    Abstract Potato market value is heavily affected by tuber quality traits such as shape, color, and skinning. Despite this, potato breeders often rely on subjective scales that fail to precisely define phenotypes. Individual human evaluators and the environments in which ratings are taken can bias visual quality ratings. Collecting quality trait data using machine vision allows for precise measurements that will remain reliable between evaluators and breeding programs. Here we present TubAR (Tuber Analysis in R), an image analysis program designed to collect data for multiple tuber quality traits at low cost to breeders. To assess the efficacy of TubAR in comparison to visual scales, red-skinned potatoes were evaluated using both methods. Broad sense heritability was consistently higher for skinning, roundness, and length to width ratio using TubAR. TubAR collects essential data on fresh market potato breeding populations while maintaining efficiency by measuring multiple traits through one phenotyping protocol.

    Abstract The shuffle model of differential privacy has gained significant interest as an intermediate trust model between the standard local and central models [EFMRTT19; CSUZZ19]. A key result in this model is that randomly shuffling locally randomized data amplifies differential privacy guarantees. Such amplification implies substantially stronger privacy guarantees for systems in which data is contributed anonymously [BEMMRLRKTS17]. In this work, we improve the state of the art privacy amplification by shuffling results both theoretically and numerically. Our first contribution is the first asymptotically optimal analysis of the R\'enyi differential privacy parameters for the shuffled outputs of LDP randomizers. Our second contribution is a new analysis of privacy amplification by shuffling. This analysis improves on the techniques of [FMT20] and leads to tighter numerical bounds in all parameter settings.

    Abstract Background: Although convincing data exist regarding the prognostic utility of positron emission tomographic (PET)-computed tomographic (CT) imaging in Hodgkin lymphoma and diffuse large B-cell lymphoma, its prognostic utility both during treatment and immediately after treatment have not been systematically evaluated in a large mantle cell lymphoma (MCL) patient cohort to support its use in clinical practice.
    Methods: The authors conducted a retrospective cohort study to examine the prognostic utility of PET-CT imaging in a uniform MCL patient cohort undergoing dose-intensive chemotherapy (R-HyCVAD) in the frontline setting. The primary study endpoints were progression-free survival (PFS) and overall survival (OS). PET-CT images were centrally reviewed for the purposes of this study using standardized response criteria.
    Results: Fifty-three patients with advanced stage MCL with PET-CT data were identified. With median follow-up of 32 months, 3-year PFS and OS estimates were 76% (95% confidence interval [CI], 64%-84%) and 84% (95% CI, 72%-90%), respectively. Interim PET-CT status was not associated with PFS (hazard ratio [HR], 0.9; 95% CI, 0.3-2.7; P = .8) or OS (HR, 0.6; 95% CI, 0.1-2.9; P = .5). Post-treatment PET-CT status was statistically significantly associated with PFS (HR, 5.2; 95% CI, 2.0-13.6; P = .001) and trended toward significant for OS (HR, 2.8; 95% CI, 0.8-9.6; P = .07).
    Conclusions: These data do not support the prognostic utility of PET-CT in pretreatment and interim treatment settings. A positive PET-CT after the completion of therapy identifies a patient subset with an inferior PFS and a trend toward inferior OS.

    Abstract Misregulation of a pluripotency-associated transcription factor network in adult tissues is associated with the expansion of rare, highly malignant tumor-initiating stem cells (TISCs) through poorly understood mechanisms. We demonstrate that robust and selective expression of the receptor for the adipocyte-derived peptide hormone leptin (OB-R) is a characteristic feature of TISCs and of a broad array of embryonic and induced pluripotent stem cells and is mediated directly by the core pluripotency-associated transcription factors OCT4 and SOX2. TISCs exhibit sensitized responses to leptin, including the phosphorylation and activation of the pluripotency-associated oncogene STAT3 and induction of Oct4 and Sox2, thereby establishing a self-reinforcing signaling module. Exposure of cultured mouse embryonic stem cells to leptin sustains pluripotency in the absence of leukemia inhibitory factor. By implanting TISCs into leptin-deficient ob/ob mice or into comparably overweight Lepr(db/db) mice that produce leptin, we provide evidence of a central role for the leptin-TISC-signaling axis in promoting obesity-induced tumor growth. Differential responses to extrinsic, adipocyte-derived cues may promote the expansion of tumor cell subpopulations and contribute to oncogenesis.
