    Serientitel Lecture notes on mathematical modelling in the life sciences
    Abstract This monograph introduces some current developments in the modelling of the spread of tick-borne diseases. Effective modelling requires the integration of multiple frameworks. Here, particular attention is given to the previously neglected issues of tick developmental and behavioral diapause, tick-borne pathogen co-feeding transmission, and their interactions. An introduction to the required basics of structured population formulations and delay differential equations is given, and topics for future study are suggested. The described techniques will also be useful in the study of other vector-borne diseases. The ultimate aim of this project is to develop a general qualitative framework leading to tick-borne disease risk predictive tools and a decision support system. The target audience is mathematical biologists interested in modelling tick population dynamics and tick-borne disease transmission, and developing computational tools for disease prevention and control.

    Abstract Lychee seed is a traditional Chinese medicine and possesses many activities, including hypoglycemia, liver protection, antioxidation, antivirus, and antitumor. However, its effect on neuroprotection is still unclear. The present study investigated the effects of lychee seed saponins (LSS) on neuroprotection and associated mechanisms. We established a rat model of Alzheimer’s disease (AD) by injecting Aβ25–35 into the lateral ventricle of rats and evaluated the effect of LSS on spatial learning and memory ability via the Morris water maze. Neuronal apoptosis was analyzed by hematoxylin and eosin stain and terminal deoxynucleotidyl transferase (Tdt)-mediated dUTP nick-end labeling analysis, and mRNA expression of caspase-3 and protein expressions of Bax and Bcl-2 by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting, respectively. The results showed that LSS remarkably improved cognitive function and alleviated neuronal injury by inhibiting apoptosis in the hippocampus of AD rats. Furthermore, the mRNA expression of caspase-3 and the protein expression of Bax were downregulated, while the protein expression of Bcl-2 and the ratio of Bcl-2/Bax were increased by LSS. We demonstrate that LSS significantly improves cognitive function and prevent neuronal injury in the AD rats via regulation of the apoptosis pathway. Therefore, LSS may be developed as a nutritional supplement and sold as a drug for AD prevention and/or treatment.

    Abstract Obesity has emerged as a major threat to public health in both the western and developing world. Essentially a disorder of energy balance, obesity occurs when energy intake and storage exceeds expenditure. Much of energy homeostasis depends on the activity and function of adipose tissue. Adipocytes in mammals fall into two categories classified by their primary functions: white fat cells that mediate energy storage and thermogenic fat cells that counteract hypothermia and obesity through adaptive thermogenesis. Whereas white fat and its function as an energy reservoir and endocrine organ have been studied for decades and are relatively well understood, until recently many aspects of the thermogenic fat biology have remained elusive. Accumulating evidence supports the hypothesis that thermogenic fat cells arise from at least two different developmental origins: the ones of a skeletal muscle-like lineage are now called "classical" brown fat cells, and the rest of the thermogenic fat cells are normally referred to as the beige fat cells. The last decade has witnessed an explosion of interest and studies focusing on the regulation of thermogenic fat cells and potential therapeutics targeting these adipocytes. Here we summarize the recent advancements in our understanding of these metabolically active fat cells

    Abstract The impact of fat intake on hypercholesterolemia and related atherosclerotic cardiovascular diseases has been studied for decades. However, the current evidence base suggests that fatty acids also influences cardiometabolic diseases through other mechanisms including effects on glucose metabolism, body fat distribution, blood pressure, inflammation, and heart rate. Furthermore, studies evaluating single fatty acids have challenged the simplistic view of shared health effects within fatty acid groups categorized by degree of saturation. In addition, investigations of endogenous fatty acid metabolism, including genetic studies of fatty acid metabolizing enzymes, and the identification of novel metabolically derived fatty acids have further increased the complexity of fatty acids' health impacts. This Special Issue aims to include original research and up-to-date reviews on genetic and dietary modulation of fatty acids, and the role and function of dietary and metabolically derived fatty acids in cardiometabolic health

    Abstract Hyperspectral Imaging (HSI) seamlessly integrates imaging and spectroscopy, capturing both spatial and spectral data concurrently. With widespread applications in medical diagnostics, HSI serves as a noninvasive tool for gaining insights into tissue characteristics. The distinctive spectral profiles of biological tissues set HSI apart from traditional microscopy in enabling in vivo tissue analysis. Despite its potential, existing HSI techniques face challenges such as alignment issues, low light throughput, and tissue heating due to intense illumination. This study introduces an innovative HSI system featuring active sequential bandpass illumination seamlessly integrated into conventional optical instruments. The primary focus is on analyzing oxyhemoglobin and deoxyhemoglobin saturation in animal tissue samples using multivariate linear regression. This approach holds promise for enhancing noninvasive medical diagnostics. A key feature of the system, active bandpass illumination, effectively prevents tissue overheating, thereby bolstering its suitability for medical applications.

    Abstract Immune checkpoint inhibitor (ICI)-based therapy has made an unprecedented impact on survival benefit for a subset of cancer patients; however, only a subset of cancer patients is benefiting from ICI therapy if all cancer types are considered. With the advanced understanding of interactions of immune effector cell types and tumors, cell-based therapies are emerging as alternatives to patients who could not benefit from ICI therapy. Pioneering work of chimeric antigen receptor T (CAR-T) therapy for hematological malignancies has brought encouragement to a broad range of development for cellular-based cancer immunotherapy, both innate immune cell-based therapies and T-cell-based therapies. Innate immune cells are important cell types due to their rapid response, versatile function, superior safety profiles being demonstrated in early clinical development, and being able to utilize multiple allogeneic cell sources. Efforts on engineering innate immune cells and exploring their therapeutic potential are rapidly emerging. Some of the therapies, such as CD19 CAR natural killer (CAR-NK) cell-based therapy, have demonstrated comparable early efficacy with CD19 CAR-T cells. These studies underscore the significance of developing innate immune cells for cancer therapy. In this review, we focus on the current development of emerging NK cells, γδ T cells, and macrophages. We also present our views on potential challenges and perspectives to overcome these challenges.
