in the SPEM lesions, and expression of trefoil factor 2 (TFF2) and clusterin was high in the human gastric mucosa.
    Conclusion: miR-7 downregulation is an early event in SPEM through regulation of TFF2 in human gastric mucosa. YWXY is able to inhibit the cell proliferation and restore the expression of miR-7 by mediating TFF2 in the SPEM mouse model.

    Abstract Objective: To investigate the effects of Fu Fang Yi Mu Cao (FFYMC) capsules combined with Yiqi Xiaoyu (YX) decoction on persistent lochia after birth.
    Methods: In this retrospective analysis, 120 patients with lochia treated in our hospital from January 2014 to October 2020 were enrolled as study subjects. Sixty of each were randomly allocated into the study group (60 cases, treated with FFYMC capsules and YX decoction) and the control group (60 cases, treated with YX decoction). The two groups were compared in terms of efficacy, volume of lochia and changes in Traditional Chinese Medicine Syndrome Score (TCMSS) before and after intervention, and the incidence of adverse reactions.
    Results: (1) The total effective rate in the study group was 100.00%, which was significantly higher than that in the control group (P<0.05); (2) The difference in volume of lochia between two groups before the intervention was not statistically significant (P>0.05), and was significantly lower in the study group than in the control group at day 4 and day 7 of the intervention (P<0.05); (3) The difference in TCMSS between two groups before the treatment was not statistically significant (P>0.05). TCMSS was lower in the study group than in the control group after treatment (P<0.05); (4) The decreased height of uterine fundus in the study group was higher than that in the control group at 5 d after treatment (P<0.05); (5) The plasma viscosity in the study group was significantly lower than that in the control group (P<0.05); (6) The total incidence of adverse reactions in the study group was 8.33%, which was not significantly different from the control group which was 11.67% (P>0.05).
    Conclusion: FFYMC capsule and YX decoction could improve the treatment effect for primiparas with lochia in terms of reducing volume of lochia and improving clinical symptoms with high safety.

    Abstract Objective: Heart failure (HF), a worldwide health condition, is the result of many cardiovascular diseases. The traditional Chinese medicine (TCM) Xiaoyu Jiangzhi capsule (XYC) has long been in use in China to treat hyperlipidemia and inhibit platelet aggregation. This study explores the effects of XYC on heart failure (HF) and its detailed mechanisms. Methods: Isoproterenol (ISO, 30 mg/kg) was injected intraperitoneally for 7 days to copy a HF model of 10–12 weeks old, 20–30 g male mice. We then compared the CON (control) group, ISO (HF model) group, MET (metoprolol) group, and XYC group. Cardiac systolic function and left wall thickness were evaluated by echocardiograph. Using western blot analysis, we detected the proteins of calmodulin dependent protein kinase II (CaMKII) and sarco/endoplasmic reticulum Ca2+-ATPase (Serca). Furthermore, tsA201 cells were cultured and the human CaV1.2 calcium channel current (hCaV1.2) were detected by patch clamp experiments. Results: XYC reduced HF, inhibiting the protein expression of CaMKII, but Serca did not change significantly. Moreover, XYC inhibited the peak amplitude of the hCaV1.2 current, depolarizing shifted the activation curve 27.6 mV, and shifted the inactivation curve toward a positive potential 17.6 mV. The fraction recovered from inaction was reduced in XYC group compared with that in CON group. Conclusion: XYC could inhibit ISO-induced HF by reducing the Ca2+/CaMKII signaling pathway in mice.

    Abstract RNA sequencing (RNA-seq) has become a cornerstone in transcriptomics, offering detailed insights into gene expression across diverse biological conditions and sample types. However, RNA-seq data often suffer from batch effects, which are systematic non-biological differences that compromise data reliability and obscure true biological variation. To address these challenges, we introduce ComBat-ref, a refined method of batch effect correction that enhances the statistical power and reliability of differential expression analysis in RNA-seq data. Building on the foundations of ComBat-seq, ComBat-ref employs a negative binomial model to adjust count data but innovates by using a pooled dispersion parameter for entire batches and preserving count data for the reference batch. Our method demonstrated superior performance in both simulated environments and real datasets, such as the growth factor receptor network (GFRN) data and NASA GeneLab transcriptomic datasets, significantly improving sensitivity and specificity over existing methods. By effectively mitigating batch effects while maintaining high detection power, ComBat-ref proves to be a robust tool for enhancing the accuracy and interpretability of RNA-seq data analyses.

    Abstract Objective To examine the effects of the temporal inverted internal limiting membrane (ILM) flap technique for foveal reconstruction in patients with highly myopic macular hole-associated retinal detachment (MHRD). Methods A retrospective case series analysis of four patients (four eyes) with MHRD was conducted. The foveal optical coherence tomography changes following treatment using the temporal inverted ILM flap technique were evaluated. Results In Patient 1, the ILM bridged the macular hole and residual subretinal fluid on postoperative day 6, and complete retinal reattachment was achieved at 19 months. Patient 2 exhibited reduced retinal detachment, with visible ILM inversion and macular hole closure after 14 days. In Patient 3, macular hole closure and fovea formation had occurred by day 25, and the ILM flap was visible. At 2 months, the outer collagenous layer connection in the central fovea and recovery of the external limiting membrane and ellipsoid zone were observed. Patient 4 had a “white hole” MHRD, with macular hole closure achieved on postoperative day 20, albeit with a suboptimal foveal shape. Conclusion The temporal inverted ILM flap technique in conjunction with vitrectomy facilitates foveal reconstruction in patients with successful treatment of MHRD, and this reconstruction process can be observed by optical coherence tomography.

    Abstract Spermatogonial stem cells (SSCs) produce haploid sperm via mitosis and meiosis in vivo. Although the technique to culture mouse SSCs has been well established, induction of meiosis in vitro has remained a challenge. Retinoic acid (RA) is required for meiosis in vivo; however, RA alone is not sufficient to induce meiosis in vitro. Here, we describe a method in which nutrient restriction and RA synergistically induce meiotic initiation into meiotic prophase I in cultured mouse SSCs.
