    Abstract In many NLP applications that involve interpreting sentences within a rich context -- for instance, information retrieval systems or dialogue systems -- it is desirable to be able to preserve the sentence in a form that can be readily understood without context, for later reuse -- a process known as ``decontextualization''. While previous work demonstrated that generative Seq2Seq models could effectively perform decontextualization after being fine-tuned on a specific dataset, this approach requires expensive human annotations and may not transfer to other domains. We propose a few-shot method of decontextualization using a large language model, and present preliminary results showing that this method achieves viable performance on multiple domains using only a small set of examples.

    Abstract Many practical applications of dialogue technology require the generation of responses according to a particular developer-specified persona. While a variety of personas can be elicited from recent large language models, the opaqueness and unpredictability of these models make it desirable to be able to specify personas in an explicit form. In previous work, personas have typically been represented as sets of one-off pieces of self-knowledge that are retrieved by the dialogue system for use in generation. However, in realistic human conversations, personas are often revealed through story-like narratives that involve rich habitual knowledge -- knowledge about kinds of events that an agent often participates in (e.g., work activities, hobbies, sporting activities, favorite entertainments, etc.), including typical goals, sub-events, preconditions, and postconditions of those events. We capture such habitual knowledge using an explicit schema representation, and propose an approach to dialogue generation that retrieves relevant schemas to condition a large language model to generate persona-based responses. Furthermore, we demonstrate a method for bootstrapping the creation of such schemas by first generating generic passages from a set of simple facts, and then inducing schemas from the generated passages.

    Abstract Aluminium matrix composites (AMCs) represent suitable materials for lightweight design applications. The abrasive ceramic reinforcements typically require diamond cutting materials to prevent excessive tool wear. In milling with diamond cutting materials the influence of cutting parameters was already examined to a significant extent. Investigations concerning the effect of modified tool geometries are limited and the potentials with regard to the geometrical and physical surface properties are unclear. Accordingly, experimental investigations in milling of a 10 vol.% <math xmlns="http://www.w3.org/1998/Math/MathML" display="inline"><semantics><mi>SiC</mi></semantics></math> particle-reinforced aluminium wrought alloy EN AW-2017 T4 were addressed. The effect of modified corner and minor cutting edge geometries were investigated based on mono crystalline diamond (MCD)-tipped tools to benefit stable process conditions. The results indicated achievable areal roughness values in the range around <math xmlns="http://www.w3.org/1998/Math/MathML" display="inline"><semantics><mrow><mn>0.2</mn><mspace width="0.166667em"></mspace><mi mathvariant="sans-serif">μ</mi><mi mathvariant="normal">m</mi></mrow></semantics></math> . Especially the application of the lowest cutting edge angle and a trailing minor cutting edge led to strong fluctuations of the surface parameters. The lowest valley void volumes were achieved with an arched minor cutting edge. Generally, finish machining led to stronger compressive residual stresses compared to the state prior to machining. The strongest increase was achieved using a corner radius combined with a straight minor cutting edge. It is concluded that reduced effective radii generating the surface enable an acceptable surface structure and strong compressive residual stresses and should be addressed in further investigations.

    Abstract Epitope-based vaccines have revolutionized vaccine research in the last decades. Due to their complex nature, bioinformatics plays a pivotal role in their development. However, existing algorithms address only specific parts of the design process or are unable to provide formal guarantees on the quality of the solution. We present a unifying formalism of the general epitope vaccine design problem that tackles all phases of the design process simultaneously and combines all prevalent design principles. We then demonstrate how to formulate the developed formalism as an integer linear program, which guarantees optimality of the designs. This makes it possible to explore new regions of the vaccine design space, analyze the trade-offs between the design phases, and balance the many requirements of vaccines.

    Abstract Motivation: Conceptually, epitope-based vaccine design poses two distinct problems: (i) selecting the best epitopes to elicit the strongest possible immune response and (ii) arranging and linking them through short spacer sequences to string-of-beads vaccines, so that their recovery likelihood during antigen processing is maximized. Current state-of-the-art approaches solve this design problem sequentially. Consequently, such approaches are unable to capture the inter-dependencies between the two design steps, usually emphasizing theoretical immunogenicity over correct vaccine processing, thus resulting in vaccines with less effective immunogenicity in vivo.
    Results: In this work, we present a computational approach based on linear programming, called JessEV, that solves both design steps simultaneously, allowing to weigh the selection of a set of epitopes that have great immunogenic potential against their assembly into a string-of-beads construct that provides a high chance of recovery. We conducted Monte Carlo cleavage simulations to show that a fixed set of epitopes often cannot be assembled adequately, whereas selecting epitopes to accommodate proper cleavage requirements substantially improves their recovery probability and thus the effective immunogenicity, pathogen and population coverage of the resulting vaccines by at least 2-fold.

    Abstract Inorganic photoacids and photobases comprising of photoactive transition metal complexes (TMCs) offer the ability to modulate proton transfer reactions through light irradiation, while utilizing the excellent optical properties of the latter. This provides a powerful tool for precise control over chemical reactions and processes, with implications for both fundamental science and practical applications. In this contribution, we present a novel molecular architecture amending an Fe-NHC complex with a pendant quinoline, as a prototypical photobase, as a representative earth-abundant TMC based inorganic photobase. We characterize the excited-state properties and proton-transfer dynamics using steady-state absorption and emission spectroscopy as well as pump wavelength dependent transient absorption spectroscopy in various protic solvents. The kinetics and thermodynamics of proton transfer in the quinoline moiety are influenced by both the presence of the metal center and the choice of the solvent. Furthermore, we see indications of intramolecular energy transfer from the quinoline to the MLCT state as a limiting factor for panchromatic photobasicity of the complex.
